Label-free enrichment of adrenal cortical progenitor cells using inertial microfluidics.
Passive and label-free isolation of viable target cells based on intrinsic biophysical cellular properties would allow for cost savings in applications where molecular biomarkers are known as well as potentially enable the separation of cells with little-to-no known molecular biomarkers. We have demonstrated the purification of adrenal cortical progenitor cells from digestions of murine adrenal glands utilizing hydrodynamic inertial lift forces that single cells and multicellular clusters differentially experience as they flow through a microchannel. Fluorescence staining, along with gene expression measurements, confirmed that populations of cells collected in different outlets were distinct from one another. Furthermore, primary murine cells processed through the device remained highly viable and could be cultured for 10 days in vitro. The proposed target cell isolation technique can provide a practical means to collect significant quantities of viable intact cells required to translate stem cell biology to regenerative medicine in a simple label-free manner

Durable and ductile double-network material for dust control
Dust generation is a world-wide issue due to its serious deleterious effects on the environment, human health and safety, and the economy. Although various dust suppression methods have been used for decades, some critical drawbacks in state-of-the-art technology still remain unsolved, such as short-lasting, ground water impact, and prone to water. This work reports a soil stabilizer based on non-toxic material and forms a ductile and durable double-network in soil, namely “D3 soil stabilizer”, which not only improves soil mechanical toughness of surface soil but also suppresses dust generation. A copolymer comprising hydrophilic and hydrophobic components combined with enzyme-induced carbonate precipitation is utilized as an in-situ gelation binder to soil particle. The tunable hydrophobic-to-hydrophilic component ratio minimizes undesirable soil matrix expansion and mechanical strength loss upon experiencing wet-dry processes, while still retains good water affinity. We further demonstrated controllable treatment depth by fine-tuning precursor composition, which is essential to minimize environmental impact. The double-network morphology with carbonate precipitate embedded uniformly in polymer matrix is observed via microscopic imaging. The nature of outstanding ductility, high durability against water, and good long-term stability were supported by systematic unconfined compressive strength (UCS) measurements on treated soil, which show strong inter-particles binding, good retention of peak strength, increased strain at peak strength, and increased toughness after soil samples have experienced wet-dry processes

An efficient semiparametric maxima estimator of the extremal index
The extremal index , a measure of the degree of local dependence in
the extremes of a stationary process, plays an important role in extreme value
analyses. We estimate semiparametrically, using the relationship
between the distribution of block maxima and the marginal distribution of a
process to define a semiparametric model. We show that these semiparametric
estimators are simpler and substantially more efficient than their parametric
counterparts. We seek to improve efficiency further using maxima over sliding
blocks. A simulation study shows that the semiparametric estimators are
competitive with the leading estimators. An application to sea-surge heights
combines inferences about with a standard extreme value analysis of
block maxima to estimate marginal quantiles.Comment: 17 pages, 7 figures. Minor edits made to version 1 prior to journal

Modelling and simulation of counter-current and confined jet reactors for hydrothermal synthesis of nano-materials
A confined jet mixer and a counter-current mixer for the continuous hydrothermal flow synthesis of TiO2 nano-materials under supercritical water conditions have been investigated using computational fluid dynamics (CFD). The fluid flow and heat transfer behaviour, including velocity and temperature profiles in both reactor configurations, are studied using the CFD tool ANSYS Fluent. The tracer concentration profiles are also simulated via solving species equations from which the mixing behaviour in the reactors is examined. A combined CFD and population balance model is used to predict the size distribution. The predicted temperature distributions for both reactors were found to be in good agreement with experimentally measured data. Detailed comparison of the hydrodynamic and thermal behaviours, and particle size distributions between the two reactors helped in the identification of key factors that affect the reactor performance, and also provided suggestions for reactor design optimisation and scale-up
Barrier-Lyapunov-Function-Based Backstepping Adaptive Hybrid Force/Position Control for Manipulator with Force and Position Constraints
In this paper, we present a backstepping adaptive hybrid force/position control based on Barrier Lyapunov Function for a robotic manipulator to prevent constraint violation of applied force and position simultaneously. First, the task space is partitioned according to the constrained and unconstrained directions, and a new representation of dynamics is introduced. Next, force/position control is applied using the strict-feedback backstepping technique, in which a time-varying Barrier Lyapunov Function is employed to ensure that the force and position do not violate their constraints. Finally, to deal with uncertainty, disturbance and non-linearity of the system, an adaptive radial basis function neural network (RBFNN) is also implemented in the control algorithm. Stability proof of the proposed control method is presented, and simulation studies on a 2-link manipulator show the effectiveness as well as the performance of the proposed controller in preventing constraint violation
Sustainability assessment of a single-use plastics ban
Governments around the world are introducing single-use plastics bans to alleviate plastic marine pollution. This paper investigates whether banning single-use plastic items is an appropriate strategy to protect the environment. Product life cycle assessment was conducted for single-use plastic and single-use non-plastic alternatives. The life cycle impacts of the two product categories were compared and scaled according to EU consumption of 2016. The results show that a single-use plastics ban would decrease plastic marine pollution in the EU by 5.5% which equates to a 0.06% decrease globally. However, such a ban would increase emissions contributing to marine aquatic toxicity in the EU by 1.4%. This paper concludes that single-use items are harmful to the environment regardless of their material. Therefore, banning or imposing a premium price on single-use items in general and not only single-use plastic items is a more effective method of reducing consumption and thereby pollution. The plastics ban only leads to a small reduction of global plastic marine pollution and thus provides only a partial solution to the problem it intends to solve.</jats:p
Early diagnosis of primary human herpesvirus 6 infection in childhood: Serology, polymerase chain reaction, and virus load
Qualitative and quantitative polymerase chain reaction (PCR) for human herpesvirus 6 (HHV-6) DNA in whole blood and plasma was correlated with serology and clinical assessment in 143 children hospitalized for undifferentiated febrile illness to evaluate options for diagnosis of primary HHV-6 infection on the acute blood specimen. PCR and serology for HHV-7 were done in parallel to define serologic cross-reactions. Using HHV-6 seroconversion as the reference standard, detection of HHV-6 DNA in whole blood in the absence of antibody in the plasma was the most reliable evidence of primary HHV-6 infection. Detection of HHV-6 DNA in plasma and a high virus load in whole blood (>3.3 log 10 copies/5 ÎĽL) had a sensitivity of 90% and 100%, respectively, in diagnosing primary HHV-6 infection. However, both were occasionally found in patients with other infections, possibly associated with HHV-6 reactivation. Maternal antibody may confound interpretation of serology in patients under 3 months of age.published_or_final_versio
