    Expression of the mucus adhesion gene Mub, surface layer protein Slp and adhesion-like factor EF-Tu by Lactobacillus acidophilus ATCC 4356 grown in the presence of mucin, bile and pancreatin and at low pH was studied using real-time PCR. None of the genes were up-regulated under increasing concentrations of mucin, while Slp and EF-Tu were up-regulated in the presence of bile and pancreatin at normal concentrations (0.3%, w/v) and under stress conditions (1.0%, w/v)

    Safety assessment of Lactobacillus plantarum 423 and Enterococcus mundtii ST4SA determined in trials with Wistar rats

    Colonization of Lactobacillus plantarum 423 and Enterococcus mundtii ST4SA in the gastro-intestinal tract was determined by using Wistar rats as model. The strains were administered through intragastric gavage over 14 days. FISH with strain-specific oligonucleotide probes indicated that Lact. plantarum 423 adhered to the surfaces of the ileum and the cecum. Enterococcus mundtii ST4SA, on the other hand, adhered to the surfaces of the cecum and colon. Results obtained by DGGE have shown that strains 423 and ST4SA excluded Enterobacteriaceae, but not lactic acid bacteria, from the cecum and colon. No signs of perforation of epithelial cells by strains 423 and ST4SA were detected. The spleen and liver appeared healthy and blood counts were normal, suggesting that the strains are not pathogenic. Both strains produce antimicrobial peptides active against a number of pathogens and may be considered as probiotics

    Investigation of a Novel Common Subexpression Elimination Method for Low Power and Area Efficient DCT Architecture

    A wide interest has been observed to find a low power and area efficient hardware design of discrete cosine transform (DCT) algorithm. This research work proposed a novel Common Subexpression Elimination (CSE) based pipelined architecture for DCT, aimed at reproducing the cost metrics of power and area while maintaining high speed and accuracy in DCT applications. The proposed design combines the techniques of Canonical Signed Digit (CSD) representation and CSE to implement the multiplier-less method for fixed constant multiplication of DCT coefficients. Furthermore, symmetry in the DCT coefficient matrix is used with CSE to further decrease the number of arithmetic operations. This architecture needs a single-port memory to feed the inputs instead of multiport memory, which leads to reduction of the hardware cost and area. From the analysis of experimental results and performance comparisons, it is observed that the proposed scheme uses minimum logic utilizing mere 340 slices and 22 adders. Moreover, this design meets the real time constraints of different video/image coders and peak-signal-to-noise-ratio (PSNR) requirements. Furthermore, the proposed technique has significant advantages over recent well-known methods along with accuracy in terms of power reduction, silicon area usage, and maximum operating frequency by 41%, 15%, and 15%, respectively

    The effects of free trade agreements on the stock market: Evidence from Vietnam.

    This study examines the effects of news events related to the European Union-Vietnam Free Trade Agreement (EVFTA) on the Vietnam stock market from 2010 to 2020. We calculate sectoral abnormal returns prior to, during, and after announcements and find that the Vietnamese stock market is susceptible to these events. We discovered that the announcement had a negative impact on the market, which might diminish the effectiveness of the Agreement. The findings show that more than half of Vietnam's sectors had an immediate reaction to EVFTA announcements, with fourteen reacting negatively and six responding positively. Two of the ten events did not have any immediate impact on these industries but all events resulted in either early or delayed reactions. We also find market scepticism and major changes in the deal led to the emergence of a diamond risk structure. We run multiple robustness tests to account for market integration and other factors that may affect stock returns. In addition, we explore potential sectoral systematic risk changes following these occurrences using different ARCH-type models. These additional tests confirm the robustness of our findings

    Identification and Characterization of 2′-Deoxyadenosine Adducts Formed by Isoprene Monoepoxides in Vitro

    Isoprene, the 2-methyl analog of 1,3-butadiene, is ubiquitous in the environment, with major contributions to total isoprene emissions stemming from natural processes despite the compound being a bulk industrial chemical. Additionally, isoprene is a combustion product and a major component in cigarette smoke. Isoprene has been classified as possibly carcinogenic to humans (group 2B) by IARC and as reasonably anticipated to be a human carcinogen by the National Toxicology Program. Isoprene, like butadiene, requires metabolic activation to reactive epoxides to exhibit its carcinogenic properties. The mode of action has been postulated to be that of a genotoxic carcinogen, with formation of promutagenic DNA adducts being essential for mutagenesis and carcinogenesis. In rodents, isoprene-induced tumors show unique point mutations (A→T transversions) in the K-ras protooncogene at codon 61. Therefore, we investigated adducts formed after reaction of 2′-deoxyadenosine (dAdo1) with the two monoepoxides of isoprene, 2-ethenyl-2-methyloxirane (IP-1,2-O) and propen-2-yloxirane (IP-3,4-O), under physiological conditions. The formation of N1–2′-deoxyinosine (N1-dIno) due to deamination of N1-dAdo adducts was of particular interest, since N1-dIno adducts are suspected to have high mutagenic potential based on in vitro experiments. Major stable adducts were identified by HPLC, UV-Spectrometry and LC-MS/MS and characterized by 1H and 1H,13C HSQC and NMR experiments. Adducts of IP-1,2-O that were fully identified are: R,S-C1-N6-dAdo, R-C2-N6-dAdo, and S-C2-N6-dAdo; adducts of IP-3,4-O are: S-C3-N6-dAdo, R-C3-N6-dAdo, R,S-C4-N6-dAdo, S-C4-N1-dIno, R-C4-N1-dIno, R-C3-N1-dIno, S-C3-N1-dIno, and C3-N7-Ade. Both monoepoxides formed adducts on the external and internal oxirane carbons. This is the first study to describe adducts of isoprene monoepoxides with dAdo. Characterization of adducts formed by isoprene monoepoxides with deoxynucleosides and subsequently with DNA represent the first step toward evaluating their potential for being converted into a mutation, or as biomarkers of isoprene metabolism and exposure

    The vast majority of studies assessing the prejudice reduction properties of imagined contact have focused so far on the immediate effects of the intervention. In an attempt to contribute to the literature examining the long-term effects of imagined contact, the two studies reported in this paper tested the immediate and long-term effects of imagined contact on outgroup attitudes, intergroup anxiety, and behavioral intentions in Experiment 1, and also on contact self-efficacy in Experiment 2. Both studies were conducted in a context of entrenched intergroup conflict, Cyprus. The results supported the effectiveness of imagined contact in eliciting more positive attitudes, lower levels of anxiety, more positive behavioral intentions, and higher contact self-efficacy when these were measured immediately after contact. However, evidence for the endurance of these effects was systematically found only for outgroup attitudes and intergroup anxiety. While these results speak to the ability of imagined contact to lead to long-term changes in important and commonly studied intergroup outcomes, lack of consistent evidence regarding its ability to yield lasting changes on variables pertaining to intended behavior toward the outgroup compose a challenge for the intervention

    Estimating the Capacity for ART Provision in Tanzania with the Use of Data on Staff Productivity and Patient Losses

    BACKGROUND: International targets for access to antiretroviral therapy (ART) have over-estimated the capacity of health systems in low-income countries in Sub-Saharan Africa. The WHO target for number on treatment by end 2005 for Tanzania was 10 times higher than actually achieved. The target of the national Care and Treatment Plan (CTP) was also not reached. We aimed at estimating the capacity for ART provision and created five scenarios for ART production given existing resource limitations. METHODS: A situation analysis including scrutiny of staff factors, such as available data on staff and patient factors including access to ART and patient losses, made us conclude that the lack of clinical staff is the main limiting factor for ART scale-up, assuming that sufficient drugs and supplies are provided by donors. We created a simple formula to estimate the number of patients on ART based on availability and productivity of clinical staff, time needed to initiate vs maintain a patient on ART and patient losses using five different scenarios with varying levels of these parameters. FINDINGS: Our scenario assuming medium productivity (40% higher than that observed in 2002) and medium loss of patients (20% in addition to 15% first-year mortality) coincides with the actual reported number of patients initiated on ART up to 2008, but is considerably below the national CTP target of 90% coverage for 2009, corresponding to 420,000 on ART and 710,000 life-years saved (LY's). Our analysis suggests that a coverage of 40% or 175,000 on treatment and 350,000 LY's saved is more achievable. CONCLUSION: A comparison of our scenario estimations and actual output 2006-2008 indicates that a simple user-friendly dynamic model can estimate the capacity for ART scale-up in resource-poor settings based on identification of a limiting staff factor and information on availability of this staff and patient losses. Thus, it is possible to set more achievable targets

    Comparative transcriptome analysis of AP2/EREBP gene family under normal and hormone treatments, and under two drought stresses in NILs setup by Aday Selection and IR64

    The AP2/EREBP genes play various roles in developmental processes and in stress-related responses in plants. Genome-wide microarrays based on the gene expression profiles of the AP2/EREBP family were analyzed under conditions of normal growth and drought stress. The preferential expression of fifteen genes was observed in specific tissues, suggesting that these genes may play important roles in vegetative and reproductive stages of growth. A large number of redundant genes were differentially expressed following phytohormone treatments (NAA, GA3, KT, SA, JA, and ABA). To investigate the gene expression responses in the root, leaf, and panicle of three rice genotypes, two drought stress conditions were applied using the fraction of transpirable soil water (FTSW) under severe (0.2 FTSW), mild (0.5 FTSW), and control (1.0 FTSW) conditions. Following treatment, transcriptomic analysis using a 44-K oligoarray from Agilent was performed on all the tissue samples. We identified common and specific genes in all tissues from two near-isogenic lines, IR77298-14-1-2-B-10 (drought tolerant) and IR77298-14-1-2-B-13 (drought susceptible), under drought stress conditions. The majority of the genes that were activated in the IR77298-14-1-2-B-10 line were members of the AP2/EREBP gene family. Non-redundant genes (sixteen) were found in the drought-tolerant line, and four genes were selected as candidate novel reference genes because of their higher expression levels in IR77298-14-1-2-B-10. Most of the genes in the AP2, B3, and B5 subgroups were involved in the panicle under severe stress conditions, but genes from the B1 and B2 subgroups were down-regulated in the root. Of the four subfamilies, RAV exhibited the highest number of up-regulated genes (80%) in the panicle under severe stress conditions in the drought-tolerant line compared to Minghui 63 under normal conditions, and the gene structures of the RAV subfamily may be involved in the response to drought stress in the flowering stage. These results provide a useful reference for the cloning of candidate genes from the specific subgroup for further functional analysis
