The sum of its parts? Sources of local legitimacy
The article analyses the sources of local actorsâ legitimacy perceptions towards international peacebuilding operations. Local legitimacy perceptions are increasingly recognised as shaping local behaviour towards international peacebuilding, which influences the effective functioning of the operation. Legitimacy debates in peacebuilding are either absent or imported from the literature on domestic legitimacy, without respect to the specific temporal and spatial situation of international operations. The article first explores which legitimacy sources influence local legitimacy perceptions of international peacebuilding operations. It finds that two sources are relevant: output and procedure. Second, it investigates how exactly legitimacy arises from them. In doing so, it demonstrates that output and procedure are umbrella terms comprising several sub-elements which influence legitimacy in different, sometimes contradictory, ways. Finally, the article empirically explores which of the sources are important to local actorsâ legitimacy perceptions using field data from the EU peacebuilding operations EULEX in Kosovo and EUPM Bosnia-Herzegovina

Se presenta al colectivo âAS6030â, conformado por arquitectos que trabajan en ĂĄreas relacionadas con la sustentabilidad ambiental desde distintos ĂĄmbitos. El grupo, formado en la Universidad de Buenos Aires entre los años ochenta y dos mil, es parte de lo que se podrĂa denominar, âuna segunda generaciĂłnâ especializada en sustentabilidad ambiental en Argentina. Inicialmente se conforma como un espacio para el intercambio de prĂĄcticas mĂĄs amigable con el ambiente, factibles en nuestro medio, sin resignar calidad ni afĂĄn proyectual. ZIC1 Is Downregulated through Promoter Hypermethylation, and Functions as a Tumor Suppressor Gene in Colorectal Cancer
The transcription factor, Zinc finger of the cerebellum (ZIC1), plays a crucial role in vertebrate development. Recently, ZIC1 has also been found to participate in the progression of human cancers, including medulloblastomas, endometrial cancers, and mesenchymal neoplasms. However, the function of ZIC1 in colon cancer progression has not been defined. In this study, we demonstrate ZIC1 to be silenced or significantly downregulated in colon cancer cell lines. These effects were reversed by demethylation treatment with 5-aza-2âČ-deoxycytidine (Aza). ZIC1 expression is also significantly downregulated in primary colorectal cancer tissues relative to adjacent non-tumor tissues (pâ=â0.0001). Furthermore, methylation of ZIC1 gene promoter is frequently detected in primary tumor tissues (85%, 34/40), but not in adjacent non-tumor tissues. Ectopic expression of ZIC1 suppresses cell proliferation and induces apoptosis, which is associated with MAPK and PI3K/Akt pathways, as well as the Bcl-xl/Bad/Caspase3 cascade. To identify target candidates of ZIC1, we employed cDNA microarray and found that 337 genes are downregulated and 95 genes upregulated by ectopic expression of ZIC1, which were verified by 10 selected gene expressions by qRT-PCR. Taken together, our results suggest that ZIC1 may potentially function as a tumor suppressor gene, which is downregulated through promoter hypermethylation in colorectal cancers
Identification of novel targets for breast cancer by exploring gene switches on a genome scale
<p>Abstract</p> <p>Background</p> <p>An important feature that emerges from analyzing gene regulatory networks is the "switch-like behavior" or "bistability", a dynamic feature of a particular gene to preferentially toggle between two steady-states. The state of gene switches plays pivotal roles in cell fate decision, but identifying switches has been difficult. Therefore a challenge confronting the field is to be able to systematically identify gene switches.</p> <p>Results</p> <p>We propose a top-down mining approach to exploring gene switches on a genome-scale level. Theoretical analysis, proof-of-concept examples, and experimental studies demonstrate the ability of our mining approach to identify bistable genes by sampling across a variety of different conditions. Applying the approach to human breast cancer data identified genes that show bimodality within the cancer samples, such as estrogen receptor (ER) and ERBB2, as well as genes that show bimodality between cancer and non-cancer samples, where tumor-associated calcium signal transducer 2 (TACSTD2) is uncovered. We further suggest a likely transcription factor that regulates TACSTD2.</p> <p>Conclusions</p> <p>Our mining approach demonstrates that one can capitalize on genome-wide expression profiling to capture dynamic properties of a complex network. To the best of our knowledge, this is the first attempt in applying mining approaches to explore gene switches on a genome-scale, and the identification of TACSTD2 demonstrates that single cell-level bistability can be predicted from microarray data. Experimental confirmation of the computational results suggest TACSTD2 could be a potential biomarker and attractive candidate for drug therapy against both ER+ and ER- subtypes of breast cancer, including the triple negative subtype.</p
Learning to Think Iconically in the Human and Social Sciences: Iconic Standards of Understanding as a Pivotal Challenge for Method Development
Theoretically as well as alongside an empirical research idea, this paper outlines conditions for the development of social scientific empirical methods able to further exploit the iconic potential of the image. Reconstructing the role of formal pictorial elements for the standards of understanding within the medium âimageâ is considered pivotal in this endeavor. Within the context of language, standards of communication have already been extensively researched. The linguistic format of the narrative, for instance, is well studied. Up to now, though, comparable formal vehicles of iconic semantics have only been examined in aesthetics and art history. Nevertheless, standards of iconic understanding are part of our implicit knowledge, are incessantly in use in everyday practice and, thus, the basis of everyday identity formation. With the help of empirical methods based on an iconic logos we can deepen our understanding of orientations, longings, and anxieties of our time that are often silently conveyed by images. The James Webb Space Telescope Mission
Twenty-six years ago a small committee report, building on earlier studies,
expounded a compelling and poetic vision for the future of astronomy, calling
for an infrared-optimized space telescope with an aperture of at least .
With the support of their governments in the US, Europe, and Canada, 20,000
people realized that vision as the James Webb Space Telescope. A
generation of astronomers will celebrate their accomplishments for the life of
the mission, potentially as long as 20 years, and beyond. This report and the
scientific discoveries that follow are extended thank-you notes to the 20,000
team members. The telescope is working perfectly, with much better image
quality than expected. In this and accompanying papers, we give a brief
history, describe the observatory, outline its objectives and current observing
program, and discuss the inventions and people who made it possible. We cite

Technikgenese: EinfluĂfaktoren der Technisierung jenseits traditioneller Technikfolgenforschung
Ausgehend davon, daĂ das VerhĂ€ltnis von Technikgeneseforschung zu Technikfolgenforschung komplementĂ€r sein muĂ, werden in dem Beitrag die strukturellen, institutionellen und organisatorischen, die kognitiven wie perzeptiven Voraussetzungen dafĂŒr untersucht, welche Technikfolgen wie systematisch und rational in frĂŒhen Phasen der Technikentstehung wahrgenommen, berĂŒcksichtigt und technisch umgesetzt werden und welche nicht, um in der sozialwissenschaftlichen Rekonstruktion der Bedingungen technischer Innovationen die gesellschaftlichen und organisatorischen EinfluĂparameter, die Selektionsprozesse bei der NeueinfĂŒhrung von Techniken prĂ€gen, manifest werden zu lassen. Ziele und Grundlagen der sozialwissenschaftlichen Technikgeneseforschung werden dargestellt. Indem Technikgenese in organisationskulturellen Kontexten analysiert wird, wird ein neuer Forschungsansatz konzipiert, der im Kern beansprucht, das Selektions- und Entscheidungsverhalten von Organisationen mit Hilfe von ErklĂ€rungskategorien zu erfassen. Der zentrale Beitrag des organisationskulturellen Forschungsansatzes der Technikgenese liegt in der Möglichkeit, diejenigen Faktoren herauszukristallisieren, die eine Organisation dazu befĂ€higen, innovative Impulse selbst zu generieren oder entsprechende Stimuli aus ihrer Umwelt aufzunehmen und umzusetzen.
