    Abstract, emotional and concrete concepts and the activation of mouth-hand effectors

    According to embodied and grounded theories, concepts are grounded in sensorimotor systems. The majority of evidence supporting these views concerns concepts referring to objects or actions, while evidence on abstract concepts is more scarce. Explaining how abstract concepts such as ‘‘freedom’’ are represented would thus be pivotal for grounded theories. According to some recent proposals, abstract concepts are grounded in both sensorimotor and linguistic experience, thus they activate the mouth motor system more than concrete concepts. Two experiments are reported, aimed at verifying whether abstract, concrete and emotional words activate the mouth and the hand effectors. In both experiments participants performed first a lexical decision, then a recognition task. In Experiment 1 participants responded by pressing a button either with the mouth or with the hand, in Experiment 2 responses were given with the foot, while a button held either in the mouth or in the hand was used to respond to catch-trials. Abstract words were slower to process in both tasks (concreteness effect). Across the tasks and experiments, emotional concepts had instead a fluctuating pattern, different from those of both concrete and abstract concepts, suggesting that they cannot be considered as a subset of abstract concepts. The interaction between type of concept (abstract, concrete and emotional) and effector (mouth, hand) was not significant in the lexical decision task, likely because it emerged only with tasks implying a deeper processing level. It reached significance, instead, in the recognition tasks. In both experiments abstract concepts were facilitated in the mouth condition compared to the hand condition, supporting our main prediction. Emotional concepts instead had a more variable pattern. Overall, our findings indicate that various kinds of concepts differently activate the mouth and hand effectors, but they also suggest that concepts activate effectors in a flexible and task-dependent wa

    Methodological factors influencing measurement and processing of plasma reelin in humans

    BACKGROUND: Reelin, intensively studied as an extracellular protein that regulates brain development, is also expressed in a variety of tissues and a circulating pool of reelin exists in adult mammals. Here we describe the methodological and biological foundation for carrying out and interpreting clinical studies of plasma reelin. RESULTS: Reelin in human plasma was sensitive to proteolysis, freeze-thawing and heating during long-term storage, sample preparation and electrophoresis. Reelin in plasma was a dimer under denaturing conditions. Boiling of samples resulted in laddering, suggesting that each of the 8 repeats expressed in reelin contains a heat-labile covalent bond susceptible to breakage. Urinary-type and tissue-type plasminogen activator converted reelin to a discrete 310 kDa fragment co-migrating with the major immunoreactive reelin fragment seen in plasma and also detected in brain. (In contrast, plasmin produced a spectrum of smaller unstable reelin fragments.) We examined archival plasma of 10 pairs of age-matched male individuals differing in repeat length of a CGG repeat polymorphism of the 5'-untranslated region of the reelin gene (both alleles < 11 repeats vs. one allele having >11 repeats). Reelin 310 kDa band content was lower in subjects having the long repeats in all 10 pairs, by 25% on average (p < 0.001). In contrast, no difference was noted for amyloid precursor protein. CONCLUSIONS: Our studies indicate the need for caution in measuring reelin in archival blood samples, and suggest that assays of plasma reelin should take into account three dimensions that might vary independently: a) the total amount of reelin protein; b) the relative amounts of reelin vs. its proteolytic processing products; and c) the aggregation state of the native protein. Reelin-plasminogen activator interactions may affect their roles in synaptic plasticity. Our results also suggest that the human CGG repeat polymorphism affects reelin gene expression, and may affect susceptibility to human disease

    The response of the male freshwater goby to natural and synthetic male courtship sound playback following exposure to different female sexual sitimuli.

    Among teleosts, field and laboratory experiments have shown the playback of male courtship sounds often elicits competitive courtship and sound emission in the territorial male. However, the importance of female stimulation for the male response to the sound is poorly understood. In this study, playback experiments with the freshwater goby, Padogobius martensii, examined the response of resident males (i.e. males individually housed within laboratory tanks for at least 5 days) to natural and synthetic male courtship sounds after exposure to chemical or visual stimuli from a ripe female. All playback tests consisted of one experimental (sound playback) and two control treatments, all of equal duration. Three experiments were conducted on a first group of 12 males using natural courtship sounds. Experiment 1 consisted of playing back the sound to a male that had not exposed to female stimulation for at least 24 hr. The same male was exposed to the “female pheromone” by dropping a few cm3 of female holding water into the male’s tank, just prior to playback (Experiment 2), or to the view of a live ripe female in a close-by tank, during playback (Experiment 3). The sound playback failed to elicit positive responses by males not exposed to female stimuli (Experiment 1), and did not increase courtship activity of the male in visual contact with the female (Experiment 3). However, it increased swimming activity and facilitated courtship and sound production in the male after an increase in sexual arousal by prior chemical stimulation (Experiment 2). In Experiment 4, the synthetic version of the courtship sound and one deprived of pulse-rate modulation and harmonic content (pure tone) were played back to 10 resident males following prior chemical stimulation. The synthetic courtship sound, but not the pure tone, was effective in eliciting positive responses by the sexually aroused male. Functional implications of the responses to the courtship sound for the territorial male are discussed. The results are relevant to the understanding of the role of sound communication and interception among gobies and to develop playback experiments in the field

    Learn to Bet: Using Reinforcement Learning to Improve Vehicle Bids in Auction-Based Smart Intersections

    With the advent of IoT, cities will soon be populated by autonomous vehicles and managed by intelligent systems capable of actively interacting with city infrastructures and vehicles. In this work, we propose a model based on reinforcement learning that teaches to autonomous connected vehicles how to save resources while navigating in such an environment. In particular, we focus on budget savings in the context of auction-based intersection management systems. We trained several models with Deep Q-learning by varying traffic conditions to find the most performance-effective variant in terms of the trade-off between saved currency and trip times. Afterward, we compared the performance of our model with previously proposed and random strategies, even under adverse traffic conditions. Our model appears to be robust and manages to save a considerable amount of currency without significantly increasing the waiting time in traffic. For example, the learner bidder saves at least 20% of its budget with heavy traffic conditions and up to 74% in lighter traffic with respect to a standard bidder, and around three times the saving of a random bidder. The results and discussion suggest practical adoption of the proposal in a foreseen future real-life scenario

    Free-carrier screening of polarization fields in wurtzite GaN/InGaN laser structures

    The free-carrier screening of macroscopic polarization fields in wurtzite GaN/InGaN quantum wells lasers is investigated via a self-consistent tight-binding approach. We show that the high carrier concentrations found experimentally in nitride laser structures effectively screen the built-in spontaneous and piezoelectric polarization fields, thus inducing a ``field-free'' band profile. Our results explain some heretofore puzzling experimental data on nitride lasers, such as the unusually high lasing excitation thresholds and emission blue-shifts for increasing excitation levels.Comment: RevTeX 4 pages, 4 figure

    Cell line derived xenograft mouse models are a suitable in vivo model for studying tumor budding in colorectal cancer

    Tumor budding (TB) is an important prognostic parameter in colorectal cancer (CRC) and associated with metastasis. However, the mechanisms of TB have not been fully elucidated and a major limitation is the absence of in vivo models. Here, we determine the suitability of human cell line derived xenografts (CDX) as models of TB in CRC. Pan-cytokeratin (CK)-stained next-generation Tissue Microarrays (ngTMA) of two CDX models (HT-29, n = 12 and HCT-8, n = 8) and human CRC (n = 27 high-grade and 25 low-grade budding tumors, each) were evaluated for TB. Immunohistochemistry for E-cadherin, β-catenin, Ki-67, ZEB1, and TWIST1 was performed. HT-29 and HCT-8 were predominantly high-grade and no/low-grade TB tumors, respectively. TB counts in the tumor center (intratumoral budding, ITB) were significantly higher in HT-29 CDX tumors compared to human CRC (p = 0.0099). No difference was found in TB counts at the invasion front (peritumoral budding, PTB; p=0.07). ITB and PTB were strongly correlated (r = 0.438 and r = 0.62 in CDX and human CRC, respectively). Immunohistochemistry profiles were comparable in CDX and human CRC tissues. TB in the CDX mouse models is phenotypically similar to human CRCs and highlights comparable protein profiles.     Are fluid inclusions in gypsum reliable paleoenvironmental indicators? An assessment of the evidence from the Messinian evaporites

    The paleosalinity of water from which the gypsum precipitated during the Messinian salinity crisis is a controversial issue. Recent microthermometry studies on primary fluid inclusions in gypsum provided very low salinity values not compatible with precipitation from seawater, and suggested strong mixing between seawater and nonmarine waters enriched in calcium sulfate. We applied a new microthermometric protocol on gypsum crystals from nine Mediterranean sections that were experimentally stretched to measure a larger population of fluid inclusions. The results show salinities ranging from 9 to 238 wt‰ NaCl equivalent, largely falling within the evaporation path of normal seawater. The data from previous studies were obtained mostly from those fluid inclusions capable of nucleating a stable bubble after a weak stretching, which probably correspond to those having a lower salinity acquired through post-depositional crack-and-seal processes. Our data suggest instead that the primary gypsum precipitated from a marine brine, later modified by post-trapping processes during tectonics and exhumation

    Sea Tide Influence on Ice Flow of David Drygalski’s Ice Tongue Inferred from Geodetic GNSS Observations and SAR Offset Tracking Analysis

    David Glacier and Drygalski Ice Tongue are massive glaciers in Victoria Land, Antarctica. The ice from the East Antarctic Ice Sheet is drained through the former, and then discharged into the western Ross Sea through the latter. David Drygalski is the largest outlet glacier in Northern Victoria Land, floating kilometers out to sea. The floating and grounded part of the David Glacier are the main focus of this article. During the XXI Italian Antarctic Expedition (2005-2006), within the framework of the National Antarctic Research Programme (PNRA), two GNSS stations were installed at different points: the first close to the grounding line of David Glacier, and the second approximately 40 km downstream of the first one. Simultaneous data logging was performed by both GNSS stations for 24 days. In the latest data processing, the kinematic PPP technique was adopted to evaluate the dominant diurnal components and the very small semi-diurnal variations in ice motion induced by the ocean tide and the mean ice flow rates of both GNSS stations. Comparison of the GNSS time series with predicted ocean tide calculated from harmonic coefficients of the nearest tide gauge stations, installed at Cape Roberts and Mario Zucchelli Station, highlight different local response of the glacier to ocean tide, with a minor amplitude of vertical motion at a point partially anchored at the bedrock close to the grounding line. During low tide, the velocity of the ice flow reaches its daily maximum, in accordance with the direction of seawater outflow from the fjord into the ocean, while the greatest daily tidal excursion generates an increase in the horizontal ice flow velocity. With the aim to extend the analysis in spatial terms, five COSMO-SkyMED Stripmap scenes were processed.     Expression of citrulline and homocitrulline residues in the lungs of non-smokers and smokers : implications for autoimmunity in rheumatoid arthritis

    Introduction: Smoking is a well-established risk factor for rheumatoid arthritis (RA), and it has been proposed that smoking-induced citrullination renders autoantigens immunogenic. To investigate this mechanism, we examined human lung tissue from 40 subjects with defined smoking status, with or without chronic obstructive pulmonary disease (COPD), and control tissues from other organs for citrullinated proteins and the deiminating enzymes peptidylarginine deiminase type-2 (PAD2) and -4 (PAD4). Methods: Lung tissue samples, dissected from lobectomy specimens from 10 never smokers, 10 smokers without airflow limitation, 13 COPD smokers and eight COPD ex-smokers, and control tissue samples (spleen, skeletal muscle, liver, ovary, lymph node, kidney and heart), were analysed for citrullinated proteins, PAD2 and PAD4 by immunoblotting. Citrulline and homocitrulline residues in enolase and vimentin were analysed by partial purification by gel electrophoresis followed by mass spectrometry in 12 of the lung samples and one from each control tissues. Band intensities were scored semi-quantitatively and analysed by two-tailed Mann-Whitney T-test. Results: Within the lung tissue samples, citrullinated proteins, PAD2 and PAD4 were found in all samples, with an increase in citrullination in COPD (P = 0.039), but minimal difference between smokers and non-smokers (P = 0.77). Citrullination was also detected at lower levels in the tissues from other organs, principally in lymph node, kidney and skeletal muscle. Mass spectrometry of the lung samples showed that vimentin was citrullinated at positions 71, 304, 346, 410 and 450 in non-smokers and smokers both with and without COPD. A homocitrulline at position 104 was found in four out of six COPD samples and one out of six non-COPD. Citrulline-450 was also found in three of the control tissues. There were no citrulline or homocitrulline residues demonstrated in a-enolase. Conclusions: We have shown evidence of citrullination of vimentin, a major autoantigen in RA, in both non-smokers and smokers. The increase in citrullinated proteins in COPD suggests that citrullination in the lungs of smokers is mainly due to inflammation. The ubiquity of citrullination of vimentin in the lungs and other tissues suggests that the relationship between smoking and autoimmunity in RA may be more complex than previously thought

    Alice in wonderland: experimental jurisprudence on the internal point of view

    Humans have this extraordinary cognitive ability: They imagine inexistent objects, they treat them as if they were real, and by doing so they make them real. They thus give rise to a shared institutional reality that enables them to cooperate in ways that would be impossible otherwise. In this paper, we would like to revisit the account that HLA Hart gives of the practice of collective acceptance that makes a legal system possible. We try to provide an explanation of what Hart calls the 'internal point of view', on the basis of experiments on institutional concepts, drawing on the paradigm known as 'embodied cognition'. Experts and non-experts in law rated the role of several cognitive dimensions for a list of words referring to two kinds of abstract concepts (institutional and theoretical/scientific) and two kinds of concrete ones (food and artifact). Institutional concepts were distinguished into pure-institutional (e.g., 'contract', 'state', 'property') and meta-institutional (e.g., 'norm', 'duty', 'justice'). The results provide an empirical account of how our way of thinking about institutions changes as we acquire expertise in the legal field, thus shading light on the cognitive underpinnings of the 'internal point of view'
