    A paperfluidic platform to detect Neisseria gonorrhoeae in clinical samples

    Globally, the microbe Neisseria gonorrhoeae (NG) causes 106 million newly documented sexually transmitted infections each year. Once appropriately diagnosed, NG infections can be readily treated with antibiotics, but high-risk patients often do not return to the clinic for treatment if results are not provided at the point of care. A rapid, sensitive molecular diagnostic would help increase NG treatment and reduce the prevalence of this sexually transmitted disease. Here, we report on the design and development of a rapid, highly sensitive, paperfluidic device for point-of-care diagnosis of NG. The device integrates patient swab sample lysis, nucleic acid extraction, thermophilic helicase-dependent amplification (tHDA), an internal amplification control (NGIC), and visual lateral flow detection within an 80 min run time. Limits of NG detection for the NG/NGIC multiplex tHDA assay were determined within the device, and clinical performance was validated retroactively against qPCR-quantified patient samples in a proof-of-concept study. This paperfluidic diagnostic has a clinically relevant limit of detection of 500 NG cells per device with analytical sensitivity down to 10 NG cells per device. In triplicate testing of 40 total urethral and vaginal swab samples, the device had 95% overall sensitivity and 100% specificity, approaching current laboratory-based molecular NG diagnostics.     Comparison of Nitrogen Incorporation in Tholins Produced by FUV Irradiation and Spark Discharge

    The discovery of very heavy ions (Coates et al., 2007) in Titan's thermosphere has dramatically altered our understanding of the processes involved in the formation of the complex organic aerosols that comprise Titan's characteristic haze. Before Cassini's arrival, it was believed that aerosol production began in the stratosphere where the chemical processes were predominantly initiated by FUV radiation. This understanding guided the design of Titan atmosphere simulation experiments. However, the energy environment of the thermosphere is significantly different than the stratosphere; in particular there is a greater flux of EUV photons and energetic particles available to initiate chemical reactions, including the destruction of N2. in the upper atmosphere. Using a High Resolution Time-of-Flight Aerosol Mass Spectrometer (HR-ToF-AMS), we have obtained in situ composition measurements of aerosol particles (so'called "tholins") produced in CH4/N2 gas mixtures subjected to either FUV radiation (deuterium lamp, 115-400 nm) (Trainer et al., 2012) or a spark discharge. A comparison of the composition of tholins produced using the two different energy sources will be presented, in particular with regard to the variation in nitrogen content of the two types of tholin. Titan's aerosols are known to contain significant amounts of nitrogen (Israel et al., 2005) and therefore understanding the role of nitrogen in the aerosol chemistry is important to further our knowledge of the formation and evolution of aerosols in Titan's atmosphere

    The hidden X-ray breaks in afterglow light curves

    Gamma-Ray Burst (GRB) afterglow observations in the Swift era have a perceived lack of achromatic jet breaks compared to the BeppoSAX, or pre-Swift era. Specifically, relatively few breaks, consistent with jet breaks, are observed in the X-ray light curves of these bursts. If these breaks are truly missing, it has serious consequences for the interpretation of GRB jet collimation and energy requirements, and the use of GRBs as standard candles. Here we address the issue of X-ray breaks which are possibly 'hidden' and hence the light curves are misinterpreted as being single power-laws. We show how a number of precedents, including GRB 990510 & GRB 060206, exist for such hidden breaks and how, even with the well sampled light curves of the Swift era, these breaks may be left misidentified. We do so by synthesising X-ray light curves and finding general trends via Monte Carlo analysis. Furthermore, in light of these simulations, we discuss how to best identify achromatic breaks in afterglow light curves via multi-wavelength analysis.Comment: 4 pages, contributed talk, submitted to the proceedings of Gamma Ray Bursts 2007, Santa Fe, New Mexico, November 5-9 200

    Longitudinal coupled-bunch instability around 1 GHZ at the CERN PS booster

    The fast-growing "Ring 4" instability occurring at intensities above 6.5 1012 protons in the top one of the four rings of the CERN PS Booster (PSB) is finally explained by an asymmetry in the 40 vacuum pump manifolds common to all rings. Impedance measurements (by wire method) and numerical calculations show a sharp resonant peak (Q~2000) at 1100 MHz and shunt impedances two times higher for the Ring 4 ports as compared to the other rings. This factor is sufficient to explain that the threshold of the instability falls below the maximum intensity only in Ring 4. A final, but labor-intensive and expensive, cure consists of inserting short-circuiting sleeves into all 160 beam ports.     Get the Story Straight: Contextual Repetition Promotes Word Learning from Storybooks

    Although shared storybook reading is a common activity believed to improve the language skills of preschool children, how children learn new vocabulary from such experiences has been largely neglected in the literature. The current study systematically explores the effects of repeatedly reading the same storybooks on both young children's fast and slow mapping abilities. Specially created storybooks were read to 3-year-old children three times during the course of 1 week. Each of the nine storybooks contained two novel name–object pairs. At each session, children either heard three different stories with the same two novel name–object pairs or the same story three times. Importantly, all children heard each novel name the same number of times. Both immediate recall and retention were tested with a four-alternative forced-choice task with pictures of the novel objects. Children who heard the same stories repeatedly were very accurate on both the immediate recall and retention tasks. In contrast, children who heard different stories were only accurate on immediate recall during the last two sessions and failed to learn any of the new words. Overall, then, we found a dramatic increase in children's ability to both recall and retain novel name–object associations encountered during shared storybook reading when they heard the same stories multiple times in succession.     Pitfalls of lateral external fixation for supracondylar humeral fractures in children

    Introduction: Closed reduction and percutaneous pinning is a standard treatment for dislocated supracondylar humeral fractures in children. However, the management of these fractures remains challenging. The aim of this study was to evaluate lateral external fixation as a treatment alternative for these fractures. Materials and methods: All supracondylar fractures treated with lateral external fixation between 2005 and 2007 were evaluated retrospectively. Long-term outcome was assessed with regards to carrying angle, malalignment, and motion. Results: Twenty-eight patients with Gartland type III fractures and one with a Y-type fracture were included in the study (mean age 6.5years). Cosmetic results were excellent in 88%, good in 8%, and fair in one patient. Functional results were excellent in 83%, good in 10%, and fair in 7%. However, 3 patients (10%) showed complete radial palsy postoperatively. In all of these patients, high insertion of the proximal pin (2.9-3.6cm above the fracture) was noted. On revision, one superficial lesion and one total transection of the nerve at the level of the proximal pin was detected. One patient showed no macroscopic damage. The transected nerve was reconstructed using an autograft, and all patients completely recovered within 2-6months. Conclusion: Lateral external fixation is an alternative method for the treatment of displaced or unstable supracondylar fractures in children, facilitating reduction and improving fracture stability. However, iatrogenic radial nerve injury is a risk, and we therefore strongly recommend inserting the proximal pin under direct vision within 2cm from the fracture line using a drill sleev

    The Peculiar Multi-Wavelength Evolution Of V1535 Sco

    We present multi-wavelength observations of the unusual nova V1535 Sco throughout its outburst in 2015. Early radio observations were consistent with synchrotron emission, and early X-ray observations revealed the presence of high-energy (>1 keV) photons. These indicated that strong shocks were present during the first ~2 weeks of the nova's evolution. The radio spectral energy distribution was consistent with thermal emission from week 2 to week 6. Starting in week 7, the radio emission again showed evidence of synchrotron emission and there was an increase in X-ray emission, indicating a second shock event. The optical spectra show evidence for at least two separate outflows, with the faster outflow possibly having a bipolar morphology.
