    Incidence and distribution of Heterobasidion and Armillaria and their influence on canopy gap formation in unmanaged mountain pine forests in the Swiss Alps

    Various disturbance factors on different spatial scales can lead to the creation of canopy gaps in forest ecosystems. In this study, we investigated the role of root rot fungi in the formation of canopy gaps in the Swiss National Park in the Central Alps. Dying or recently dead mountain pine (Pinus mugo subsp. uncinata trees (n=172) and saplings (n=192) from 42 canopy gaps were assessed for Armillaria and Annosum root rot. Heterobasidion annosum s.str. proved to be the dominant pathogen and was isolated from 49% of the trees and 64% of the saplings. Armillaria was found on 13% of the trees and 20% of the saplings. Three Armillaria species, A. borealis, A. cepistipes, and A. ostoyae, were identified. Armillaria ostoyae was the most frequent species, accounting for 72% of all Armillaria isolates. A total of 31 (74%) gaps were associated with H. annosum, and six (14%) with A. ostoyae. The remaining gaps were either associated with both pathogens (7%) or with other, unknown, factors (5%). Our findings suggest that the two pathogenic fungi, H. annosum s.str. and A. ostoyae, are the main reason for the large-scale mortality of mountain pines and the creation of canopy gaps in high elevation forests of the Swiss National Par

    Overexpression of connexin 43 using a retroviral vector improves electrical coupling of skeletal myoblasts with cardiac myocytes in vitro.

    BACKGROUND: Organ transplantation is presently often the only available option to repair a damaged heart. As heart donors are scarce, engineering of cardiac grafts from autologous skeletal myoblasts is a promising novel therapeutic strategy. The functionality of skeletal muscle cells in the heart milieu is, however, limited because of their inability to integrate electrically and mechanically into the myocardium. Therefore, in pursuit of improved cardiac integration of skeletal muscle grafts we sought to modify primary skeletal myoblasts by overexpression of the main gap-junctional protein connexin 43 and to study electrical coupling of connexin 43 overexpressing myoblasts to cardiac myocytes in vitro. METHODS: To create an efficient means for overexpression of connexin 43 in skeletal myoblasts we constructed a bicistronic retroviral vector MLV-CX43-EGFP expressing the human connexin 43 cDNA and the marker EGFP gene. This vector was employed to transduce primary rat skeletal myoblasts in optimised conditions involving a concomitant use of the retrovirus immobilising protein RetroNectin and the polycation transduction enhancer Transfectam. The EGFP-positive transduced cells were then enriched by flow cytometry. RESULTS: More than four-fold overexpression of connexin 43 in the transduced skeletal myoblasts, compared with non-transduced cells, was shown by Western blotting. Functionality of the overexpressed connexin 43 was demonstrated by microinjection of a fluorescent dye showing enhanced gap-junctional intercellular transfer in connexin 43 transduced myoblasts compared with transfer in non-transduced myoblasts. Rat cardiac myocytes were cultured in multielectrode array culture dishes together with connexin 43/EGFP transduced skeletal myoblasts, control non-transduced skeletal myoblasts or alone. Extracellular field action potential activation rates in the co-cultures of connexin 43 transduced skeletal myoblasts with cardiac myocytes were significantly higher than in the co-cultures of non-transduced skeletal myoblasts with cardiac myocytes and similar to the rates in pure cultures of cardiac myocytes. CONCLUSION: The observed elevated field action potential activation rate in the co-cultures of cardiac myocytes with connexin 43 transduced skeletal myoblasts indicates enhanced cell-to-cell electrical coupling due to overexpression of connexin 43 in skeletal myoblasts. This study suggests that retroviral connexin 43 transduction can be employed to augment engineering of the electrocompetent cardiac grafts from patients own skeletal myoblasts

    Palaeoclimate characteristics in interior Siberia of MIS 6–2: first insights from the Batagay permafrost mega-thaw slump in the Yana Highlands

    Syngenetic permafrost deposits formed extensively on and around the arising Beringian subcontinent during the Late Pleistocene sea level lowstands. Syngenetic deposition implies that all material, both mineral and organic, freezes parallel to sedimentation and remains frozen until degradation of the permafrost. Permafrost is therefore a unique archive of Late Pleistocene palaeoclimate. Most studied permafrost outcrops are situated in the coastal lowlands of northeastern Siberia; inland sections are, however, scarcely available. Here, we describe the stratigraphical, cryolithological, and geochronological characteristics of a permafrost sequence near Batagay in the Siberian Yana Highlands, the interior of the Sakha Republic (Yakutia), Russia, with focus on the Late Pleistocene Yedoma ice complex (YIC). The recently formed Batagay mega-thaw slump exposes permafrost deposits to a depth of up to 80 m and gives insight into a climate record close to Verkhoyansk, which has the most severe continental climate in the Northern Hemi- sphere. Geochronological dating (optically stimulated luminescence, OSL, and 14C ages) and stratigraphic implications delivered a temporal frame from the Middle Pleistocene to the Holocene for our sedimentological interpretations and also revealed interruptions in the deposition. The sequence of lithological units indicates a succession of several distinct climate phases: a Middle Pleistocene ice complex indicates cold stage climate. Then, ice wedge growth stopped due to highly increased sedimentation rates and eventually a rise in temperature. Full interglacial climate conditions existed during accumulation of an organic-rich layer – plant macrofossils reflected open forest vegetation existing under dry conditions during Marine Isotope Stage (MIS) 5e. The Late Pleistocene YIC (MIS 4–MIS 2) suggests severe cold-stage climate conditions. No alas deposits, potentially indicating thermokarst processes, were detected at the site. A detailed comparison of the permafrost deposits exposed in the Batagay thaw slump with well-studied permafrost sequences,both coastal and inland, is made to highlight common features and differences in their formation processes and palaeo-climatic histories. Fluvial and lacustrine influence is tem-orarily common in the majority of permafrost exposures, but has to be excluded for the Batagay sequence. We interpret the characteristics of permafrost deposits at this location as a result of various climatically induced processes that are partly seasonally controlled. Nival deposition might have been dominant during winter time, whereas proluvial and aeolian deposition could have prevailed during the snowmelt period and the dry summer season

    Integrating biodiversity: a longitudinal and cross-sectoral analysis of Swiss politics

    The effective conservation and promotion of biodiversity requires its integration into a wide range of sectoral policies. For this to happen, the issue must receive attention across policy sectors. Yet, we know little about how attention to the issue evolves over time and across sectors. Drawing from the literature on environmental policy integration/mainstreaming and policy process theories, we develop competing hypotheses, expecting either increasing or fluctuating attention to the biodiversity issue. We tested the hypotheses using the case of Swiss politics between 1999 and 2018. Applying a combination of computational methods, we analyze the content of a comprehensive collection of policy documents (n ≈ 440,000) attributed to 20 policy sectors. Comparing the sectors, we find that (1) a persistent increase in attention is the exception, (2) if there is an increase in attention, it is likely to be temporary, and (3) the most common pattern is that of invariant attention over time. Biodiversity integration—if it does happen at all—tends to occur in cycles rather than in steady long-term shifts. This implies that the conservation of biodiversity does not follow the cross-sectoral nature of the problem, but is subject to the dynamics of "politics," where actors, because of limited resources, engage with (aspects of) an issue only for a certain amount of time

    Summary:: Since traditional agriculture and forestry are no longer economically viable in many regions of the European Alps, subalpine forests will become less managed or completely abandoned in the near future. Therefore, the interest in understanding how forest stands will develop after abandonment has increased considerably over the past two decades. While much is known about stand structure and stand development of Norway spruce (Picea abies L.) forests, almost no knowledge is available about the same processes in forest communities of the Central Alps. In the Swiss National Park (SNP), the forested area is comprised of mountain pine (Pinus montana Miller), Swiss stone pine/larch, (Pinus cembra L./Larix decidua L.). and mixed stands. When the Park was founded in 1914 all management activities were stopped. Therefore, this area offers the opportunity to study stand development and changes in stand structure after abandonment. We compared historic (1957) and present data (2001/02) from 19 stands that were grouped into characteristic stand types: "mountain pine”, "mixed”, and "stone pine”. We detected significant decreases in total tree density (stem/ha) and sapling density (saplings/ha) of 45 to 57%, and 64 to 76%, respectively, over the 45 years of observation for all stand types. These changes were strongly related to decreases in the number of shade intolerant mountain pine trees. Simultaneously, the amount of non-standing woody residue increased from less than 4 t/ha to 36 to67.7 t/ha, and the density of standing dead wood (stems/ha) decreased significantly between 72 and 94%. The biomass of standing dead wood (t/ha), however, changed only slightly between 1957 and 01/02. Our results describe the successional development of continental subalpine forests after abandonment and outlines changes that might take place in similar areas in the near futur

    Atlantic water inflow to Labrador Sea and its interaction with ice sheet dynamics during the Holocene

    The hydrodynamics of the Labrador Sea, controlled by the complex interplay of oceanographic, atmospheric and ice-sheet processes, play a crucial role for the Atlantic Meridional Overturning Circulation (AMOC). An improved understanding of the hydrodynamics and its forcing in the past could therefore hold a key to understanding its future behaviour. At present, there is a remarkable temporal mismatch, in that the largely microfossil-based reconstructions of Holocene Atlantic-water inflow/influence in the Labrador Sea and Baffin Bay appear to lag grain size-based current strength reconstructions from the adjacent North Atlantic by > 2ka. Here, we present the first current strength record from the West Greenland shelf off Nuuk to reconstruct Atlantic Water (AW)-inflow to the Labrador Sea via the West Greenland Current. Our data show that the Holocene AW-inflow into Labrador Sea is well aligned with the Holocene Speed Maximum documented in the North Atlantic (McCave and Andrews, 2019; Quat. Sci. Rev. 223), suggesting a close coupling with the AMOC. The observed lag between the microfossil-based records and the Holocene Speed Maximum can be explained when considering the presence of an extended meltwater lens that prevented the shoaling of the inflowing Atlantic waters. Once the meltwater discharge waned after the cessation of large-scale melting of the surrounding ice sheets, the AW could influence the surface waters, independently of the strength of its inflow. Only then was an effective ocean-atmosphere heat transfer enabled, triggering the comparably late onset of the regional Holocene Thermal Maximum. Furthermore, sediment geochemical analyses show that short term cooling events, such as the 8.2 ka event related to the final drainage of glacial Lake Agassiz, lead to glacier advances of the Greenland Ice Sheet. Since the grain size data show that these events had no influence on the AW-inflow to the north eastern Labrador Sea, these advances must have been caused by atmospheric cooling. Consequently, we argue that (i) in this region, surface water-based proxies register AW influence rather than inflow (ii) the AW inflow into the Labrador Sea is controlled by the AMOC, but (iii) its impact on an effective ocean-atmosphere heat transfer was hindered by a prevailing meltwater lens in the early Holocene, i.e. until the cessation of large-scale melting of the surrounding ice sheets

    Molecular basis for the bifunctional Uba4-Urm1 sulfur-relay system in tRNA thiolation and ubiquitin-like conjugation.

    The chemical modification of tRNA bases by sulfur is crucial to tune translation and to optimize protein synthesis. In eukaryotes, the ubiquitin-related modifier 1 (Urm1) pathway is responsible for the synthesis of 2-thiolated wobble uridine (U34 ). During the key step of the modification cascade, the E1-like activating enzyme ubiquitin-like protein activator 4 (Uba4) first adenylates and thiocarboxylates the C-terminus of its substrate Urm1. Subsequently, activated thiocarboxylated Urm1 (Urm1-COSH) can serve as a sulfur donor for specific tRNA thiolases or participate in ubiquitin-like conjugation reactions. Structural and mechanistic details of Uba4 and Urm1 have remained elusive but are key to understand the evolutionary branch point between ubiquitin-like proteins (UBL) and sulfur-relay systems. Here, we report the crystal structures of full-length Uba4 and its heterodimeric complex with its substrate Urm1. We show how the two domains of Uba4 orchestrate recognition, binding, and thiocarboxylation of the C-terminus of Urm1. Finally, we uncover how the catalytic domains of Uba4 communicate efficiently during the reaction cycle and identify a mechanism that enables Uba4 to protect itself against self-conjugation with its own product, namely activated Urm1-COSH

    Relating landscape ecological metrics with public survey data on perceived landscape quality and place attachment

    Context It is essential for policy-making and planning that we understand landscapes not only in terms of landscape ecological patterns, but also in terms of their contribution to people's quality of life. Objectives In this study our objective is to test relationships between landscape ecology and social science indicators, by investigating how landscape patterns are linked to people’s perception of landscape quality. Methods To assess public views on landscapes we conducted a survey among 858 respondents in Switzerland. We combined this survey data on perceived landscape quality and place attachment with landscape metrics (e.g. diversity, naturalness of land cover, urban sprawl, fragmentation) in a statistical model to test hypotheses about the relationships between the different variables of interest. Results Our results illustrate the contribution of both landscape composition metrics and social science indicators to understanding variation in people’s perception and assessment of landscape. For example, we found the landscape ecology metrics on urban sprawl and fragmentation to be a negative predictor of overall satisfaction with landscape, and that perceived landscape quality positively predicted place attachment and satisfaction with the municipality landscape. Conclusions This study highlights the importance and feasibility of combining landscape ecology metrics and public survey data on how people perceive, value and relate to landscape in an integrated manner. Our approach has the potential for implementation across a variety of settings and can contribute to holistic and integrated landscape assessments that combine ecological and socio-cultural aspects
