    Teachers valuation and implementation of formative assessment strategies in elementary science classrooms.

    Formative assessment is frequently used by educators, but when asked to define, responses tend to vary widely. In essence, formative assessment is when teachers and students engage in instructional conversations discussing content information and gathering data about present levels of understanding. This feedback exchanged between teachers and students about knowledge gained is a critical element that provides structure and support to move student learning forward. For teachers who effectively use formative assessment strategies, it is the responsive element of adjusting lesson plans, instruction and assessments to give students multiple avenues to develop deeper understanding and to address student learning needs. For students it is the clarification provided by the teacher to link what was known, to what is known to achieve desired learning. In this study elementary teachers in one school district were surveyed by means of a questionnaire to ascertain their perspectives on value and implementation of formative assessment. Four teachers were selected for intensive observations of science lessons utilizing what each perceived to be formative assessment strategies. This study used a mixed methods approach to analyze data from three sources: a questionnaire, classroom observations and teacher interviews. Factor analysis and confirmatory structural equation modeling were used to determine the validity of the questionnaire about teacher\u27s beliefs and use of formative assessment strategies; SEM also provided underlying structural relationships of formative assessment strategies. Teachers\u27 value of formative assessment strategies strongly correlated with their reported use of same strategies. Regression analysis determined no significant correlation between years of experience and use of formative assessment. Interview analysis of case studies indicated that those who had a more student-focused or constructivist approach to science teaching, were more likely to effectively use a wider variety of formative assessment strategies in their instruction; conversely, teacher-focused instructors used fewer formative assessment strategies and did not exemplify those characteristics a constructivist philosophy. A Teacher Profile has been proposed which categorizes teaching behaviors that are more consistent with who embodies the knowledge, beliefs and dispositions about effective formative assessment. This profile has suggested uses for university teacher educators, education administrators, professional development trainers, and professional learning communities

    Tetracycline-Based Measurements of The Bone Dynamics in the Rib of A Girl with Hyperphosphatasia

    Tetracycline-based quantitative histological analysis was made of a rib biopsy taken from a girl afflicted with hyperphosphatasia. The study revealed essentially normal bone dynamics on trabeculae, but greatly increased turnover in the compacta, which was largely composed of woven bone. Since both bone resorption and lamellar bone formation could be unequivocally demonstrated on periosteal, Haversian, and cortical-endosteal surfaces, it is suggested that the direct cause of the cell behavioral abnormality responsible for this disease may arise locally within the bone, but external to the bone cells. Because of its familial nature, however, the ultimate cause probably represents an inborn intracellular defect

    Wood Creek Tidal Marsh Enhancement Project Benthic Macroinvertebrate Monitoring Report 2019

    The focus of this report was to monitor benthic macroinvertebrate communities on the Freshwater Farms Reserve, which underwent two phases of restoration as part of the Wood Creek Tidal Marsh Enhancement Project in 2009-2010 and 2016-2018. Objectives for the restoration activities were to increase winter rearing refugia habitat for several threatened/endangered fish species such as the tidewater goby (Eucyclogobius newberryi), Coho salmon (Oncorhynchus kisutch) and steelhead trout (Oncorhynchus mykiss). The goals of this project were to (1) sample and identify BMIs along a salinity gradient in Wood Creek; (2) assess water quality; and (3) report general trajectory of community composition over time. Results show that the abundance of benthic macroinvertebrates increased dramatically in Wood Creek in 2019 for all sampled sites when compared to previous years of monitoring data. Three taxa accounted for over 99% of the overall composition at each of the sample sites. Increased abundance of benthic macroinvertebrates may provide additional nutritional support for fish present in Wood Creek and Freshwater Creek. Overall, Freshwater Farms Reserve’s post-restoration ecological trajectory seems to be improving in relation to the goals of supporting fish refugia for threatened/endangered species

    Critical Mathematics Pedagogy: Transforming Teachers’ Practices

    This study reports the effects of a graduate-level mathematics education course that focused on critical theory and teaching for social justice on the pedagogical philosophies and practices of three mathematics teachers (middle, high school, and 2-year college). The study employed Freirian participatory research methodology; in fact, the participants were not only co-researchers, but also co-authors of the study. Data collection included reflective essays, journals, and “storytelling”; data analysis was a combination of textual analysis and autoethnography. The findings report that the teachers believed that the course provided not only a new language but also a legitimization to transform their pedagogical philosophies and practices (and research agendas) away from the “traditional” and toward a mathematics for social justice

    Altered gene expression in asymptomatic SHIV-infected rhesus macaques (Macacca mulatta)

    Simian-Human immunodeficiency virus is a chimeric virus which, in rhesus macaques (Macacca mulatta) closely imitates immunodeficiency virus infection in human (HIV). A relatively new way to study pathogenesis of viral infection is to study alterations in host gene expression induced by the virus. SHIV infection with certain strains does not result in clinical signs. We hypothesized that alterations in gene expression relating to the immune system would be present in SHIV-infected animals despite the lack of clinical signs. Splenic tissue from four adult male Indian-origin Rhesus monkeys serologically positive for non-pathogenic SHIV 89.6 was processed by cDNA microarray analysis. Results were compared with the corresponding outcome using splenic tissues from four unexposed adult male Rhesus monkeys. Subsequent gene analysis confirmed statistically significant variations between control and infected samples. Interestingly, SHIV-infected monkeys exhibited altered expression in genes related to apoptosis, signal transduction, T and B lymphocyte activation and importantly, to immune regulation. Although infected animals appeared asymptomatic, our study demonstrated that SHIV-infected monkeys cannot reliably be used in studies of other infectious agents as their baseline gene expression differs from that of normal Rhesus monkeys. The gene expression differences in SHIV-infected animals relative to uninfected animals offer additional clues to the pathogenesis of altered immune function in response to secondary infection

    Perceptions of an Ideal Point-of-Care Test for Sexually Transmitted Infections – A Qualitative Study of Focus Group Discussions with Medical Providers

    A point-of-care test (POCT) for sexually transmitted infections (STIs), which offers immediate diagnosis resulting in patients receiving diagnosis and treatment in a single visit, has the ability to address some of the STI control needs. However, needs assessment from STI experts and end users about currently available STI POCTs and their future new development has not been evaluated since World Health Organization Sexually Transmitted Diseases Diagnostics Initiative was formed over 15 years ago. Therefore, our objective was to explore the perceptions of the ideal types of STI POCT for use in health care settings.A qualitative study, encompassing eight focus groups, was conducted from March 2008 through April 2009. Participants included 6 STD clinic directors, 63 clinicians, and 7 public health/laboratory/epidemiology professionals in the STI field. Discussion topics included currently available POCT, perceived barriers to using POCT in clinics, priority STI for the development of new POCT, and characteristics of the ideal POCT. All discussions were recorded and transcribed verbatim. Themes raised as barriers for current POCT included complexity, long time frames of the so-called "rapid" test, multiple time-driven steps, requiring laboratory technician, difficulty in reading result, interruption of workflow, unreliability, and invasiveness. Chlamydia trachomatis was identified as the priority organism for development of a new STI POCT. Themes indicated for the ideal POCT included rapid turnaround (up to 20 minutes), ease of use, non-invasive, accurate (preferred sensitivity and specificity in the range of high 90s), Clinical Laboratory Improvement Amendments (CLIA)-waived, user-friendly (for both patients and staff), compact, durable, and sturdy.Focus group discussions with STI experts and professionals highlighted chlamydia as the top priority pathogen for POCT development, and identified the qualities of new POCT for STIs. Participants endorsed ease of use, rapid turnaround and high accuracy as essential characteristics of an ideal POCT

    Src Binds Cortactin Through An Sh2 Domain Cystine-Mediated Linkage

    Tyrosine-kinase-based signal transduction mediated by modular protein domains is critical for cellular function. The Src homology (SH)2 domain is an important conductor of intracellular signaling that binds to phosphorylated tyrosines on acceptor proteins, producing molecular complexes responsible for signal relay. Cortactin is a cytoskeletal protein and tyrosine kinase substrate that regulates actin-based motility through interactions with SH2-domain-containing proteins. The Src kinase SH2 domain mediates cortactin binding and tyrosine phosphorylation, but how Src interacts with cortactin is unknown. Here we demonstrate that Src binds cortactin through cystine bonding between Src C185 in the SH2 domain within the phosphotyrosine binding pocket and cortactin C112/246 in the cortactin repeats domain, independent of tyrosine phosphorylation. Interaction studies show that the presence of reducing agents ablates Src-cortactin binding, eliminates cortactin phosphorylation by Src, and prevents Src SH2 domain binding to cortactin. Tandem MS/MS sequencing demonstrates cystine bond formation between Src C185 and cortactin C112/246. Mutational studies indicate that an intact cystine binding interface is required for Src-mediated cortactin phosphorylation, cell migration, and pre-invadopodia formation. Our results identify a novel phosphotyrosine-independent binding mode between the Src SH2 domain and cortactin. Besides Src, one quarter of all SH2 domains contain cysteines at or near the analogous Src C185 position. This provides a potential alternative mechanism to tyrosine phosphorylation for cysteine-containing SH2 domains to bind cognate ligands that may be widespread in propagating signals regulating diverse cellular functions

    Clinical and Experimental Applications of NIR-LED Photobiomodulation

    This review presents current research on the use of far-red to near-infrared (NIR) light treatment in various in vitro and in vivo models. Low-intensity light therapy, commonly referred to as “photobiomodulation,” uses light in the far-red to near-infrared region of the spectrum (630–1000 nm) and modulates numerous cellular functions. Positive effects of NIR–light-emitting diode (LED) light treatment include acceleration of wound healing, improved recovery from ischemic injury of the heart, and attenuated degeneration of injured optic nerves by improving mitochondrial energy metabolism and production. Various in vitro and in vivo models of mitochondrial dysfunction were treated with a variety of wavelengths of NIR-LED light. These studies were performed to determine the effect of NIR-LED light treatment on physiologic and pathologic processes. NIRLED light treatment stimulates the photoacceptor cytochrome c oxidase, resulting in increased energy metabolism and production. NIR-LED light treatment accelerates wound healing in ischemic rat and murine diabetic wound healing models, attenuates the retinotoxic effects of methanol-derived formic acid in rat models, and attenuates the developmental toxicity of dioxin in chicken embryos. Furthermore, NIR-LED light treatment prevents the development of oral mucositis in pediatric bone marrow transplant patients. The experimental results demonstrate that NIR-LED light treatment stimulates mitochondrial oxidative metabolism in vitro, and accelerates cell and tissue repair in vivo. NIR-LED light represents a novel, noninvasive, therapeutic intervention for the treatment of numerous diseases linked to mitochondrial dysfunction
