    Rapid Identification of Bio-Molecules Applied for Detection of Biosecurity Agents Using Rolling Circle Amplification

    Detection and identification of pathogens in environmental samples for biosecurity applications are challenging due to the strict requirements on specificity, sensitivity and time. We have developed a concept for quick, specific and sensitive pathogen identification in environmental samples. Target identification is realized by padlock- and proximity probing, and reacted probes are amplified by RCA (rolling-circle amplification). The individual RCA products are labeled by fluorescence and enumerated by an instrument, developed for sensitive and rapid digital analysis.     Feedback Inhibition in the PhoQ/PhoP Signaling System by a Membrane Peptide

    The PhoQ/PhoP signaling system responds to low magnesium and the presence of certain cationic antimicrobial peptides. It regulates genes important for growth under these conditions, as well as additional genes important for virulence in many gram-negative pathogens. PhoQ is a sensor kinase that phosphorylates and activates the transcription factor PhoP. Since feedback inhibition is a common theme in stress-response circuits, we hypothesized that some members of the PhoP regulon may play such a role in the PhoQ/PhoP pathway. We therefore screened for PhoP-regulated genes that mediate feedback in this system. We found that deletion of mgrB (yobG), which encodes a 47 amino acid peptide, results in a potent increase in PhoP-regulated transcription. In addition, over-expression of mgrB decreased transcription at both high and low concentrations of magnesium. Localization and bacterial two-hybrid studies suggest that MgrB resides in the inner-membrane and interacts directly with PhoQ. We further show that MgrB homologs from Salmonella typhimurium and Yersinia pestis also repress PhoP-regulated transcription in these organisms. In cell regulatory circuits, feedback has been associated with modulating the induction kinetics and/or the cell-to-cell variability in response to stimulus. Interestingly, we found that elimination of MgrB-mediated feedback did not have a significant effect on the kinetics of reporter protein production and did not decrease the variability in expression among cells. Our results indicate MgrB is a broadly conserved membrane peptide that is a critical mediator of negative feedback in the PhoQ/PhoP circuit. This new regulator may function as a point of control that integrates additional input signals to modulate the activity of this important signaling system

    International audienceQue le temps du sujet ne soit pas superposable au temps médical, le temps de la rémission le dit sans doute suffisamment. Quels peuvent donc bien être les enjeux d'une " consultation rémission " telle qu'ouverte récemment au Centre de Lutte Contre le Cancer de Reims, organisée sur la base d'un binôme médecin généraliste travaillant en cancérologie/psychologue-psychanalyste ? De la rupture de l'illusion d'identité ouverte par le choc du diagnostic à la chute, au temps de la rémission, de l'identité substitutive de surface produite par " l'effet cadre " des traitements de longue durée, des points de repère se dessinent pour un renouvellement de la clinique en cancérologie : problématique identitaire, effet prothétique de la structure de soins, perte de l'étayage, élaboration du deuil du maternel... Car, nous savons à peu près ce que c'est qu'être malade ou non, mais que savons-nous de celui qui " a eu un cancer " ? L'extension des Soins de Support à la phase de rémission de la maladie s'impose sans doute..

    Cloning and characterization of cspL and cspP, two cold-inducible genes from Lactobacillus plantarum.

    Two cold shock genes, cspL and cspP, have been cloned from two Lactobacillus plantarum strains. These genes, which are nonallelic, were present in all strains tested. The genes encode 66-amino-acid polypeptides related to each other and to the cold shock Csp family. Transcription of cspP rendered a single mRNA, while two cspL mRNAs were found with common 5' ends. The amounts of these transcripts increased moderately upon exposure of the cultures to cold
