    Dynamin- and Rab5-Dependent Endocytosis of a Ca<sup>2+</sup>-Activated K<sup>+</sup> Channel, KCa2.3

    Regulation of the number of ion channels at the plasma membrane is a critical component of the physiological response. We recently demonstrated that the Ca2+-activated K+ channel, KCa2.3 is rapidly endocytosed and enters a Rab35- and EPI64C-dependent recycling compartment. Herein, we addressed the early endocytic steps of KCa2.3 using a combination of fluorescence and biotinylation techniques. We demonstrate that KCa2.3 is localized to caveolin-rich domains of the plasma membrane using fluorescence co-localization, transmission electron microscopy and co-immunoprecipitation (co-IP). Further, in cells lacking caveolin-1, we observed an accumulation of KCa2.3 at the plasma membrane as well as a decreased rate of endocytosis, as assessed by biotinylation. We also demonstrate that KCa2.3 and dynamin II are co-localized following endocytosis as well as demonstrating they are associated by co-IP. Further, expression of K44A dynamin II resulted in a 2-fold increase in plasma membrane KCa2.3 as well as a 3-fold inhibition of endocytosis. Finally, we evaluated the role of Rab5 in the endocytosis of KCa2.3. We demonstrate that expression of a dominant active Rab5 (Q79L) results in the accumulation of newly endocytosed KCa2.3 on to the membrane of the Rab5-induced vacuoles. We confirmed this co-localization by co-IP; demonstrating that KCa2.3 and Rab5 are associated. As expected, if Rab5 is required for the endocytosis of KCa2.3, expression of a dominant negative Rab5 (S34N) resulted in an approximate 2-fold accumulation of KCa2.3 at the plasma membrane. This was confirmed by siRNA-mediated knockdown of Rab5. Expression of the dominant negative Rab5 also resulted in a decreased rate of KCa2.3 endocytosis. These results demonstrate that KCa2.3 is localized to a caveolin-rich domain within the plasma membrane and is endocytosed in a dynamin- and Rab5-dependent manner prior to entering the Rab35/EPI64C recycling compartment and returning to the plasma membrane. © 2012 Gao et al

    Laser-plasma interactions with a Fourier-Bessel Particle-in-Cell method

    A new spectral particle-in-cell (PIC) method for plasma modeling is presented and discussed. In the proposed scheme, the Fourier-Bessel transform is used to translate the Maxwell equations to the quasi-cylindrical spectral domain. In this domain, the equations are solved analytically in time, and the spatial derivatives are approximated with high accuracy. In contrast to the finite-difference time domain (FDTD) methods that are commonly used in PIC, the developed method does not produce numerical dispersion, and does not involve grid staggering for the electric and magnetic fields. These features are especially valuable in modeling the wakefield acceleration of particles in plasmas. The proposed algorithm is implemented in the code PLARES-PIC, and the test simulations of laser plasma interactions are compared to the ones done with the quasi-cylindrical FDTD PIC code CALDER-CIRC.Comment: submitted to Phys.     Kepuasan Pasien Pengguna Kartu Jakarta Sehat di Unit Rawat Jalan Rumah Sakit

    Peluncuran Kartu Jakarta Sehat (KJS) pada bulan November 2012 mengakibatkan jumlah kunjungan pasien yang melonjak drastis di fasilitas kesehatan, khususnya rumah sakit, baik itu rumah sakit pemerintah pusat, rumah sakit pemerintahdaerah maupun rumah sakit swasta yang berkomitmen dengan Pemerintah Daerah Jakarta. KJS memberikan kemudahan akses layanan kesehatan agar pasien dapat berobat secara gratis. Namun persoalan yang muncul kemudian adalah pasien harus mengantre lama di loket pendaftaran, ruang pemeriksaan, maupun ruang pengambilan obat. Penelitian ini bertujuan untuk mengetahui hubungan faktor sosial budaya, pribadi, dan psikologis dengan kepuasan pasien pengguna KJS pada 110 responden di Unit Rawat Jalan RSU UKI, dan telah dilaksanakan pada bulan Maret 2013. Rancangan penelitian yang digunakan adalah deskriptif kuantitatif dengan pendekatan potong lintang. Hasil penelitian menunjukkan kepuasan pasien KJS sebesar 40,9%. Akses ke rumah sakit, motivasi, informasi baik dan informasi tidak baik berhubungan signifikan dengan kepuasan pasien KJS, tetapi motivasi adalah hal yang paling dominan berhubungan. Selain itu, kepuasan pasien KJS juga berhubungan bermakna dengan pemanfaatan kembali layanan dan minat merekomendasikan layanan pada orang lain.Jakarta Health Card (JHC) was launched on November 2012, followed by sudden increase number of patient in any hospital accordingly. Jakarta Health Card ease the patient to afford health care without payment. We conducted study on the relation among socio-culture, demographic characteristics, and psychology factors toward patient satisfaction in using JHC. The study subjects were 110 outpatient of UKI Hospital, with study design descriptive quantitative with cross sectional approach. The result showed 40,9% of the patients were satisfied with the hospital service. Factors such as hospital access, motivation, good information and bad information were associated with patient satisfaction significantly, and motivation was the dominant factor. Besides, patient satisfaction also has significant correlation with revisiting/ reusing and recommendation of the hospital to other patients

    PT. PLN P2B JB Depok merupakan sebuah Perusahaan yang bergerak dalam bidang jasa penyaluran dan pusat pengaturan beban listrik pada wilayah Jawa-Bali. Setiap divisi memiliki target dan harus selesai tepat waktu. Beban pekerjaan yang berat memaksa pegawai untuk bekerja lebih keras sehingga berakibat banyak pegawai yang sakit. Untuk mengurangi jumlah pegawai yang sakit PLN P2B JB Depok melakukan tindakan pencegahan setiap enam bulan sekali guna mencegah terjadinya penyakit yang sama pada semester berikutnya, tetapi jumlah penyakit masih tetap bahkan bertambah, karena itu dibutuhkan cara baru untuk merencanakan program kesehatan pegawai di semester berikutnya. Bu Fatma ingin menggabungkan database kesehatan pegawai dengan kualitas kerja pegawai agar mendapatkan penyakit dengan kelompok kualitas kerja yang berbeda-beda. Algoritma K-Means++ adalah algoritma yang mampu mengelompokkan data yang besar menjadi beberapa kelompok, tetapi K-Means++ tidak mampu mengelompokkan pegawai yang sama dalam satu kelompok kualitas kerja maka dari itu penggunaan COP-Kmeans dibutuhkan. Hasil dari perhitungan K-Means++ dan COP-Kmeans pada penelitian ini adalah 5 kelompok kualitas kerja dengan pegawai yang berbeda dan penyakit yang berbeda, dimana penyakit dan jumlah penyakit dalam kelompok tersebut akan digunakan sebagai bahan pertimbangan Bu Fatma untuk merencanakan program kesehatan pegawai di semester berikutnya

    Investigating the functionality of an OCT4-short response element in human induced pluripotent stem cells.

    Pluripotent stem cells offer great therapeutic promise for personalized treatment platforms for numerous injuries, disorders, and diseases. Octamer-binding transcription factor 4 (OCT4) is a key regulatory gene maintaining pluripotency and self-renewal of mammalian cells. With site-specific integration for gene correction in cellular therapeutics, use of the OCT4 promoter may have advantages when expressing a suicide gene if pluripotency remains. However, the human OCT4 promoter region is 4 kb in size, limiting the capacity of therapeutic genes and other regulatory components for viral vectors, and decreasing the efficiency of homologous recombination. The purpose of this investigation was to characterize the functionality of a novel 967bp OCT4-short response element during pluripotency and to examine the OCT4 titer-dependent response during differentiation to human derivatives not expressing OCT4. Our findings demonstrate that the OCT4-short response element is active in pluripotency and this activity is in high correlation with transgene expression in vitro, and the OCT4-short response element is inactivated when pluripotent cells differentiate. These studies demonstrate that this shortened OCT4 regulatory element is functional and may be useful as part of an optimized safety component in a site-specific gene transferring system that could be used as an efficient and clinically applicable safety platform for gene transfer in cellular therapeutics

    Bioliquids and their use in power generation - a technology review

    The first EU Renewable Energy Directive (RED) served as an effective push for world-wide research efforts on biofuels and bioliquids, i.e. liquid fuels for energy purposes other than for transport, including electricity, heating, and cooling, which are produced from biomass. In December 2018 the new RED II was published in the Official Journal of the European Union. Therefore, it is now the right time to provide a comprehensive overview of achievements and practices that were developed within the current perspective. To comply with this objective, the present study focuses on a comprehensive and systematic technical evaluation of all key aspects of the different distributed energy generation pathways using bioliquids in reciprocating engines and micro gas turbines that were overseen by these EU actions. Methodologically, the study originates from the analyses of feedstock and fuel processing technologies, which decisively influence fuel properties. The study systematically and holistically highlights the utilisation of these bioliquids in terms of fuel property specific challenges, required engine adaptations, and equipment durability, culminating in analyses of engine performance and emissions. In addition, innovative proposals and future opportunities for further technical improvements in the whole production-consumption cycle are presented, thus serving as a guideline for upcoming research and development activities in the fast-growing area of bioliquids. Additionally, the paper systematically addresses opportunities for the utilisation of waste streams, emerging from the ever increasing circular use of materials and resources. With this, the present review provides the sorely needed link between past efforts, oriented towards the exploitation of bio-based resources for power generation, and the very recent zero-waste oriented society that will require a realistic exploitation plan for residuals originating from intensive material looping

    ‘It’s better than daytime television’: questioning the socio-spatial impacts of massage parlours on residential communities

    It has been shown that street sex work is problematic for some communities, but there is less evidence of the effects of brothels. Emerging research also suggests that impact discourses outlined by residential communities and in regulatory policies should be critiqued, because they are often based on minority community voices, and limited tangible evidence is used to masquerade wider moral viewpoints about the place of sex work. Using a study of residents living in close proximity to brothels in Blackpool, this paper argues that impact is socially and spatially fluid. Impact needs to be evaluated in a more nuanced manner, which is considerate of the heterogeneity of (even one type of) sex work, and the community in question. Brothels in Blackpool had a variety of roles in the everyday socio-spatial fabric; thus also questioning the common assumption that sex work only impacts negatively on residential communities

    Dysfunction of Mitral Ball Valve Prosthesis

    Three case histories of patients with malfunctions of Smeloff-Cutter mitral ball valve prostheses are presented here - one underwent successful replacement. The clinical diagnosis was made by observing a marked variation in the A2OC interval and intermittent absence of the opening click. Phonocardiograms were diagnostic in all three cases. while echocardiograms and even angiograms did not uniformly diagnose prosthetic valve dysfunction. Periodic phonocardiographic evaluations may be helpful in early detection of the prosthetic valve dysfunction. Once the diagnosis is established, immediate surgical treatment is imperative to prevent sudden death
