    Effects of loperamide on the human hypothalamo-pituitary-adrenal axis in vivo and in vitro.

    Loperamide, an opiate agonist of high specificity for p-receptors, was recently reported to suppress ACTH and cortisol levels in normal subjects, but not in patients with proven ACTH-dependent Cushing’s disease. However, there is little information on the site of action of loperamide in the hypothalamo-pituitary-adrenal axis of man. We investigated the effect of loperamide on pituitary hormone secretion in uiuo and in vitro. In seven normal subjects, basal ACTH plasma levels were significantly suppressed 3 h after loperamide administration (16 mg, orally) from 5 + 1 to 2 f 0 pmol/L (P < 0.0001). After the combined pituitary stimulation test (100 pg human CRH, 100 rg GnRH, 100 pg GH-releasing hormone, and 200 pg TRH), the ACTH peak (maximum increase at 30 min) was significantly blunted by loperamide from 9 + 1 to 4 of: 1 pmol/L (P < 0.001) and the area under the curve of ACTH from O-120 min was reduced from 35 + 5 to 23 + 4 pmol/L.2 h (P < 0.05). In the insulin-hypoglycemia test (0.15 IU/kg BW), neither the ACTH peak nor the area under the curve of ACTH was affected by loperamide. In six patients with Cushing’s disease and one patient with secondary adrenal insufficency due to hypothalamic failure, neither basal ACTH and cortisol levels nor CRH-stimulated levels were influenced by loperamide. In four cultured human corticotropic adenomas, loperamide was not able to reduce basal and CRH-induced ACTH secretion. In summary, loperamide is able to reduce basal and CRHinduced ACTH and cortisol levels in normal subjects, but not in patients with Cushing’s disease or secondary adrenal failure of hypothalamic origin. Loperamide has no significant effect on insulin-hypoglycemia- induced ACTH and cortisol levels and, therefore, no effect on stress-induced elevation of cortisol levels. Loperamide might act at a suprapituitary site in man in viuo, but, nevertheless, a pituitary site cannot be excluded

    Time-resolved X-ray microscopy of nanoparticle aggregates under oscillatory shear

    Of all current detection techniques with nanometer resolution, only X-ray microscopy allows imaging nanoparticles in suspension. Can it also be used to investigate structural dynamics? When studying response to mechanical stimuli, the challenge lies in applying them with precision comparable to spatial resolution. In the first shear experiments performed in an X-ray microscope, we accomplished this by inserting a piezo actuator driven shear cell into the focal plane of a scanning transmission X-ray microscope (STXM). Thus shear-induced reorganization of magnetite nanoparticle aggregates could be demonstrated in suspension. As X-ray microscopy proves suitable for studying structural change, new prospects open up in physics at small length scales.Comment: submitted to J. Synchrot. Radia

    Remote brainstorming: Methodological interventions in designing from a distance

    This paper examines the immediate effects of group methods in facilitating remote team collaboration. We recruited seven teams with prior experience working together. All teams completed two current, complex, and open-ended design challenges using remote tools. We examined design activities before and after teams were given a design method intervention. The interventions were a Brainstorming Method to promote divergent thinking and the Five-Whys method to promote analytical thinking. Using OpenFace, we observed changes in emotion by examining facial expressions. We found that the brainstorming intervention did not have a change in ideation performance and the problem analysis intervention had a decrease in ideation performance. Teams used digital media to facilitate communication but were constrained by the media\u27s tools. Our results can inform teams in organizations interested in promoting divergent thinking to not expect immediate improvements in ideation performance following the introduction of a design method. Future research is required to identify relevant abilities and social skills needed to facilitate remote ideation through design methods

    Reversible magnetomechanical collapse: virtual touching and detachment of rigid inclusions in a soft elastic matrix

    Soft elastic composite materials containing particulate rigid inclusions in a soft elastic matrix are candidates for developing soft actuators or tunable damping devices. The possibility to reversibly drive the rigid inclusions within such a composite together to a close-to-touching state by an external stimulus would offer important benefits. Then, a significant tuning of the mechanical properties could be achieved due to the resulting mechanical hardening. For a long time, it has been argued whether a virtual touching of the embedded magnetic particles with subsequent detachment can actually be observed in real materials, and if so, whether the process is reversible. Here, we present experimental results that demonstrate this phenomenon in reality. Our system consists of two paramagnetic nickel particles embedded at finite initial distance in a soft elastic polymeric gel matrix. Magnetization in an external magnetic field tunes the magnetic attraction between the particles and drives the process. We quantify the scenario by different theoretical tools, i.e., explicit analytical calculations in the framework of linear elasticity theory, a projection onto simplified dipole-spring models, as well as detailed finite-element simulations. From these different approaches, we conclude that in our case the cycle of virtual touching and detachment shows hysteretic behavior due to the mutual magnetization between the paramagnetic particles. Our results are important for the design and construction of reversibly tunable mechanical damping devices. Moreover, our projection on dipole-spring models allows the formal connection of our description to various related systems, e.g., magnetosome filaments in magnetotactic bacteria.Comment: 14 pages, 7 figure

    The role of the novel Th17 cytokine IL-26 in intestinal inflammation

    Background and aims: Interleukin 26 (IL-26), a novel IL-10-like cytokine without a murine homologue, is expressed in T helper 1 (Th1) and Th17 cells. Currently, its function in human disease is completely unknown. The aim of this study was to analyse its role in intestinal inflammation.Methods: Expression studies were performed by reverse transcription-PCR (RT-PCR), quantitative PCR, western blot and immunohistochemistry. Signal transduction was analysed by western blot experiments and ELISA. Cell proliferation was measured by MTS (3-(4,5-dimethylthiazol-2-yl)-5-(carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) assay. IL-26 serum levels were determined by an immunoluminometric assay (ILMA).Results: All examined intestinal epithelial cell (IEC) lines express both IL-26 receptor subunits IL-20R1 and IL-10R2. IL-26 activates extracellular signal-related kinase (ERK)-1/2 and stress-activated protein kinase/c-Jun N-terminal kinase (SAPK/JNK) mitogen-activated protein (MAP) kinases, Akt and signal transducers and activators of transcription (STAT) 1/3. IL-26 stimulation increases the mRNA expression of proinflammatory cytokines but decreases cell proliferation. In inflamed colonic lesions of patients with Crohn's disease, an elevated IL-26 mRNA expression was found that correlated highly with the IL-8 and IL-22 expression. Immunohistochemical analysis demonstrated IL-26 protein expression in colonic T cells including Th17 cells expressing the orphan nuclear receptor ROR\textgreekgt, with an increased number of colonic IL-26-expressing cells in active Crohn's disease.Conclusion: Intestinal cells express the functional IL-26 receptor complex. IL-26 modulates IEC proliferation and proinflammatory gene expression and its expression is upregulated in active Crohn's disease, indicating a role for this cytokine system in the innate host cell response during intestinal inflammation. For the first time, IL-26 expression is demonstrated in colonic ROR\textgreekgt-expressing Th17 cells in situ, supporting a role for this cell type in the pathogenesis of Crohn's disease

    Autopoietic organisation of knowledge, creativity and innovation: a case study of the automotive manufacturer Daimler AG

    This doctoral thesis presents an analysis of the context in which creativity and innovation emerges. It is based on an investigation of a single case organisation.The theoretical framework for the study derived from a number of fields, including organisational studies, social science and systems theory. Of particular importance were studies of organisational systems, their function, and the dynamics that allow creativity and innovation to emerge. Autopoietic system theory is relevant here.The empirical work was conducted in a single large case study organisation based in Germany. Daimler AG, served as the site of data collection. The data for analysis was generated from a survey, focus groups and interviews.The study identified large organisational structures such as organisational design, culture and information and knowledge, which constrain or enable the fluid process of routines, individuals' interactions, and knowledge and idea creation. These structures are highly interrelated and correlate with the organisational innovation performance. These structures turn into fluid patterns of individual and group creativity. Nine patterns were identified, which build a pattern language of creativity in organisations. This pattern language consists of three main "pattern rules".The contribution of the study is the identification of three main factors or "pattern rules" that underpin creativity and innovation. These are (1) diverse experienced experts within the "thick of the action"; (2) innovation willingness to create and support change; (3) "free space" where employees can explore, create and prototype new ideas. In this thesis this concept is labelled "Freiraum", which is the German word for "free space" in which individuals and teams can achieve their potential.These rules build a model of two spaces, which facilitate a spiral of creativity driven by the innovation willingness. This model provides explanation of how creativity and innovation emerges within the context of an organisation. This work draws particular attention to the dynamics of creativity and innovation, and the influence of organisational control on redundancy of the system, where high control leads to low redundancy and vice versa
