    Mammalian oocytes are targets for prostaglandin E2 (PGE2) action

    <p>Abstract</p> <p>Background</p> <p>The ovulatory gonadotropin surge increases synthesis of prostaglandin E2 (PGE2) by the periovulatory follicle. PGE2 actions on granulosa cells are essential for successful ovulation. The aim of the present study is to determine if PGE2 also acts directly at the oocyte to regulate periovulatory events.</p> <p>Methods</p> <p>Oocytes were obtained from monkeys and mice after ovarian follicular stimulation and assessed for PGE2 receptor mRNA and proteins. Oocytes were cultured with vehicle or PGE2 and assessed for cAMP generation, resumption of meiosis, and in vitro fertilization.</p> <p>Results</p> <p>Germinal vesicle intact (GV) oocytes from both monkeys and mice expressed mRNA for the PGE2 receptors EP2, EP3, and EP4. EP2 and EP4 proteins were detected by confocal microscopy in oocytes of both species. Monkey and mouse oocytes responded to PGE2 as well as agonists selective for EP2 and EP4 receptors with elevated cAMP, consistent with previous identification of EP2 and EP4 as GĪ±s/adenylyl cyclase coupled receptors. Incubation of mouse GV stage oocytes with PGE2 delayed oocyte nuclear maturation in vitro, but PGE2 treatment did not alter the percentage of mouse oocytes that fertilized successfully. PGE2 treatment also decreased the percentage of monkey oocytes that resumed meiosis in vitro. In contrast with mouse oocytes, the percentage of monkey oocytes which fertilized in vitro was lower after treatment with PGE2. Monkey oocytes with intact cumulus showed delayed nuclear maturation, but fertilization rate was not affected by PGE2 treatment.</p> <p>Conclusions</p> <p>Monkey and mouse oocytes express functional PGE2 receptors. PGE2 acts directly at mammalian oocytes to delay nuclear maturation. Surrounding cumulus cells modulate the effect of PGE2 to alter subsequent fertilization.</p

    Correlation between the optical veiling and accretion properties: A case study of the classical T Tauri star DK Tau

    Classical T Tauri stars (cTTs) accrete from their circumstellar disk. The material falls onto the stellar surface, producing an accretion shock, which generates veiling in a star's spectra. In addition, the shock causes a localized accretion spot at the level of the chromosphere. Our goal is to investigate the accretion, particularly the mass accretion rates (Macc), for the cTTs DK Tau, over two periods of 17 and 29 days, using two different procedures for comparison purposes. The first method relies on the derivation of the accretion luminosity via accretion-powered emission lines. The second compares the variability of the optical veiling with accretion shock models to determine mass accretion rates. We used observations taken in 2010 and 2012 with the ESPaDOnS spectropolarimeter at the CFHT. We find peak values of the veiling (at 550 nm) ranging from 0.2 to 1.3, with a steeper trend across the wavelength range for higher peak values. When using the accretion-powered emission lines, we find mass accretion rate values ranging from log(Macc[Msol/yr]) = -8.20 to log(Macc[Msol/yr]) = -7.40. This agrees with the values found in the literature, as well as the values calculated using the accretion shock models and the veiling. In addition, we identify a power-law correlation between the values of the accretion luminosity and the optical veiling. For the 2010 observations, using the values of the filling factors (which represent the area of the star covered by an accretion spot) derived from the shock models, we infer that the accretion spot was located between +45 degrees and +75 degrees in latitude. We show that both methods of determining the mass accretion rate yield similar results.     Proliferated LEO Architecture Enabling Beyond Line of Sight Fires (pLEO BLOS Fires)

    NPS NRP Project PosterChief of Naval Operations, ADM Gilday, and Commandant of the Marine Corps, Gen Berger, directed modernization efforts across the Naval Services. The concept of Naval Operational Architecture (NOA) enables the development of additional beyond-line-of-sight (BLOS) targeting and fires delivered from widely distributed points within and outside a near-peer's weapons engagement zones (WEZ). Research into proliferated low-earth orbiting communications satellites (pLEO) reveal an intersection with the Joint All Domain Command and Control (JADC2) priority and the Navy and Marine components leveraging efforts such as Task Force Overmatch (DEVSECOPS). There is an immediate need to develop requirements and analyze alternate architectures for delivering BLOS precision fires in future contested environments in the context of Great Power Competition. Faculty experience with space mission architecture design will be leveraged to advise students in achieving the following objectives: 1) Research and develop operational, functional objective and threshold requirements for a proliferated LEO systems within the JADC2 framework to enable BLOS precision fires. 2) Analyze alternative architectures for the requirements which enable BLOS precision fires in a contested environment. A phased approach will be used to meet the research objectives. First, a capabilities or system requirements-like document will be generated to include validation criteria and in-phase lessons learned. Second, standard aerospace modeling/simulation techniques will be used to define the mission and architecture requirements and analyze the alternatives for architectures enabling BLOS precision fires through a contested environment to include in-phase lessons learned.     Aquatic Eddy Correlation: Quantifying the Artificial Flux Caused by Stirring-Sensitive O2 Sensors

    In the last decade, the aquatic eddy correlation (EC) technique has proven to be a powerful approach for non-invasive measurements of oxygen fluxes across the sediment water interface. Fundamental to the EC approach is the correlation of turbulent velocity and oxygen concentration fluctuations measured with high frequencies in the same sampling volume. Oxygen concentrations are commonly measured with fast responding electrochemical microsensors. However, due to their own oxygen consumption, electrochemical microsensors are sensitive to changes of the diffusive boundary layer surrounding the probe and thus to changes in the ambient flow velocity. The so-called stirring sensitivity of microsensors constitutes an inherent correlation of flow velocity and oxygen sensing and thus an artificial flux which can confound the benthic flux determination. To assess the artificial flux we measured the correlation between the turbulent flow velocity and the signal of oxygen microsensors in a sealed annular flume without any oxygen sinks and sources. Experiments revealed significant correlations, even for sensors designed to have low stirring sensitivities of ~0.7%. The artificial fluxes depended on ambient flow conditions and, counter intuitively, increased at higher velocities because of the nonlinear contribution of turbulent velocity fluctuations. The measured artificial fluxes ranged from 2-70 mmol m(-2) d(-1) for weak and very strong turbulent flow, respectively. Further, the stirring sensitivity depended on the sensor orientation towards the flow. For a sensor orientation typically used in field studies, the artificial flux could be predicted using a simplified mathematical model. Optical microsensors (optodes) that should not exhibit a stirring sensitivity were tested in parallel and did not show any significant correlation between O2 signals and turbulent flow. In conclusion, EC data obtained with electrochemical sensors can be affected by artificial flux and we recommend using optical microsensors in future EC-studies

    Psychometric Characteristics of the Connor-Davidson Resilience Scale (CD-RISC) in Postpartum Mothers with Histories of Childhood Maltreatment

    Background: There is increased awareness that resilience serves as a protective factor against adverse psychophysiological sequelae in the context of stress. However, there are few instruments to assess this construct in adult populations. The Connor-Davidson resilience scale (CD-RISC) has been developed to assess adaptation following stress exposure. While this instrument has previously demonstrated impressive reliability and construct validity, prior research has not supported the consistency of the originally described factor structure. There is also limited evidence regarding the measurement of resilience in the context of cumulative stress exposure. Objectives: This research explores the psychometric properties of the CD-RISC in mothers with childhood histories of maltreatment Materials and Methods: Postpartum women who endorsed a history of childhood abuse or neglect (N = 141) completed the CD-RISC, the childhood trauma questionnaire and other surveys measuring positive and negative health and functioning. We calculated descriptive statistics with percentage counts and means as appropriate. Internal reliability was evaluated by Cronbachā€™s alpha and the calculation of item-to-total score correlations. Parallel analysis (PA) was utilized to derive the number of retained factors. Results: A recent parenting transition concomitant with a history of maltreatment was associated with lower CD-RISC scores. Internal reliability and concurrent validity analyses were satisfactory and consistent with predicted hypotheses. Exploratory factor analysis (EFA) supported a four-factor model of resilience with this population. Conclusions: This research offers further evidence of the reliability and validity of the CD-RISC. Further, the results of the EFA with parallel analysis offer an empirically-driven derivation of factors for this population

    White privilege and teacher perceptions of teacher-child relationship quality

    In this study, we investigated differences in teachersā€™ perceptions of the teacher-child relationship from kindergarten through second grade as a function of child race and gender from the perspective of critical race theory and the cultural synchrony hypothesis. Given the extensive evidence of White privilege and anti-Black racism in the US education system, we expected that teachers, particularly White teachers, would perceive their relationships with White children more positively than with Black children. Controlling for family SES and child gender, results supported this hypothesis. Black boys had the highest risk of being perceived by teachers as having poor relationships with teachers in kindergarten (highest conflict and lowest closeness) and White girls had the lowest risk. In addition, teachers perceived relationships with Black boys as increasing in conflict across first and second grades at higher rates than with White and female children. These findings remained after examining teacher-child racial match as a moderator. Our results indicate that racism and sexism work together to explain the perceptions teachers have of children in the early elementary grades. Implications for training teachers and school psychologists on anti-racism and cultural competency are discussed

    Quantitative changes in intracellular calcium and extracellular-regulated kinase activation measured in parallel in CHO cells stably expressing serotonin (5-HT) 5-HT2A or 5-HT2C receptors

    <p>Abstract</p> <p>Background</p> <p>The serotonin (5-HT) 2A and 2C receptors (5-HT<sub>2A</sub>R and 5-HT<sub>2C</sub>R) are involved in a wide range of physiological and behavioral processes in the mammalian central and peripheral nervous systems. These receptors share a high degree of homology, have overlapping pharmacological profiles, and utilize many of the same and richly diverse second messenger signaling systems. We have developed quantitative assays for cells stably expressing these two receptors involving minimal cell sample manipulations that dramatically improve parallel assessments of two signaling responses: intracellular calcium (<it>Ca<sub>i</sub></it><sup>++</sup>) changes and activation (phosphorylation) of downstream kinases. Such profiles are needed to begin to understand the simultaneous contributions from the multiplicity of signaling cascades likely to be initiated by serotonergic ligands.</p> <p>Results</p> <p>We optimized the <it>Ca<sub>i</sub></it><sup>++ </sup>assay for stable cell lines expressing either 5-HT<sub>2A</sub>R or 5-HT<sub>2C</sub>R (including dye use and measurement parameters; cell density and serum requirements). We adapted a quantitative 96-well plate immunoassay for pERK in the same cell lines. Similar cell density optima and time courses were observed for 5-HT<sub>2A</sub>R- and 5-HT<sub>2C</sub>R-expressing cells in generating both types of signaling. Both cell lines also require serum-free preincubation for maximal agonist responses in the pERK assay. However, 5-HT<sub>2A</sub>R-expressing cells showed significant release of <it>Ca<sub>i</sub></it><sup>++ </sup>in response to 5-HT stimulation even when preincubated in serum-replete medium, while the response was completely eliminated by serum in 5-HT<sub>2C</sub>R-expressing cells. Response to another serotonergic ligand (DOI) was eliminated by serum-replete preincubation in both cells lines.</p> <p>Conclusions</p> <p>These data expand our knowledge of differences in ligand-stimulated signaling cascades between 5-HT<sub>2A</sub>R and 5-HT<sub>2C</sub>R. Our parallel assays can be applied to other cell and receptor systems for monitoring and dissecting concurrent signaling responses.</p
