    Omacetaxine may have a role in chronic myeloid leukaemia eradication through downregulation of Mcl-1 and induction of apoptosis in stem/progenitor cells

    Chronic myeloid leukaemia (CML) is maintained by a rare population of tyrosine kinase inhibitor (TKI)-insensitive malignant stem cells. Our long-term aim is to find a BcrAbl-independent drug that can be combined with a TKI to improve overall disease response in chronic-phase CML. Omacetaxine mepesuccinate, a first in class cetaxine, has been evaluated by clinical trials in TKI-insensitive/resistant CML. Omacetaxine inhibits synthesis of anti-apoptotic proteins of the Bcl-2 family, including (myeloid cell leukaemia) Mcl-1, leading to cell death. Omacetaxine effectively induced apoptosis in primary CML stem cells (CD34<sup>+</sup>38<sup>lo</sup>) by downregulation of Mcl-1 protein. In contrast to our previous findings with TKIs, omacetaxine did not accumulate undivided cells <i>in vitro</i>. Furthermore, the functionality of surviving stem cells following omacetaxine exposure was significantly reduced in a dose-dependant manner, as determined by colony forming cell and the more stringent long-term culture initiating cell colony assays. This stem cell-directed activity was not limited to CML stem cells as both normal and non-CML CD34<sup>+</sup> cells were sensitive to inhibition. Thus, although omacetaxine is not leukaemia stem cell specific, its ability to induce apoptosis of leukaemic stem cells distinguishes it from TKIs and creates the potential for a curative strategy for persistent disease

    Los patines como contenido de educación física en primaria: desarrollo del perfil competencial

    Las actividades físicas con patines ofrecen grandes posibilidades pedagógicas para desarrollar en el área de Educación Física (EF). El objetivo de este trabajo es presentar una propuesta didáctica coherente basada en las actividades físicas con patines para los tres últimos cursos de la etapa de Educación Primaria. Se comienza con el desarrollo de los aprendizajes fundamentales hasta la adquisición de los aprendizajes específicos del patinaje artístico y del hockey sobre patines. Para ello se va utilizar el juego como eje central en el proceso de enseñanza-aprendizaje. La metodología está basada en la enseñanza comprensiva y constructivista del deporte. Skates-based physical activities have a great pedagogy potential to develop in the area of Physical Education (PE). The purpose of this study is to present a consistent didactic proposal based on skates-based physical activities for the last three years of Elementary School. It starts with the fundamental learning development of figure skating and roller hockey until the acquisition of the more specific learning.     Physico-chemical characterization of chitosan-based edible films incorporating bioactive compounds of different molecular weight

    Chitosan packaging films containing different bioactive compounds (a peptide fraction from whey protein concentrate (WPC) hydrolysate, glycomacropeptide (GMP) and lactoferrin) were produced and their mechanical and barrier properties were evaluated. The molecular weight of protein-based compounds was determined using SDS–PAGE. The addition of GMP and lactoferrin to chitosan film caused a significant reduction of tensile strength and the elongation-at-break significantly increased with the incorporation of lactoferrin. The addition of protein-based compounds also affected gas permeability: a significant decrease in water vapor permeability was observed with the incorporation of lactoferrin; oxygen permeability significantly decreased with the addition of GMP and lactoferrin and carbon dioxide permeability significantly decreased with the incorporation of all of the protein-based compounds. Such results were related with film’s hydrophilicity and crystallinity. This manuscript contributes to the establishment of an approach to optimize edible films performance based on physico-chemical properties, aiming at a higher benefit for the consumer.The authors gratefully acknowledge LoicHilliou for the fruitful discussions on the results and Ana Nicolau for helping in confocal analysis.     Tratamiento de las disciplinas de raqueta y pala en educación física

    Los deportes de raqueta y pala albergan grandes posibilidades pedagógicas sensibles de desarrollar en el área de Educación Física. Desde la Educación Primaria al Bachillerato pasando por la Educación Secundaria Obligatoria, estas disciplinas pueden y deben abordarse en nuestra área. En su tratamiento pedagógico se deben determinar tanto los objetivos didácticos como los aprendizajes específicos. La metodología a seguir parte de unos ejes de progreso que irán desarrollándose a lo largo de toda la etapa educativa. Finalmente, la evaluación cierra el proceso a partir de unas orientaciones específicas para los deportes de raqueta y pala, así como con la selección de los instrumentos de evaluación idóneos. Racket sports have a great potential for pedagogy in Physical Education. All these disciplines should and could be taught in all kind of education, from primary till secondary school to the higher education. The didactic and specific learning objectives must be identified in the pedagogical treatment of racket sports. The methodological aspects are based on some fundamental axes which will be developed throughout the whole stage of education. Eventually, the evaluation process closes the circle with some specific orientations for racket sports as well as the selection of suitable evaluation methods

    The Q2Q^2 dependence of the measured asymmetry A1A_1: the test of the Bjorken sum rule

    We analyse the proton and deutron data on spin dependent asymmetry A1(x,Q2)A_1(x,Q^2) supposing the DIS structure functions g1(x,Q2)g_1(x,Q^2) and F3(x,Q2)F_3(x,Q^2) have the similar Q2Q^2-dependence. As a result, we have obtained that Γ1p−Γ1n=0.190±0.038\Gamma_1^p - \Gamma_1^n = 0.190 \pm 0.038 at Q2=10GeV2Q^2= 10 GeV^2 and Γ1p−Γ1n=0.165±0.026\Gamma_1^p - \Gamma_1^n = 0.165 \pm 0.026 at Q2=3GeV2Q^2= 3 GeV^2, what is in the best agreement with the Bjorken sum rule predictions.Comment: LaTeX, 5 pages, no figures, to be published in JETP Letter

    A genetic interaction of NRXN2 with GABRE, SYT1 and CASK in migraine patients: a case-control study

    Background: Migraine is a multifactorial disorder that is more frequent (two to four times) in women than in men. In recent years, our research group has focused on the role of neurotransmitter release and its regulation. Neurexin (NRXN2) is one of the components of the synaptic vesicle machinery, responsible for connecting intracellular fusion proteins and synaptic vesicles. Our aim was to continue exploring the role and interaction of proteins involved in the control and promotion of neurotransmission in migraine susceptibility. Methods: A case-control study was performed comprising 183 migraineurs (148 females and 35 males) and 265 migraine-free controls (202 females and 63 males). Tagging single nucleotide polymorphisms of NRXN2 were genotyped to assess the association between NRXN2 and migraine susceptibility. The χ 2 test was used to compare allele frequencies in cases and controls and odds ratios were estimated with 95% confidence intervals. Haplotype frequencies were compared between groups. Gene-gene interactions were analysed using the Multifactor Dimensionality Reduction v2.0. Results: We found a statistically significant interaction model (p = 0.009) in the female group between the genotypes CG of rs477138 (NRXN2) and CT of rs1158605 (GABRE). This interaction was validated by logistic regression, showing a significant risk effect [OR = 4.78 (95%CI: 1.76–12.97)] after a Bonferroni correction. Our data also supports a statistically significant interaction model (p = 0.011) in the female group between the GG of rs477138 in NRXN2 and, the rs2244325's GG genotype and rs2998250’s CC genotype of CASK. This interaction was also validated by logistic regression, with a protective effect [OR = 0.08 (95%CI: 0.01–0.75)]. A weak interaction model was found between NRXN2-SYT1.     Análisis de los parámetros temporales que intervienen en la competición del pádel

    El estudio de la estructura temporal de un deporte informa sobre el tipo de esfuerzo que se produce en la competición. El objetivo de la presente investigación ha sido determinar las variables temporales que intervienen en la competición del pádel. Se analizaron 13 partidos cuantificándose los parámetros temporales de juego y la densidad de trabajo. Los resultados señalan que la competición masculina tiene una mayor duración que la femenina. El tiempo medio de cada jugada es superior a otros deportes de raqueta. No se han encontrado diferencias significativas entre géneros en las variables analizadas. Las características de juego del pádel muestran un predominio del tiempo de pausa sobre el de esfuerzo. El análisis de la estructura temporal de una competición es una herramienta útil para conocer los indicadores de rendimiento. Los resultados obtenidos permiten desarrollar programas de entrenamiento adecuados a las necesidades de juego durante la competición en el pádel. The evaluation of a sport temporal structure provides information on the effort produced during competitions. The purpose of this study was that of evaluating the temporal variables involved in padel competition. Thirteen matches were evaluated in order to quantity game time parameters and work density. The results showed than male competitions are longer than female ones. Mean play time is longer than that of other racket sports. Rest time is predominant over play time. The analysis of temporal game structure is useful to understand padel performances parameters. The results obtained allow developing more adequate and effective padel competitions training programs
