    The anticancer effect related to disturbances in redox balance on Caco-2 cells caused by an alkynyl gold(I) complex

    The alkynyl gold(I) derivative Au(C=CPh)(PTA)] (PTA = 1, 3, 5-triaza-7-phosphaadamantane) induces apoptosis in colorectal carcinoma tumour cells (Caco-2) without affecting to normal enterocytes. Au(C=CPh)(PTA)] is a slight lipophilic drug, stable in PBS (Phosphate Buffered Saline) and able to bind BSA (Bovin Serum Albumin) by hydrophobic interactions. Once inside the cell, Au(C=CPh)(PTA)] targets seleno proteins such as Thioredoxin Reductase 1, increasing ROS (Reactive Oxygen Species) levels, reducing cell viability and proliferation and inducing mitochondrial apoptotic pathway, pro-apoptotic and anti-apoptotic protein imbalance, loss of mitochondrial membrane potential, cytochrome c release and activation of caspases 9 and 3. Moreover, unlike other metal-based drugs such as cisplatin, Au(C=CPh)(PTA)] does not target nucleic acid, reducing the risk of side mutation in the DNA. In consequence, our results predict a promising future for Au(C=CPh)(PTA)] as a chemotherapeutic agent for colorectal carcinoma

    Phenolic compounds reduce the fat content in caenorhabditis elegans by affecting lipogenesis, lipolysis, and different stress responses

    Supplementation with bioactive compounds capable of regulating energy homeostasis is a promising strategy to manage obesity. Here, we have screened the ability of different phenolic compounds (myricetin, kaempferol, naringin, hesperidin, apigenin, luteolin, resveratrol, curcumin, and epicatechin) and phenolic acids (p-coumaric, ellagic, ferulic, gallic, and vanillic acids) regulating C. elegans fat accumulation. Resveratrol exhibited the strongest lipid-reducing activity, which was accompanied by the improvement of lifespan, oxidative stress, and aging, without affecting worm development. Whole-genome expression microarrays demonstrated that resveratrol affected fat mobilization, fatty acid metabolism, and unfolded protein response of the endoplasmic reticulum (UPRER), mimicking the response to calorie restriction. Apigenin induced the oxidative stress response and lipid mobilization, while vanillic acid affected the unfolded-protein response in ER. In summary, our data demonstrates that phenolic compounds exert a lipid-reducing activity in C. elegans through different biological processes and signaling pathways, including those related with lipid mobilization and fatty acid metabolism, oxidative stress, aging, and UPR-ER response. These findings open the door to the possibility of combining them in order to achieve complementary activity against obesity-related disorders

Moreover, few of them have been encapsulated in drug delivery systems to improve their therapeutic potential to achieve a sustained or controlled release compared to the administration of equivalent doses of the free compounds. Nanoscience has arisen in the last decades as a potential field of study in drug delivery because nanomaterials may overcome the main current limitations to achieve an efficient and localized drug delivery by improving the targeted delivery and providing a sustained or controlled delivery to prolong the therapeutic effect.     Segovia's 'Green Plan' is the putting into practice of a methodology for the action, both general and peculiar, in order to defend the town's sustainability. The aim of the Plan is the recovery, preservation and improvement of the landscape and ecological quality of the town and surroundings, starting from the analysis of the urban and peripheral environment, and on the basis of a programme founded upon the comprehensive understanding of environmental aspects. This article describes not only the steps taken for the writing of the Plan and the subsequent elaboration of a proposal for general action, but also the development of this action. The results of the revision and evaluation of the Plan after eight years of execution are also explained, as well as the ensuing strategy for the completion and improvement of the initial Plan.El Plan Verde de Segovia es la puesta en practica de una metodología de actuación general y peculiar para defender la sostenibilidad de la ciudad. El objeto del referido Plan es la recuperación, conservación y mejora de la calidad ecológica y paisajística de la ciudad y su entorno, a partir de un análisis del medio urbano y periurbano, y sobre la base de una programación fundamentada en la visión integral de los aspectos ambientales.     The application of plant extracts for therapeutic purposes has been used in traditional medicine since the plants are a source of a great variety of chemical compounds that possess biological activity. Actually, the effect of these extracts on diseases such as cancer is being widely studied. Colorectal adenocarcinoma is one of the main causes of cancer related to death and the second most prevalent carcinoma in Western countries. The aim of this work is to study the possible effect of two fenugreek (Trigonella foenum graecum) protein hydrolysates on treatment and progression of colorectal cancer. Fenugreek proteins from seeds were hydrolysed by using two enzymes separately, which are named Purafect and Esperase, and were then tested on differentiated and undifferentiated human colonic adenocarcinoma Caco2/TC7 cells. Both hydrolysates did not affect the growth of differentiated cells, while they caused a decrease in undifferentiated cell proliferation by early apoptosis and cell cycle arrest in phase G1. This was triggered by a mitochondrial membrane permeabilization, cytochrome C release to cytoplasm, and caspase-3 activation. In addition, the hydrolysates of fenugreek proteins displayed antioxidant activity since they reduce the intracellular levels of ROS. These findings suggest that fenugreek protein hydrolysates could be used as nutraceutical molecules in colorectal cancer treatment

    Colloidal crystals of polystyrene particles of 1.0, 1.4 and 2.8 μm diameter have been prepared by vertical deposition. The influence of parameters such as temperature, particle size and concentration as well as dispersion medium has been studied. The size of domain and the crystalline structure of the particle arrays have been analyzed by optical microscopy. The quality of the crystals has been improved (minimizing cracks) by controlling sedimentation (density matching), evaporation (volatility of the medium) and drying (co-solvents)

    Association of the Gut Microbiota with the Host's Health through an Analysis of Biochemical Markers, Dietary Estimation, and Microbial Composition

This research includes 60 Spanish adults aged 47.3 ± 11.2 years old. Biochemical and anthropometric measurements, and a self-referred dietary survey (food frequency questionnaire), were analyzed and compared with the participant's gut microbiota composition analyzed by 16s rDNA sequencing. Several bacterial strains differed significantly with the biochemical markers analyzed, suggesting an involvement in the participant's metabolic health. Lower levels of Lactobacillaceae and Oscillospiraceae and an increase in Pasteurellaceae, Phascolarctobacterium, and Haemophilus were observed in individuals with higher AST levels. Higher levels of the Christensenellaceae and a decrease in Peptococcaceae were associated with higher levels of HDL-c. High levels of Phascolarctobacterium and Peptococcus and low levels of Butyricicoccus were found in individuals with higher insulin levels. This study also identified associations between bacteria and specific food groups, such as an increase in lactic acid bacteria with the consumption of fermented dairy products or an increase in Verrucomicrobiaceae with the consumption of olive oil. In conclusion, this study reinforces the idea that specific food groups can favorably modulate gut microbiota composition and have an impact on host's health

    Influence of Extraction Solvent on the Biological Properties of Maritime Pine Bark (Pinus pinaster)

    Maritime pine bark (Pinus pinaster Aiton subsp. atlantica) is rich in polyphenols with known bioactive properties which are beneficial for human health. However, biological activities of bark extracts depend on the type of polyphenols extracted and the characteristics of these extractives depend on several factors such as the type of solvents used. The in uence of the extraction solvent on the composition and consequently on the properties of the extracts has been poorly described. Thus, in this study the in uence of the extraction solvent (water, ethanol and ethanol-water (50/50 v/v%)) on the antibacterial and anticancer properties of P. pinaster bark samples were evaluated. LC-DAD-MS profiling of the different extracts was also carried out to study their polyphenol composition. Results show that extraction solvent must be carefully chosen with respect to foreseeing use of bark extracts, since ethanolic and hydroethanolic extracts displayed the greatest antibacterial activity whereas water extracts showed increased anticancer properties. © 2022.     CLOCK, PER2 and BMAL1 DNA methylation: association with obesity and metabolic syndrome characteristics and monounsaturated fat intake

    The circadian clock system instructs 24-h rhythmicity on gene expression in essentially all cells, including adipocytes, and epigenetic mechanisms may participate in this regulation. The aim of this research was to investigate the influence of obesity and metabolic syndrome (MetS) features in clock gene methylation and the involvement of these epigenetic modifications in the outcomes. Sixty normal-weight, overweight and obese women followed a 16-weeks weight reduction program. DNA methylation levels at different CpG sites of CLOCK, BMAL1 and PER2 genes were analyzed by Sequenom's MassARRAY in white blood cells obtained before the treatment. Statistical differences between normal-weight and overweight + obese subjects were found in the methylation status of different CpG sites of CLOCK (CpGs 1, 5-6, 8 and 11-14) and, with lower statistical significance, in BMAL1 (CpGs 6-7, 8, 15 and 16-17). The methylation pattern of different CpG sites of the three genes showed significant associations with anthropometric parameters such as body mass index and adiposity, and with a MetS score. Moreover, the baseline methylation levels of CLOCK CpG 1 and PER2 CpGs 2-3 and 25 correlated with the magnitude of weight loss. Interestingly, the percentage of methylation of CLOCK CpGs 1 and 8 showed associations with the intake of monounsaturated and polyunsaturated fatty acids. This study demonstrates for the first time an association between methylation status of CpG sites located in clock genes (CLOCK, BMAL1 and PER2) with obesity, MetS and weight loss. Moreover, the methylation status of different CpG sites in CLOCK and PER2 could be used as biomarkers of weight-loss success, particularly CLOCK CPGs 5-6

    Fenugreek proteins and their hydrolysates prevent hypercholesterolemia and enhance the HDL antioxidant properties in rats

    Purpose This paper aims to investigate the in vivo hypocholesterolemic property of fenugreek proteins (FP), Purafect-fenugreek protein hydrolysate (PFPH) and Esperase-fenugreek protein hydrolysate (EFPH) on high cholesterol (HC)-fed rats. Design/methodology/approach Rats were randomized into five groups: four were fed for four weeks a hypercholesterolemic diet and the tested products were given by gavage. The fifth group was taken as control (C) receiving the same diet without cholesterol. Findings Results showed that the elevated aspartate aminotransferase activity in HC group plasma was significantly corrected by FP and EFPH administration (-33 per cent; p = 0.0003). HC liver lipids and total cholesterol (TC) contents were not markedly affected by FP and EFPH. However, liver triglycerides (TG) contents trended to decrease in FP rats vs HC (p = 0.07), while, the TG decrease was significant in groups fed the proteins hydrolysates (p = 0.02). On the other hand, serum TC and TG decreased by 53 per cent (p = 0.0003) and 20 per cent (p = 0.04), respectively, in FP treated rats compared to HC group. This decrease was associated with a high fecal cholesterol excretion (2.5-fold higher in FP vs HC; p = 0.0001). Likewise, EFPH-treated rats exhibited lower TC compared to HC rats (p = 0.004). The very low density lipoprotins was the main affected fraction in these two groups, while there were no significant difference in apolipoproteins (Apo) B, A-I and A-IV contents between the different groups, except in FP group, where Apo A-I and A-IV decreased by 26 and 17 per cent, respectively, compared to C rats (p = 0.02). The high density lipoproteins (HDL) of rats treated with proteins hydrolysates showed a better antioxidant property compared to those of HC rats, which was accompanied with an increase in paraoxonase activity when compared to HC group. Originality/value Unlike PFPH which had almost no effect, FPs and EFPH could constitute a nutraceutical ingredient in cardiovascular disease management
