    In vitro effects of regulators on growth and morphogenesis of Ocimum basilicum L. ‘Alfavaca Green’ stem apexes

    Large-scale cultivation of contamination free plants requires a good standardization protocol and production methods. Basil is widely used for cosmetics, food and pharmaceutical industries as it is rich in many bioactive compounds. This present study aimed to evaluate the growth and in vitro anatomical aspects of apical buds of basil grown under different concentrations rowth regulators like: NAA (Naphthalenoacetic Acid), BAP (6-benzylaminopurine), and KIN (Kinetin). The in vitro establishment was evaluated every 20 days to calculate the, the percentage of plants with calluses, appearance of the roots, any abnormal seedlings, any oxidized seedlings, and the number of sprouts per plant. Growth, physiological, and morpho-anatomical evaluations were performed at 80 days. Basal callogenesis was observed when cytokinin’s and auxins are used in combination. Auxin treatments caused hyperhydricity in the stems and leaves. Medium A2 (0.05 mg L -1 of NAA and 0.1 mg L -1 of BAP), and A3 (0.05 mg L -1 of NAA and 0.1 mg L -1 of KIN) resulted in the best development of basil plants, cultivar ‘Alfavaca Green’. The A2 produced plants with greater numbers of leaves, an average bud length of 59.81 mm, and the best root properties. A2 and A1 have a higher percentage of hyperhydricity (83 and 67%). The A3 resulted in an acceptable number of leaves (range: 21–39), and this treatment produced the best shoot properties as well as fewer plants with hyperhydricity. In addition, the A3 treatment produced plants with a shoot length, high shoot fresh and dry mass (2.82 and 0.23 g), high chlorophyll index and leaf anatomy that was similar to the control. Excluding the control, the other treatments presented more than 90% of the explants with calluses in their bases

    Copper modulates the biochemical and enzymatic activity and growth of tomato cultivars grown in vitro

This study sought to identify the effects of exposure to Cu on the growth and biochemical activity of two tomato cultivars. Tomato seeds of ‘Carolina’ and ‘Cereja’ cultivars were disinfected and inoculated in MS medium plus copper sulfate concentrations (CuSO4) (default MS, 25, 50, and 100 µm) and had their growth monitored for 30 days. It was estimated that the growth and biomass accumulation of tomato plants ‘Carolina’ and ‘Cereja’, both from the aerial part and the roots, were benefited by 25 e 50 µm of CuSO4. However, it was observed that these concentrations were inefficient in controlling hyperhydricity and leaf deformation. There was a reduction of these phenomena in the treatment with 100 µm, in both cultivars. Tomato of ‘Carolina’ cultivar subjected to 100 µm showed an increase in anthocyanins and superoxide dismutase (SOD) activity in the root system. There was a reduction of catalase (CAT) activity in shoots exposed to Cu. ‘Cereja’ tomatoes subjected to 100 µm showed an increase in CAT and SOD activity in shoots and roots, respectively. It was concluded that the ‘Carolina’ and ‘Cereja’ tomatoes have their growth impaired when exposed to 100 µm CuSO4. Concentrations higher than 50 µm of CuSO4 cause an increase in the antioxidant activity in the shoot of tomato plants from the ‘Carolina’ cultivar.     Dentre os fatores físicos que exercem influência sobre os insetos, os mais importantes são a temperatura, a umidade, luz e o vento. Quanto à temperatura, os insetos, de um modo geral, desenvolvem-se melhor em temperaturas próximas a 25o C, podendo, no entanto, viver numa larga faixa de temperaturas. Dentro da faixa favorável, os insetos apresentam estreitas faixas de melhor desenvolvimento. De forma análoga à temperatura, as exigências de água pelos insetos variam muito, principalmente devido seus hábitos alimentares. A luz é um dos fatores limitantes aos seres vivos, podendo influenciar na reprodução, dispersão, emergência, alimentação e escolha do hospedeiro (BRENNAN & WEINBAUM 2001d). O vento, além de afetar a temperatura e umidade do ambiente, auxilia os insetos nos processos de dispersão e migração, principalmente quando procuram novo habitat, para alimentação e reprodução (LARA, 1979)

    Differential involvement of the canonical and noncanonical inflammasomes in the immune response against infection by the periodontal bacteria Porphyromonas gingivalis and Fusobacterium nucleatum

    We examined the involvement of the P2 × 7 receptor and the canonical and noncanonical inflammasomes in the control of single-species or dual-species infection by the periodontal bacteria Porphyromonas gingivalis and Fusobacterium nucleatum in cells and mice. Stimulation of the P2 × 7 receptor leads to activation of the canonical NLRP3 inflammasome and activation of caspase-1, which leads to cleavage of pro-IL-1β to IL-1β, a key cytokine in the host inflammatory response in periodontal disease. The non-canonical inflammasome pathway involves caspase-11. Thus, wildtype (WT), P2 × 7−/−, caspase-11−/− and caspase-1/11−/− mice were co-infected with both bacterial species. In parallel, bone marrow-derived macrophages (BMDMs) from WT mice and the different knockout mice were infected with P. gingivalis and/or F. nucleatum, and treated or not with extracellular ATP, which is recognized by P2 × 7. F. nucleatum infection alone promoted secretion of IL-1β in BMDMs. Conversely, the canonical pathway involving P2 × 7 and caspase-1 was necessary for secretion of IL-1β in BMDMs infected with P. gingivalis and in the mandible of mice coinfected with P. gingivalis and F. nucleatum. The P2 × 7 pathway can limit bacterial load in single-species and dual-species infection with P. gingivalis and F. nucleatum in BMDMs and in mice. The non-canonical pathway involving caspase-11 was required for secretion of IL-1β induced by F. nucleatum infection in BMDMs, without treatment with ATP. Caspase-11 was also required for induction of cell death during infection with F. nucleatum and contributed to limiting bacterial load during F. nucleatum infection in BMDMs and in the gingival tissue of mice coinfected with P. gingivalis and F. nucleatum. Together, these data suggest that the P2 × 7-caspase-1 and caspase-11 pathways are involved in the immune response against infection by P. gingivalis and F. nucleatum, respectively

    Dissecting stellar chemical abundance space with t-SNE

    In the era of large-scale Galactic astronomy and multi-object spectroscopic stellar surveys, the sample sizes and the number of available stellar chemical abundances have reached dimensions in which it has become difficult to process all the available information in an effective manner. In this paper we demonstrate the use of a dimensionality-reduction technique (t-distributed stochastic neighbour embedding; t-SNE) for analysing the stellar abundance-space distribution. While the non-parametric non-linear behaviour of this technique makes it difficult to estimate the significance of any abundance-space substructure found, we show that our results depend little on parameter choices and are robust to abundance errors. By reanalysing the high-resolution high-signal-to-noise solar-neighbourhood HARPS-GTO sample with t-SNE, we find clearer chemical separations of the high- and low-[α/Fe] disc sequences, hints for multiple populations in the high-[α/Fe] population, and indications that the chemical evolution of the high-[α/Fe] metal-rich stars is connected with the super-metal-rich stars. We also identify a number of chemically peculiar stars, among them a high-confidence s-process-enhanced abundance-ratio pair (HD 91345/HD 126681) with very similar ages and v X and v Y velocities, which we suggest have a common birth origin, possibly a dwarf galaxy. Our results demonstrate the potential of abundance-space t-SNE and similar methods for chemical-tagging studies with large spectroscopic surveys

    Studies regarding parasite fauna in farmed fish are of great relevance to lhe knowledge of the parasites species. allowing interference in their proliferation in order to avoid epizooties and consequently. economical losses, This study was designed to investigate the prevalence and intensity of parasites in fry Arapaima gigas maintained in ponds of a semi-intensive fish farm in Amazonas State, Brazil. On necropsy, 96,0% of A. gigas were found parasitized by Dawestrema cycloancistrioides. Dawestrema cycloancistrioides (Monogenoidea). Trichodina sp., Ichthyobodo sp. (Protozoa). Camallamus tridentatus, Terranova serrata, Goezia spinulosa (Nematoda) and Argulidae. However, D. cycloancistrium. D, cycloancistrioides and Trichodina fariai were the parasites of' greatest intensity. This study is the first to report parasitic infections in farmed A. gigas and the results indicated a high rate of infection that might lead to important changes in the health of the hosts
