    Green Fluorescent Protein (GFP) in Vector Systems Played Sense Role of Epigenetic in Plants

    The green fluorescent protein (GFP) of jellyfish (_Aequorea victoria_) has significant advantages over other reporter genes, because expression can be detected in living cells without any substrates. Recently, epigenetic phenomena are important to consider in plant biotechnology experiments for elucidate unknown mechanism. Therefore, soybean immature cotyledons were generated embryogenesis cells and engineered with two different gene constructs (pHV and pHVS) using gene gun method. Both constructs contain a gene conferring resistance to hygromycin (_hpt_) as a selective marker and a modified glycinin (11S globulin) gene (_V3-1_) as a target. However, sGFP(_S65T_) as a reporter gene was used only in pHVS as a reporter gene for study the relation between using sGFP(_S65T_) and gene silencing phenomena. Fluorescence microscopic was used for screening after the selection of hygromycin, identified clearly the expression of sGFP(_S65T_) in the transformed soybean embryos bombarded with the pHVS construct. Protein analysis was used to detect gene expression overall seeds using SDS-PAGE. Percentage of gene down regulation was highly in pHV construct compared with pHVS. Thus, sGFP(_S65T_) as a reporter gene in vector system may be play useful role for transgenic evaluation and avoid gene silencing in plants for the benefit of plant transformation system

    Five enzyme systems, PGM 1 , PGM 2 , ADA, 6-PGD and AK, were examined by electrophoresis in over 4000 samples from Hiroshima and Nagasaki for the frequencies of common and rare variants. In the PGM 1 , system, the PGM 2 1 allele and PGM 7 1 ; allele were found in polymorphic proportions. I n addition, five kinds of slow variants and three types of fast variants of PGM 1 were detected. The PGM 3 NGS 1 1 allele was found in five individuals from Nagasaki, but was not observed in samples from Hiroshima. There were no variants of PGM 2 . Three kinds of fast variants of 6-PGD were detected. NO variation in AK was observed. There were no rare variants of ADA. The 6-PGD c allele had a frequency of 0.084 in Hiroshima, and 0.093 in Nagasaki, and the ADA 2 allele frequencies of 0.025 in Hiroshima and 0.032 in Nagasaki.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/65524/1/j.1469-1809.1977.tb01912.x.pd

    Canonical orbits for rapidly deforming planar microswimmers in shear flow

    Classically, the rotation of ellipsoids in shear Stokes flow is captured by Jeffery’s orbits. Here, we demonstrate that Jeffery’s orbits also describe general shape-deforming swimmers moving in the plane of a shear flow, employing only basic properties of Stokes flow and a multiple-scales asymptotic analysis. In doing so, we support the use of these simple models for capturing shape-changing swimmer dynamics in studies of active matter and highlight the ubiquity of ellipsoid-like dynamics in complex systems. This result is robust to weakly confounding effects, such as distant boundaries

    Neutron scattering study of magnetic ordering and excitations in the ternary rare-earth diborocarbide Ce^{11}B_2C_2

    Neutron scattering experiments have been performed on the ternary rare-earth diborocarbide Ce11^{11}B2_2C2_2. The powder diffraction experiment confirms formation of a long-range magnetic order at TN=7.3T_{\rm N} = 7.3 K, where a sinusoidally modulated structure is realized with the modulation vector q=[0.167(3),0.167(3),0.114(3)]{\bm q} = [0.167(3), 0.167(3), 0.114(3)]. Inelastic excitation spectra in the paramagnetic phase comprise significantly broad quasielastic and inelastic peaks centered at ω0,8\hbar \omega \approx 0, 8 and 65 meV. Crystalline-electric-field (CEF) analysis satisfactorily reproduces the observed spectra, confirming their CEF origin. The broadness of the quasielastic peak indicates strong spin fluctuations due to coupling between localized 4f4f spins and conduction electrons in the paramagnetic phase. A prominent feature is suppression of the quasielastic fluctuations, and concomitant growth of a sharp inelastic peak in a low energy region below TNT_{\rm N}. This suggests dissociation of the conduction and localized 4f4f electrons on ordering, and contrasts the presently observed incommensurate phase with spin-density-wave order frequently seen in heavy fermion compounds, such as Ce(Ru1x_{1-x}Lax_x)2_2Si2_2.Comment: accepted for publication in Phys.     Efficient production of transgenic soybean (Glycine max [L] Merrill) plants mediated via whisker-supersonic (WSS) method

    The present study was designed to evaluate the transformation efficiency and proof the capability of whisker supersonic (WSS) method as an alternative option for soybean (Glycine max [L] Merrill)transformation. We compared soybean transformation efficiency obtained by WSS-mediated with that of particle bombardment transformation by carrying out molecular analysis of the T0 plants in two independent experiments. For this, we used for both transformation techniques the same genotype, the same plasmid and the same selection method. To assess the efficiency of soybean genetictransformation, we evaluated the efficiency of multi gene transformation by the selection with hygromycin and the expression of green fluorescent protein [sGFP (S65T)] resulted from both techniques. Regenerable embryogenic cells were induced from immature cotyledons of soybean c.v Jack on MSD40 media within 3 weeks then proliferated on FN lite liquid media and engineered with pUHG gene construct through both WSS and particle bombardment-mediated transformation. The pUHG was constructed with pUC 19 and contain the hpt gene conferring resistance to hygromycin as a selective marker and sGFP(S65T) as a reporter gene. Fluorescence microscopy screening after the selection of hygromycin, identified the clearly expression of sGFP(S65T) in the transformed soybean embryos. Stable integration of the transgenes was confirmed by polymerase chain reaction (PCR) andSouthern blot analysis. The average transformation efficiency achieved with WSS was higher than that obtained by particle bombardment and hence it may represent an alternative method for soybeantransformation
