    A Novel Putative miRNA Target Enhancer Signal

    It is known that miRNA target sites are very short and the effect of miRNA-target site interaction alone appears as being unspecific. Recent experiments suggest further context signals involved in miRNA target site recognition and regulation. Here, we present a novel GC-rich RNA motif downstream of experimentally supported miRNA target sites in human mRNAs with no similarity to previously reported functional motifs. We demonstrate that the novel motif can be found in at least one third of all transcripts regulated by miRNAs. Furthermore, we show that motif occurrence and the frequency of miRNA target sites as well as the stability of their duplex structures correlate. The finding, that the novel motif is significantly associated with miRNA target sites, suggests a functional role of the motif in miRNA target site biology.     Turbiditic trench deposits at the South-Chilean active margin: A PleistoceneHolocene record of climate and tectonics

    The active plate margin of South America is characterized by a frequent occurrence of large and devastating subduction earthquakes. Here we focus on marine sedimentary records off Southern Chile that are archiving the regional paleoseismic history over the Holocene and Late Pleistocene. The investigated records - Ocean Drilling Program (ODP) Site 1232 and SONNE core 50SL - are located at similar to 40 degrees S and similar to 38 degrees S, within the Peru-Chilc trench, and are characterized by frequent interbedded strata of turbiditic and hemipelagic origin. On the basis of the sedimentological characteristics and the association with the active margin of Southern Chile, we assume that the turbidites are mainly scismically triggered, and may be considered as paleo-megaearthquake indicators. However, the long-term changes in turbidite recurrence times appear to be strongly influenced by climate and sea level changes as well. During sea level highstands in the Holocene and Marine Isotope Stage (MIS) 5, recurrence times of turbiditic layers are substantially higher, primarily reflecting a climate-induced reduction of sediment availability and enhanced slope stability. In addition, segmented tectonic uplift changes and related drainage inversions likely influenced the postglacial decrease in turbidite frequencies. Glacial turbidite recurrence times (including MIS 2, MIS 3, cold substages of MIS 5, and MIS 6), on the other hand, are within the same order of magnitude as earthquake recurrence times derived from the historical record and other terrestrial paleoseismic archives of the region. Only during these cold stages sediment availability and slope instability were high enough to enable recording of the complete sequence of large earthquakes in Southern Chile. Our data thus suggest that earthquake recurrence times on the order of 100 to 200 years are a persistent feature at least during the last glacial period

    Rumen microbiota and its relation to fermentation in lactose-fed calves

    In our previous studies, we revealed the effect of lactose inclusion in calf starters on the growth performance and gut development of calves. We conducted the present study as a follow-up study to identify the shift in rumen microbiota and its relation to rumen fermentation when calves are fed a lactose-containing starter. Thirty Holstein bull calves were divided into 2 calf starter treatment groups: texturized calf starter (i.e., control; n = 15) or calf starter in which starch was replaced with lactose at 10% (i.e., LAC10; n = 15) on a dry matter basis. All calves were fed their respective treatment calf starter ad libitum from d 7, and kleingrass hay from d 35. Rumen digesta were collected on d 80 (i.e., 3 wk after weaning) and used to analyze rumen microbiota and fermentation products. There was no apparent effect of lactose feeding on the alpha-diversity and overall composition of rumen microbiota. Amplicon sequencing and real-time PCR quantification of the 16S rRNA gene confirmed that the abundance of butyrate producing bacteria (i.e., Butyrivibrio group and Megasphaera elsdenii) did not differ between the control and LAC10 groups. Conversely, the relative abundance of Mitsuokella spp., which produce lactate, succinate, and acetate, was significantly higher in the rumen of calves that were fed lactose, whereas the lactate concentration did not differ between the control and LAC10 groups. These findings suggest that the lactate production can be elevated by an increase of Mitsuokella spp. and then converted into butyrate, not propionate, since the proportion of propionate was lower in lactose-fed calves. In addition, we observed a higher abundance of Coriobacteriaceae and Pseudoramibacter-Eubacterium in the LAC10 group. Both these bacterial taxa include acetate-producing bacteria, and a positive correlation between the acetate-to-propionate ratio and the abundance of Pseudoramibacter-Eubacterium was observed. Therefore, the higher abundance of Coriobacteriaceae, Mitsuokella spp., and Pseudoramibacter-Eubacterium in the rumen of lactose-fed calves partially explains the increase in the proportion of rumen acetate that was observed in our previous study

    Interaction of N-Alkylanthracyclines With Lipid Bilayers: Correlations Between Partition Coefficients, Lipid Phase Distributions and Thermotropic Behavior

    The thermotropic behavior of multilamellar vesicles of dipalmitoylphosphatidylcholine (DPPC), or of DPPC in admixture with cardiolipin or cholesterol, in the presence of various N-alkyl derivatives of both adriamycin-14-valerate has been investigated by high sensitivity differential scanning calorimetry. The analogues, particularly the 14-valerate derivatives, which were most lipophilic as judged by their lipid/buffer, and to a lesser extent by their octanol/buffer, partition coefficients, were the most effective in depressing the tm of the investigated lipids; correlations, however, were not absolute. Other factors, such as the distribution of the drugs between the solid and liquid-crystalline phases of the bilayer, were also important to the observed membrane perturbations. With all anthracyclines, however, no major changes in the transition enthalpy were observed. In the case of vesicles prepared from pure DPPC, curve fitting analysis based on ideal solution theory (J.M. Sturtevant (1984) Proc. Natl. Acad. Sci. USA 81, 1398-1400) applied at relatively low drug concentrations where single peak transitions were produced, adequately described the differential scanning calorimetric results. At high drug concentrations, however, the presence of multipeak transitions were indicative of non-ideality
