    New Methodological Approaches for Change in Traditional Sectors: The Case of the Portuguese Fisheries Socio-Economic System

    This paper summarises the methodological approach and main results of the MARHE project (Employment and Human Resources in the Fisheries Socio-Economic System). This project had as its main aim the search for alternative futures for the fisheries sector in Portugal, with particular attention being paid to the human resources situation and the working and living conditions of the fisheries-dependent populations in the coastal areas. This is a particularly interesting case, since fisheries were once an important activity and they are now in deep recession, even though it is generally recognised that the future utilisation of maritime resources offer an immense potential. As part of the research, a Delphi exercise was implemented involving in two successive stages some of the leading actors and experts dealing with the sector in Portugal. Other initiatives were held in the context of the MARHE project providing direct and indirect inputs to the scenarios and recommendations that were put forward in the sequence of the Delphi exercise. Overall the activities described in the paper contributed to the mobilisation of major actors and to discussions that may have practical implication for the future of the sector, if certain conditions are now met in the follow up to the project.Fisheries; Portugal; human resources; scenarios; labour market

    Microwave Imaging to Improve Breast Cancer Diagnosis

    Breast cancer is the most prevalent type of cancer worldwide. The correct diagnosis of Axillary Lymph Nodes (ALNs) is important for an accurate staging of breast cancer. The performance of current imaging modalities for both breast cancer detection and staging is still unsatisfactory. Microwave Imaging (MWI) has been studied to aid breast cancer diagnosis. This thesis addresses several novel aspects of the development of air-operated MWI systems for both breast cancer detection and staging. Firstly, refraction effects in air-operated setups are evaluated to understand whether refraction calculation should be included in image reconstruction algorithms. Then, the research completed towards the development of a MWI system to detect the ALNs is presented. Anthropomorphic numerical phantoms of the axillary region are created, and the dielectric properties of ALNs are estimated from Magnetic Resonance Imaging exams. The first pre-clinical MWI setup tailored to detect ALNs is numerically and experimentally tested. To complement MWI results, the feasibility of using machine learning algorithms to classify healthy and metastasised ALNs using microwave signals is analysed. Finally, an additional study towards breast cancer detection is presented by proposing a prototype which uses a focal system to focus the energy into the breast and decrease the coupling between antennas. The results show refraction calculation may be neglected in low to moderate permittivity media. Moreover, MWI has the potential as an imaging technique to assess ALN diagnosis as estimation of dielectric properties indicate there is sufficient contrast between healthy and metastasised ALNs, and the imaging results obtained in this thesis are promising for ALN detection. The performance of classification models shows these models may potentially give complementary information to imaging results. The proposed breast imaging prototype also shows promising results for breast cancer detection

    CVD of CrO2 Thin Films: Influence of the Deposition Parameters on their Structural and Magnetic Properties

    This work reports on the synthesis of CrO2 thin films by atmospheric pressure CVD using chromium trioxide (CrO3) and oxygen. Highly oriented (100) CrO2 films containing highly oriented (0001) Cr2O3 were grown onto Al2O3(0001) substrates. Films display a sharp magnetic transition at 375 K and a saturation magnetization of 1.92 Bohr magnetons per f.u., close to the bulk value of 2 Bohr magnetons per f.u. for the CrO2.     Determination of Peroxisomal pH in Living Mammalian Cells Using pHRed

The nature of the peroxisomal pH remains somewhat controversial, with several studies reporting conflicting results. Here, we describe in detail a rapid and accurate method for the measurement of peroxisomal pH, using the pHRed sensor protein and confocal microscopy of living mammalian cells. pHRed, a ratiometric sensor of pH, is targeted to the peroxisomes by virtue of a C-terminal targeting sequence. The probe has a maximum fluorescence emission at 610 nm while exhibiting dual excitation peaks at 440 nm and 585 nm, allowing for ratiometric imaging and determination of intracellular pH in live cell microscopy.This work was supported by the Portuguese Foundation for Science and Technology (FCT) (SFRH/BPD/90084/2012 to L.     Spontaneous telomere to telomere fusions occur in unperturbed fission yeast cells

    Telomeres protect eukaryotic chromosomes from illegitimate end-to-end fusions. When this function fails, dicentric chromosomes are formed, triggering breakage-fusion-bridge cycles and genome instability. How efficient is this protection mechanism in normal cells is not fully understood. We created a positive selection assay aimed at capturing chromosome-end fusions in Schizosaccharomyces pombe. We placed telomere sequences with a head to head arrangement in an intron of a selectable marker contained on a plasmid. By linearizing the plasmid between the telomere sequences, we generated a stable mini-chromosome that fails to express the reporter gene. Whenever the ends of the mini-chromosome join, the marker gene is reconstituted and fusions are captured by direct selection. Using telomerase mutants, we recovered several fusion events that lacked telomere sequences. The end-joining reaction involved specific homologous subtelomeric sequences capable of forming hairpins, suggestive of ssDNA stabilization prior to fusing. These events occurred via microhomology-mediated end-joining (MMEJ)/single-strand annealing (SSA) repair and also required MRN/Ctp1. Strikingly, we were able to capture spontaneous telomere-to-telomere fusions in unperturbed cells. Similar to disruption of the telomere regulator Taz1/TRF2, end-joining reactions occurred via non-homologous end-joining (NHEJ) repair. Thus, telomeres undergo fusions prior to becoming critically short, possibly through transient deprotection.     Manifolds associated with (Z2)n(Z_2)^n-colored regular graphs

    In this article we describe a canonical way to expand a certain kind of (Z2)n+1(\mathbb Z_2)^{n+1}-colored regular graphs into closed nn-manifolds by adding cells determined by the edge-colorings inductively. We show that every closed combinatorial nn-manifold can be obtained in this way. When n3n\leq 3, we give simple equivalent conditions for a colored graph to admit an expansion. In addition, we show that if a (Z2)n+1(\mathbb Z_2)^{n+1}-colored regular graph admits an nn-skeletal expansion, then it is realizable as the moment graph of an (n+1)(n+1)-dimensional closed (Z2)n+1(\mathbb Z_2)^{n+1}-manifold.Comment: 20 pages with 9 figures, in AMS-LaTex, v4 added a new section on reconstructing a space with a (Z2)n(Z_2)^n-action for which its moment graph is a given colored grap
