    In vivo evaluation of the delivery and efficacy of a sirolimus-laden polymer gel for inhibition of hyperplasia in a porcine model of arteriovenous hemodialysis graft stenosis

    Journal ArticleSynthetic arteriovenous (AV) hemodialysis grafts are plagued by hyperplasia resulting in occlusion and graft failure yet there are no clinically available preventative treatments. Here the delivery and degradation of a sirolimus-laden polymer gel were monitored in vivo by magnetic resonance imaging (MRI) and its efficacy for inhibiting hyperplasia was evaluated in a porcine model of AV graft stenosis. Synthetic grafts were placed between the carotid artery and ipsilateral jugular vein of swine. A biodegradable polymer gel loaded with sirolimus (2.5 mg/mL) was immediately applied perivascularly to the venous anastomosis, and reapplied by ultrasound-guided injections at one, two and three weeks. Control grafts received neither sirolimus nor polymer. The lumen cross-sectional area at the graft-vein anastomosis was assessed in vivo by non-invasive MRI. The explanted tissues also underwent histological analysis. A specifically developed MRI pulse sequence provided a high contrast-to-noise ratio (CNR) between the polymer and surrounding tissue that allowed confirmation of gel location after injection. Polymer signal decreased up to 80% at three to four weeks after injection, slightly faster than its degradation kinetics in vitro. The MR image of the polymer was confirmed by visual assessment at necropsy. On histological assessment, the mean hyperplasia surface area of the treated graftwas 52% lower than that of the control grafts (0.43 mm2 vs. 0.89mm2; pb0.003), while the minimum cross-sectional lumen area, as measured on MRI, was doubled (5.3mm2 vs 2.5 mm2; pb0.05)

    The Struggle Never Ends (The Reflection on the History of Struggle between Socio-Political Groups in the Creation of Painting)

    This artistic research aims for the creation of contemporary painting. State of art issues of art creation consists of paintings, socio-political, social functions, and visual symbols. The creation goals are to create painting from the ideas of creation and aesthetics visual form, departing from socio-political history condition in the struggle among mass organizations and politic organizations in Indonesia. By utilizing practice-based research method, the researcher is united with the object being worked in an in and through appreciation, must also refer to published creation method, thus that it is not subjective and the explanation is detailed. David Campbell’s creation method is used as the reference, i.e., Preparation, concentration, incubation, illumination, and verification-production. The study results and art design are contemporary painting “Pertarungan tak Kunjung Usai” (The Struggle Never Ends). It portrays the reflection of ideology struggle and socio-political outlook of figures or among community groups. Subject matter shows two figures group who facing and opposing each other with allegory or symbol attributes, such as weaponries, normative books, bulls, and garudas. The setting is on a ruins or ship deck wreckage. The painting is shown in decorative-expressive style, within contemporary aesthetic artistic images. Penelitian artistik ini bertujuan untuk penciptaan karya seni lukis kontemporer. State of art permasalahan penciptaan karya meliputi seni lukis, sosial-politik, fungsi sosial, dan simbol visual. Tujuan penciptaannya menciptakan lukisan dari ide penciptaan dan bentuk visual yang estetik, dari fenomena kondisi sejarah sosial politik dalam pertarungan ormas dan orpol yang ada di Indonesia. Metodenya practice based research, peneliti menyatu dengan objek yang dikerjakan dalam penghayatan secara timbal balik (in and through), juga harus merujuk metode penciptaan terpublikasi, sehingga tidak subjektif dan pemaparannya rinci. Metode penciptaan David Campbell digunakan sebagai rujukan, yaitu preparation, concentration, incubation, illumination, verification-production. Hasil penelitian dan perancangan karya ini adalah karya seni lukis kontemporer "Pertarungan Tak Kunjung Usai". Menggambarkan refleksi pertarungan ideologi dan pandangan sosial politik dari figur-figur atau antarkelompok masyarakat. Subject matter menampilkan dua kelompok figur-figur yang berhadapan dan saling bertentangan dengan masing-masing atribut alegori atau simbolnya, seperti senjata, kitab normatif, banteng, dan garuda. Setingnya ada dalam reruntuhan bangunan atau potongan geladak kapal. Lukisan ini ditampilkan dalam gaya dekoratif ekspresif, dalam citra artistik estetika kontemporer

    : Diarrhea is one of the most frequent diseases of the babies and children in the world. many efforts were made to improve the absorption function of the small intestine, one of the providing of a combination of probiotics (live microorganisms) and prebiotics (substrate), which is known as synbiotic, synbiotic in vivo studies are still very few, especially its handling of non-infectious acute diarrhea is still not yet. This study was aimed to investigate the effect of synbiotic for acute diarrhea in children. This is an a randomized clinical trial double blind in 40 children aged 1-14 years in department of Pediatric, Prof. dr. R. D. Kandou Manado General Hospital. Subjects were randomly allocated into 2 groups, synbiotic and control group. Based on a sample of 40 children study. More men (62.5%) than women (37.5%). Age group 1-5 years is the largest age group with 22 patients. There were no significant differences between treatment groups (sinbiotik) 2.9 days (SB 0.64) and controls 3.2 days (SB 0.61) p = 0.138. conclusion is There is no effect of synbiotic in shortening long of acute diarrhea in children who were treated in the department of Prof. dr. R.D Kandou Manado general hospital

    : Public believes that Lead Tree seeds (Leucaena leucochepala L) as a kind of medicinal plant that have capability of treating a lot of diseases, among them is diabetes. This study aimed to test whether the Lead tree seeds extract can reduce blood glucose levels of Wistar rats (R. norvegicus) which has induced alloxan. The subjects in these research were male wistar rat with total of 18 samples divided into six groups consisting of a negative control group and five groups induced by alloxan with dose 130 mg/kgBW of rat, which can cause wistar rats in hyperglycemic state. Hyperglycemic rats were given lead tree seed extract at a dose of 0,14 mg/200 gBW, 0,3 mg/200 gBW, and 0,6 mg/200 gBW, positive control group was given insulin analogue, and one group was given alloxan. Data obtained from the examination of blood glucose levels in all groups of Wistar rats on day zero, day one and day two every six hours on 0, 6, 12, 18, and 24. The results showed that giving lead tree seed extract does not have the effect to reduce blood glucose levels Wistar rats

    Education in the present has become a necessity, the need for knowledge gained through the learning process both in school and learning from the environment of residence. Mathematical skills are important for everyday life, when students work in activities that are based on everyday life situations; they see ways of applying the mathematical skills they are learning to the real world. Thelearning strategy REACT is one of the learning strategies that use the environment as a source of learning. Besides the teaching learning process with active student learning methods (CBSA) the teacher must be more active in preparing lesson material, planning the process to be carried out such as RPP, preparing evaluation such as LKS and follow-up. From the results of the study the development of learning tools for student activities reached 91.25% and the success rate of students reached 70%. Thus the development of a square and rectangular learning device using theREACTstrategy has a good effect on student activity and student learning outcome

    : People of Indonesia especially in North Sulawesi often use nutmeg powder for wound treatment. Comensalist bacterias that live in human skin, like Staphylococcus aureus and Streptococcus pyogenescan infect open wound especially those that are not treatedwell. This study aimed to find out the result of nutmeg extract inhibition test on Staphylococcus aureus and Streptococcus pyogenes. This was a laboratory experimental study using modified Kirby-Bauer wells at Phytochemistry and Micobriology Laboratory of Sam Ratulangi University Faculty of Mathematics and Science. Nutmeg extract was obtained from maceration process with 96% ethanol. Concentrations of the thick extract that was used were 100 mg/ml, 50 mg/ml, 25 mg/ml, and 12,5 mg/ml. Cefuroxime was used as positive control and aquades was used as negative control. The results showed that aquades had no inhibition zone. Cefuroxime had the widest diameter of inhibition zone. The average of inhibition diameter that was produced by Cefuroxime was 37,33 mm toward S. Aureus bacteria and 42,67 mm toward S. Pyogenes bacteria. Nutmeg extract with 100 mg/ml concentration produced a 25,16 mm inhibition zone diameter on S. Aureus bacteria and 24,16 mm on S. Pyogenes bacteria. Nutmeg extract with 25 mg/ml concentration produced a 19,16 mm diameter on S. Aureus bacteria and 20,66 mm on S. Pyogenes bacteria. While nutmeg extract with 12,5 mg/ml concentration prodyced a 16,16 mm diameter on S. Aureus bacteria and 18,16 mm on S. Pyogenes bacteria. Conclusion: Nutmeg extract has a potential inhibition effect on the growth of S. Aureus and S. Pyogenes bacteria. The inhibition effect of nutmeg extract is stronger on S. Pyogenes than S. Aureus
