    Targeting interleukin-1β protects from aortic aneurysms induced by disrupted transforming growth factor β signaling

    Aortic aneurysms are life-threatening conditions with effective treatments mainly limited to emergency surgery or trans-arterial endovascular stent grafts, thus calling for the identification of specific molecular targets. Genetic studies have highlighted controversial roles of transforming growth factor β (TGF-β) signaling in aneurysm development. Here, we report on aneurysms developing in adult mice after smooth muscle cell (SMC)-specific inactivation of Smad4, an intracellular transducer of TGF-β. The results revealed that Smad4 inhibition activated interleukin-1β (IL-1β) in SMCs. This danger signal later recruited innate immunity in the adventitia through chemokine (C-C motif) ligand 2 (CCL2) and modified the mechanical properties of the aortic wall, thus favoring vessel dilation. SMC-specific Smad4 deletion in Il1r1- or Ccr2-null mice resulted in milder aortic pathology. A chronic treatment with anti-IL-1β antibody effectively hampered aneurysm development. These findings identify a mechanistic target for controlling the progression of aneurysms with compromised TGF-β signaling, such as those driven by SMAD4 mutations

    The efficacy against mosquitoes (Diptera: Culicidae) of a bednet treated with carbamate insecticide [carbosulfan capsule suspension (CS) 200 mg/m(2)] was compared with four types of pyrethroid-treated nets in veranda-trap huts at Yaokoffikro near Bouaké, Côte d'Ivoire, where the malaria vector Anopheles gambiae Giles carries the kdr gene (conferring pyrethroid resistance) at high frequency and Culex quinquefasciatus Say is also pyrethroid resistant. Pyrethroids compared were lambdacyhalothrin CS 18 mg/m(2), alphacypermethrin water dispersible granules (WG) 20 mg/m(2), deltamethrin 50 mg/m(2) (Permanet) and permethrin emulsifiable concentrate (EC) 500 mg/m(2). Insecticidal power and personal protection from mosquito bites were assessed before and after the nets were used for 8 months and hand washed five times in cold soapy water. Before washing, all treatments except permethrin significantly reduced blood-feeding and all had significant insecticidal activity against An. gambiae. The carbosulfan net gave significantly higher killing of An. gambiae than all pyrethroid treatments except the Permanet. Against Culex spp., carbosulfan was more insecticidal and gave a significantly better protective effect than any of the pyrethroid treatments. After washing, treated nets retained various degrees of efficacy against both mosquito genera - but least for the carbosulfan net. Washed nets with three types of pyrethroid treatment (alphacypermethrin, lambdacyhalothrin, permethrin) gave significantly higher mortality rates of Culex than in huts with the same pyrethroid-treated nets before washing. After five washes, the Permanet, which is sold as a long-lasting insecticidal product, performed no better than the other nets in our experimental conditions

They project that the forces fueling the demand for college-educated workers today will continue to grow, along with the income divide between those who have some postsecondary education and those who do not

    Damage identification in warren truss bridges by two different time–frequency algorithms

    Recently, a number of authors have been focusing on drive-by monitoring methods, exploiting sensors mounted on the vehicle rather than on the bridge to be monitored, with clear advantages in terms of cost and flexibility. This work aims at further exploring the feasibility and effectiveness of novel tools for indirect health monitoring of railway structures, by introducing a higher level of accuracy in damage modelling, achieve more close-to-reality results. A numerical study is carried out by means of a FE 3D model of a short span Warren truss bridge, simulating the dynamic interaction of the bridge/track/train structure. Two kinds of defects are simulated, the first one affecting the connection between the lower chord and the side diagonal member, the second one involving the joint between the cross-girder and the lower chord. Accelerations gathered from the train bogie in different working conditions and for different intensities of the damage level are analyzed through two time-frequency algorithms, namely Continuous Wavelet and Huang-Hilbert transforms, to evaluate their robustness to disturbing factors. Compared to previous studies, a complete 3D model of the rail vehicle, together with a 3D structural scheme of the bridge in place of the 2D equivalent scheme widely adopted in the literature, allow a more detailed and realistic representation of the effects of the bridge damage on the vehicle dynamics. Good numerical results are obtained from both the two algorithms in the case of the time-invariant track profile, whereas the Continuous Wavelet Transform is found to be more robust when a deterioration of track irregularity is simulated

    Gas Turbine are nowadays largely used for aircraft propulsion and land-based power generation. The increased attention to environmental aspects has promoted research and development efforts both from manufacturers and research centres. The latest developments in turbinecooling technologies play a critical role in the attempt to increase the efficiency and the specific power of the most advanced designs. Pin fin arrays, in particular, are widely used in jet engine application because of their ability to enhance cooling by providing extended surfaces for conduction and convection. They are also known to be an effective means to create turbulence which naturally increases heat transfer. Pin fin turbulators are typically located inside the trailing edge of high pressure turbine blade where they also act as a structural support. The optimum shapes and spacing of such elements are usually determined experimentally, or more recently, by using Computational Fluid Dynamics (CFD). On the other hand, the comprehension of the real physics controlling the heat transfer enhancement process and the role played by the large scale vortical structures generated by the inserts, still represent a great challenge for fluid mechanic researchers. The problem has been intensively investigated by Ames et al. (2005) by means of an experimental campaign on pin fin matrix. From the numerical point of view, the principal bottleneck of the CFD approach as applied to this kind of massively unsteady flow is related to the high computational cost and to the reliability of the turbulence models. The main objective of this work is to offer a critical analysis of the performance of a cooling device consisting of a pin fin turbulators geometry, as predicted by different CFD models of various complexity, using similar computational technology to integrate the corresponding governing equations. Local velocity and turbulence distributions are presented and compared with available experimental data
