Histidine Residue 94 Is Involved in pH Sensing by Histidine Kinase ArsS of Helicobacter pylori
The ArsRS two-component system is the master regulator of acid adaptation in the human gastric pathogen Helicobacter pylori. Low pH is supposed to trigger the autophosphorylation of the histidine kinase ArsS and the subsequent transfer of the phosphoryl group to its cognate response regulator ArsR which then acts as an activator or repressor of pH-responsive genes. Orthologs of the ArsRS two-component system are also present in H. pylori's close relatives H. hepaticus, Campylobacter jejuni and Wolinella succinogenes which are non-gastric colonizers.In order to investigate the mechanism of acid perception by ArsS, derivatives of H. pylori 26695 expressing ArsS proteins with substitutions of the histidine residues present in its periplasmic input domain were constructed. Analysis of pH-responsive transcription of selected ArsRS target genes in these mutants revealed that H94 is relevant for pH sensing, however, our data indicate that protonatable amino acids other than histidine contribute substantially to acid perception by ArsS. By the construction and analysis of H. pylori mutants carrying arsS allels from the related epsilon-proteobacteria we demonstrate that WS1818 of W. succinogenes efficiently responds to acidic pH.We show that H94 in the input domain of ArsS is crucial for acid perception in H. pylori 26695. In addition our data suggest that ArsS is able to adopt different conformations depending on the degree of protonation of acidic amino acids in the input domain. This might result in different activation states of the histidine kinase allowing a gradual transcriptional response to low pH conditions. Although retaining considerable similarity to ArsS the orthologous proteins of H. hepaticus and C. jejuni may have evolved to sensors of a different environmental stimulus in accordance with the non gastric habitat of these bacteria
Conductivity of interacting spinless fermion systems via the high dimensional approach

Symmetries and Triplet Dispersion in a Modified Shastry-Sutherland Model for SrCu_2(BO_3)_2
We investigate the one-triplet dispersion in a modified Shastry-Sutherland
Model for SrCu_2(BO_3)_2 by means of a series expansion about the limit of
strong dimerization. Our perturbative method is based on a continuous unitary
transformation that maps the original Hamiltonian to an effective, energy
quanta conserving block diagonal Hamiltonian H_{eff}. The dispersion splits
into two branches which are nearly degenerated. We analyse the symmetries of
the model and show that space group operations are necessary to explain the
degeneracy of the dispersion at k=0 and at the border of the magnetic Brillouin
zone. Moreover, we investigate the behaviour of the dispersion for small |k|

Systematic Mapping of the Hubbard Model to the Generalized t-J Model
The generalized t-J model conserving the number of double occupancies is
constructed from the Hubbard model at and in the vicinity of half-filling at
strong coupling. The construction is realized by a self-similar continuous
unitary transformation. The flow equation is closed by a truncation scheme
based on the spatial range of processes. We analyze the conditions under which
the t-J model can be set up and we find that it can only be defined for
sufficiently large interaction. There, the parameters of the effective model

Interactionist theories are considered to have resolved the classic person-situation debate by demonstrating that human behavior is most accurately described as a function of both personal characteristics as well as environmental cues. According to these theories, personality traits form part of the personal characteristics that drive behavior. We suggest that psychological theory stands to gain from also considering personality traits as an important environmental characteristic that shapes sociocultural norms and institutions, and, in turn, behavior. Building on research in geographical psychology, we support this proposition by presenting evidence on the relationship of individual and regional personality with spending behavior. Analyzing the spending records of 111,336 participants (31,915,942 unique transactions) across 374 Local Authority Districts (LAD) in the United Kingdom, we first show that geographic regions with higher aggregate scores on a given personality trait collectively spend more money on categories associated with that trait. Shifting the focus to individual level spending as our behavioral outcome (N = 1,716), we further demonstrate that regional personality of a participant's home LAD predicts individual spending above and beyond individual personality. That is, a person's spending reflects both their own personality traits as well as the personality traits of the people around them. We use conditional random forest predictions to highlight the robustness of these findings in the presence of a comprehensive set of individual and regional control variables. Taken together, our findings empirically support the proposition that spending behaviors reflect personality traits as both personal and environmental characteristics. Accessing Multi-triplons in Spin Ladders using Resonant Inelastic X-ray Scattering
Resonant inelastic x-ray scattering (RIXS) has proven a powerful tool to
investigate dynamic magnetic correlations, yielding complementary insights
compared to neutron scattering. A key issue is the interpretation of
spectroscopic measurements in terms of quasi-particle excitations. In this
paper we study the spin-conserving channel of RIXS spectra for two-leg quantum
spin ladders using continuous unitary transformations. We find that
multi-triplon continua and bound states are the leading contribution, opening
new venues for observing and characterizing multi-triplons in quantum spin

Wet-chemical Passivation of Anisotropic Plasmonic Nanoparticles for LSPR-sensing by a Silica Shell
Metal nanoparticles showing the effect of localized surface plasmon resonance (LSPR), a collective oscillation of the conduction electrons upon interaction with light, represent an interesting tool for bioanalytics. This resonance is influenced by changes in the environment, and can be therefore used for the detection of molecular layers. The sensitivity, this means the extent of wavelength resonance shift per change in refractive index in the environment, represents an important performance parameter. It is higher for silver compared to gold particles, and is also increased for anisotropic particles. So silver triangles show a high potential for highly sensitive plasmonic nanoparticles. However, the stability under ambient conditions is rather poor.
The paper demonstrates the passivation of silver triangles by silica coating using a wet-chemical approach. It compares the sensitivity for particles with and without passivation, and visualizes the passivation effect in a high resolution, single particle TEM study
Staphylococcal Peptidoglycan Co-Localizes with Nod2 and TLR2 and Activates Innate Immune Response via Both Receptors in Primary Murine Keratinocytes
In mammalian host cells staphylococcal peptidoglycan (PGN) is recognized by Nod2. Whether PGN is also recognized by TLR2 is disputed. Here we carried out PGN co-localization and stimulation studies with TLR2 and Nod2 in wild type and mutant host cells. To exclude contamination with lipoproteins, polymeric staphylococcal PGN (PGNpol) was isolated from Staphylococcus aureus Δlgt (lacking lipidated prelipoproteins). PGNpol was biotinylated (PGN-Bio) for fluorescence monitoring with specific antibodies. Keratinocytes from murine oral epithelium (MK) readily internalized PGN-Bio in an endocytosis-like process. In wt MK, PGNpol induced intracellular accumulation of Nod2 and TLR2 and co-localized with Nod2 and TLR2, but not with TLR4. In TLR2-deficient MK Nod2 and in Nod2-deficient MK TLR2 was induced, indicating that PGNpol recognition by Nod2 is independent of TLR2 and vice versa. In both mutants IL-6 and IL-1B release was decreased by approximately 50% compared to wt MK, suggesting that the immune responses induced by Nod2 and TLR2 are comparable and that the two receptors act additively in MK. In TLR2-tranfected HEK293 cells PGNpol induced NFkB-promoter fused luciferase expression. To support the data, co-localization and signaling studies were carried out with SHL-PGN, a lipase protein covalently tethered to PGN-fragments of varying sizes at its C-terminus. SHL-PGN also co-localized with Nod2 or TLR2 and induced their accumulation, while SHL without PGN did not. The results show that staphylococcal PGN not only co-localizes with Nod2 but also with TLR2. PGN is able to stimulate the immune system via both receptors
