    In vitro - Identifizierung fettassimilationsinhibierender BioaktivitÀt

    The aim of this study was to identify substances by in vitro trials which lower postprandial triglycerides by their ability to inhibit pancreatic lipase or the intestinal fatty acid uptake. One of the 24 most effective lipase inhibitory test substances is Tabebuia impetiginosa (Lapacho tea), a tea manufacture of the inner bark of the amazonian taheebo tree. It inhibited the lipase strongly in vitro. In addition, this extract resulted in a significant reduction of postprandial triglycerides in subsequent animal trials with Wistar rats [personal information by JĂŒrgen Schrezenmeir]. The effect of several synthetic PPARgamma- und PPARalpha ligands was investigated on palmitic acid uptake in Caco-2 cells. Only the incubation of the PPARalpha agonist GW7647 in the concentration 1 ”mol/L resulted in a low but significant inhibition of palmitic acid uptake (p = 0,027). Natural PPARgamma and PPARalpha ligands are not applicable for the delay of fatty acid uptake in the intestine. Lapacho tea was identified as a promising candidate for the development of functional food or food additives for the Metabolic Syndrome

    SĂŒdostasien 2001: Transformation, KrĂ€ftevergleich und Kooperation

    "Ein Anfang der 90er Jahre noch boomendes SĂŒdostasien steht 2001 wirtschaftlich und politisch am Scheideweg. WĂ€hrend Nordostasien, jedenfalls momentan, wieder wachsende Investitionen und einen neuen technologischen Schub verzeichnet, bleibt eine dauerhafte Erholung der sĂŒdostasiatischen Volkswirtschaften angesichts schlechter politischer Rahmenbedingungen, steigender Ölpreise und einer AbschwĂ€chung der amerikanischen Konjunktur fraglich. Auch die exportgetriebene, makroökonomische Beruhigung der Jahre 1999/2000 könnte wieder auf dem Spiel stehen, wenn ihr keine konsequente Reform der mikroökonomischen, politischen und Rechtssysteme folgt. DafĂŒr fehlt in vielen FĂ€llen das Bewußtsein und in den meisten FĂ€llen der finanzielle Spielraum. In dem Maße aber, in dem auslĂ€ndische Investoren ihr Heil in Nordostasien suchen, mĂŒssen sĂŒdostasiatische Regierungen einmal mehr ihre WĂ€hrungen verteidigen, ohne das Problem der öffentlichen und privaten Verschuldung auch nur ansatzweise einer Lösung nĂ€herzubringen. Die bis vor einigen Jahren noch hochgepriesene Gemeinschaft SĂŒdostasiatischer Nationen (Association of Southeast Asian Nations, ASEAN) erscheint angesichts der neuen Herausforderungen nahezu gelĂ€hmt. Der traditionelle Konsens ĂŒber gegenseitige Nichteinmischung, durch die noch vor der Krise von 1997/98 erfolgte Norderweiterung untergraben, könnte durch die mit der SchwĂ€chung Indonesiens einhergehenden Verschiebungen im regionalen KrĂ€ftegleichgewicht vollends obsolet werden. Zu den Folgen gehört eine Wiederbelebung der Konkurrenz externer MĂ€chte in SĂŒdostasien, aber auch die abnehmende FĂ€higkeit, gemeinsam ĂŒberzeugend auf nichtmilitĂ€rische Risiken zu reagieren. Die vorliegende Analyse unternimmt eine Bestandsaufnahme der ökonomischen und politischen Transformationsprozesse in den ASEAN-Staaten und versucht, Wechselwirkungen zwischem wirtschaftlichen und politischem Wandel, sowie zwischen Transformation, KrĂ€ftegleichgewicht und Regionalisierung zu identifizieren."     Towards a practical approach for self-consistent large amplitude collective motion

    We investigate the use of an operatorial basis in a self-consistent theory of large amplitude collective motion. For the example of the pairing-plus-quadrupole model, which has been studied previously at equilibrium, we show that a small set of carefully chosen state-dependent basis operators is sufficient to approximate the exact solution of the problem accuratly. This approximation is used to study the interplay of quadrupole and pairing degrees of freedom along the collective path for realistic examples of nuclei. We show how this leads to a viable calculational scheme for studying nuclear structure, and discuss the surprising role of pairing collapse.Comment: 19 pages, 8 figures Revised version To be published in Phys.     In Vitro Analyses of Novel HCN4 Gene Mutations

    Background/Aims: The hyperpolarization-activated cyclic nucleotide-gated cation channel HCN4 contributes significantly to the generation of basic cardiac electrical activity in the sinus node and is a mediator of modulation by ÎČ–adrenergic stimulation. Heterologous expression of sick sinus syndrome (SSS) and bradycardia associated mutations within the human HCN4 gene results in altered channel function. The main aim was to describe the functional characterization of three (two novel and one known) missense mutations of HCN4 identified in families with SSS. Methods: Here, the two-electrode voltage clamp technique on Xenopus laevis oocytes and confocal imaging on transfected COS7 cells respectively, were used to analyze the functional effects of three HCN4 mutations; R378C, R550H, and E1193Q. Membrane surface expressions of wild type and the mutant channels were assessed by confocal microscopy, chemiluminescence assay, and Western blot in COS7 and HeLa cells. Results: The homomeric mutant channels R550H and E1193Q showed loss of function through increased rates of deactivation and distinctly reduced surface expression in all three homomeric mutant channels. HCN4 channels containing R550H and E1193Q mutant subunits only showed minor effects on the voltage dependence and rates of activation/deactivation. In contrast, homomeric R378C exerted a left-shifted activation curve and slowed activation kinetics. These effects were reduced in heteromeric co-expression of R378C with wild-type (WT) channels. Conclusion: Dysfunction of homomeric/heteromeric mutant HCN4-R378C, R550H, and E1193Q channels in the present study was primarily caused by loss of function due to decreased channel surface expression

    Survival benefit with checkpoint inhibitors versus chemotherapy is modified by brain metastases in patients with recurrent small cell lung cancer

    IntroductionSmall cell lung cancer (SCLC) is a rapidly growing malignancy with early distant metastases. Up to 70% will develop brain metastases, and the poor prognosis of these patients has not changed considerably. The potential of checkpoint inhibitors (CPI) in treating recurrent (r/r) SCLC and their effect on brain metastases remain unclear.MethodsIn this retrospective multicenter study, we analyzed r/r SCLC patients receiving second or further-line CPI versus chemotherapy between 2010 and 2020. We applied multivariable-adjusted Cox regression analysis to test for differences in 1-year mortality and real-world progression. We then used interaction analysis to evaluate whether brain metastases (BM) and/or cranial radiotherapy (CRT) modified the effect of CPI versus chemotherapy on overall survival.ResultsAmong 285 patients, 99 (35%) received CPI and 186 (65%) patients received chemotherapy. Most patients (93%) in the CPI group received nivolumab/ipilimumab. Chemotherapy patients were entirely CPI-naĂŻve and only one CPI patient had received atezolizumab for first-line treatment. CPI was associated with a lower risk of 1-year mortality (adjusted Hazard Ratio [HRadj] 0.59, 95% CI 0.42 to 0.82, p=0.002). This benefit was modified by BM and CRT, indicating a pronounced effect in patients without BM (with CRT: HRadj 0.34, p=0.003; no CRT: HRadj 0.50, p=0.05), while there was no effect in patients with BM who received CRT (HRadj 0.85, p=0.59).ConclusionCPI was associated with a lower risk of 1-year mortality compared to chemotherapy. However, the effect on OS was significantly modified by intracranial disease and radiotherapy, suggesting the benefit was driven by patients without BM

    CSF1R inhibitor JNJ-40346527 attenuates microglial proliferation and neurodegeneration in P301S mice

    Neuroinflammation and microglial activation are significant processes in Alzheimer’s disease pathology. Recent genome-wide association studies have highlighted multiple immune-related genes in association with Alzheimer’s disease, and experimental data have demonstrated microglial proliferation as a significant component of the neuropathology. In this study, we tested the efficacy of the selective CSF1R inhibitor JNJ-40346527 (JNJ-527) in the P301S mouse tauopathy model. We first demonstrated the anti-proliferative effects of JNJ-527 on microglia in the ME7 prion model, and its impact on the inflammatory profile, and provided potential CNS biomarkers for clinical investigation with the compound, including pharmacokinetic/pharmacodynamics and efficacy assessment by TSPO autoradiography and CSF proteomics. Then, we showed for the first time that blockade of microglial proliferation and modification of microglial phenotype leads to an attenuation of tau-induced neurodegeneration and results in functional improvement in P301S mice. Overall, this work strongly supports the potential for inhibition of CSF1R as a target for the treatment of Alzheimer’s disease and other tau-mediated neurodegenerative diseases
