    Electronic structure of intentionally disordered AlAs/GaAs superlattices

    We use realistic pseudopotentials and a plane-wave basis to study the electronic structure of non-periodic, three-dimensional, 2000-atom (AlAs)_n/(GaAs)_m (001) superlattices, where the individual layer thicknesses n,m = {1,2,3} are randomly selected. We find that while the band gap of the equivalent (n = m = 2) ordered superlattice is indirect, random fluctuations in layer thicknesses lead to a direct gap in the planar Brillouin zone, strong wavefunction localization along the growth direction, short radiative lifetimes, and a significant band-gap reduction, in agreement with experiments on such intentionally grown disordered superlattices.Comment: 10 pages, REVTeX and EPSF macros, 4 figures in postscript. e-mail to [email protected]

    Vascular Injury After Whole Thoracic X-Ray Irradiation in the Rat

    Purpose To study vascular injury after whole thoracic irradiation with single sublethal doses of X-rays in the rat and to develop markers that might predict the severity of injury. Methods and Materials Rats that received 5- or 10-Gy thorax-only irradiation and age-matched controls were studied at 3 days, 2 weeks, and 1, 2, 5, and 12 months. Several pulmonary vascular parameters were evaluated, including hemodynamics, vessel density, total lung angiotensin-converting enzyme activity, and right ventricular hypertrophy. Results By 1 month, the rats in the 10-Gy group had pulmonary vascular dropout, right ventricular hypertrophy, increased pulmonary vascular resistance, increased dry lung weights, and decreases in total lung angiotensin-converting enzyme activity, as well as pulmonary artery distensibility. In contrast, irradiation with 5 Gy resulted in only a modest increase in right ventricular weight and a reduction in lung angiotensin-converting enzyme activity. Conclusion In a previous investigation using the same model, we observed that recovery from radiation-induced attenuation of pulmonary vascular reactivity occurred. In the present study, we report that deterioration results in several vascular parameters for ≀1 year after 10 Gy, suggesting sustained remodeling of the pulmonary vasculature. Our data support clinically relevant injuries that appear in a time- and dose-related manner after exposure to relatively low radiation doses

    Mutation screening of the medium-chain acyl-CoA dehydrogenase (MCAD) and the ornithine transcarbamylase (OTC) genes by multiplex PCR amplification and sequencing

    Background: Sequencing based mutation screening assays of genes encompassing large numbers of exons could be substantially optimized by multiplex PCR, which enables simultaneous amplification of many targets in one reaction. In the present study, a multiplex PCR protocol originally developed for fragment analysis was evaluated for sequencing based mutation screening of the ornithine transcarbamylase (OTC) and the medium-chain acyl-CoA dehydrogenase (MCAD) genes. Methods: Single exon and multiplex PCR protocols were applied to generate PCR templates for subsequent DNA sequencing of all exons of the OTC and the MCAD genes. For each PCR protocol and using the same DNA samples, 66 OTC and 98 MCAD sequence reads were generated. The sequences derived from the two different PCR methods were compared at the level of individual signal-to-noise ratios of the four bases and the proportion of high-quality base-signals. Results: The single exon and the multiplex PCR protocol gave qualitatively comparable results for the two genes. Conclusions: Many existing sequencing based mutation analysis protocols may be easily optimized with the proposed method, since the multiplex PCR protocol was successfully applied without any re-design of the PCR primers and other optimization steps for generating sequencing templates for the OTC and MCAD genes, respectively.     Grass-clover leys are an integral part of organic rotations. We performed an experiment with reduced tillage (RT) and conventional tillage (CT) using mouldboard ploughing in a rotation in Frick (Switzerland) on a heavy soil and 1000 mm mean annual precipitation. The grass-clover mixture was sawn in autumn 2005 after uniform seed bed preparation with a rotary hoe in both tillage systems without ploughing. After emergence most of the clover seedlings collapsed in the CT plots due to draught, while they survived in the RT plots. This led to a much higher share of clover in the mixture under RT. Grass-clover yields were 29 and 23% higher in RT than in CT plots in the first and second year of cultivation in 2006 and 2007, respectively. Grass grown in RT plots was higher in nitrogen (N), phosphorous (P), potassium (K) and magnesium (Mg) content than in CT plots; clover contained solely more P in RT plots. Over all grass-clover had better growing conditions in RT compared to CT plots in our experiment, reflecting after-effects of the differentiated tillage schemes applied for the preceding arable crops. It is suggested that reduced tillage has a high potential to improve water stress tolerance of cropping systems

    Long-term organic matter application reduces cadmium but not zinc concentrations in wheat

    Wheat is a staple food crop and a major source of both the essential micronutrient zinc (Zn) and the toxic heavy metal cadmium (Cd) for humans. Since Zn and Cd are chemically similar, increasing Zn concentrations in wheat grains (biofortification), while preventing Cd accumulation, is an agronomic challenge. We used two Swiss agricultural long-term field trials, the “Dynamic-Organic-Conventional System Comparison Trial” (DOK) and the “Zurich Organic Fertilization Experiment” (ZOFE), to investigate the impact of long-term organic, mineral and combined fertilizer inputs on total and phytoavailable concentrations of soil Zn and Cd and their accumulation in winter wheat ( L.). “Diffusive gradients in thin films” (DGT) and diethylene-triaminepentaacetic acid (DTPA) extraction were used as proxies for plant available soil metals. Compared to unfertilized controls, long-term organic fertilization with composted manure or green waste compost led to higher soil organic carbon, cation exchange capacity and pH, while DGT-available Zn and Cd concentrations were reduced. The DGT method was a strong predictor of shoot and grain Cd, but not Zn concentrations. Shoot and grain Zn concentrations correlated with DTPA-extractable and total soil Zn concentrations in the ZOFE, but not the DOK trial. Long-term compost fertilization led to lower accumulation of Cd in wheat grains, but did not affect grain Zn. Therefore, Zn/Cd ratios in the grains increased. High Zn and Cd inputs with organic fertilizers and high Cd inputs with phosphate fertilizers led to positive Zn and Cd mass balances when taking into account atmospheric deposition and fertilizer inputs. On the other hand, mineral fertilization led to the depletion of soil Zn due to higher yields and thus higher Zn exports than under organic management. The study supports the use of organic fertilizers for reducing Cd concentrations of wheat grains in the long-term, given that the quality of the fertilizers is guaranteed
