Isotopic Signature of Massive, Buried Ice in Eastern Taylor Valley, Antarctica: Implications for Its Origin
The coastal regions of the McMurdo Dry Valleys, Antarctica, contain deposits of the Ross Sea Drift, sedimentary material left from the Ross Sea ice sheet from the advance of the West Antarctic ice sheet during the Last Glacial Maximum. Much of this deposit is ice-cored, but data on the stable isotopic composition of water from this ice, which may contain a valuable climate archive, are sparse or incomplete. Widespread thermokarstic ground subsidence in this “coastal thaw zone” of the McMurdo Dry Valleys suggests that these potential records are rapidly being lost due to the melting of ground ice and permafrost. We collected samples of massive buried ice from the Ross Sea Drift in eastern Taylor Valley for δ18O-H2O and δ2H-H2O and measured a broad range of values (δ18O = −27.7 to −37.3 ‰; δ2H = −210 to −295 ‰). These buried ice deposits do not show evidence of alteration through sublimation or evaporation, plot along the local meteoric water line, and have values that indicate ice deposition under a colder climate than present conditions. We propose that this ice was sourced from the Ross Sea ice sheet during the Last Glacial Maximum and contains a valuable and accessible climate record
Season of Prescribed Fire Determines Grassland Restoration Outcomes After Fire Exclusion and Overgrazing
Fire exclusion and mismanaged grazing are globally important drivers of environmental change in mesic C4 grasslands and savannas. Although interest is growing in prescribed fire for grassland restoration, we have little long-term experimental evidence of the influence of burn season on the recovery of herbaceous plant communities, encroachment by trees and shrubs, and invasion by exotic grasses. We conducted a prescribed fire experiment (seven burns between 2001 and 2019) in historically fire-excluded and overgrazed grasslands of central Texas. Sites were assigned to one of four experimental treatments: summer burns (warm season, lightning season), fall burns (early cool season), winter burns (late cool season), or unburned (fire exclusion). To assess restoration outcomes of the experiment, in 2019, we identified old-growth grasslands to serve as reference sites. Herbaceous-layer plant communities in all experimental sites were compositionally and functionally distinct from old-growth grasslands, with little recovery of perennial C4 grasses and long-lived forbs. Unburned sites were characterized by several species of tree, shrub, and vine; summer sites were characterized by certain C3 grasses and forbs; and fall and winter sites were intermediate in composition to the unburned and summer sites. Despite compositional differences, all treatments had comparable plot-level plant species richness (range 89–95 species/1000 m2). At the local-scale, summer sites (23 species/m2) and old-growth grasslands (20 species/m2) supported greater richness than unburned sites (15 species/m2), but did not differ significantly from fall or winter sites. Among fire treatments, summer and winter burns most consistently produced the vegetation structure of old-growth grasslands (e.g., mean woody canopy cover of 9%). But whereas winter burns promoted the invasive grass Bothriochloa ischaemum by maintaining areas with low canopy cover, summer burns simultaneously limited woody encroachment and controlled B. ischaemum invasion. Our results support a growing body of literature that shows that prescribed fire alone, without the introduction of plant propagules, cannot necessarily restore old-growth grassland community composition. Nonetheless, this long-term experiment demonstrates that prescribed burns implemented in the summer can benefit restoration by preventing woody encroachment while also controlling an invasive grass. We suggest that fire season deserves greater attention in grassland restoration planning and ecological research

Garwood Valley, Antarctica: A new record of Last Glacial Maximum to Holocene glaciofluvial processes in the McMurdo Dry Valleys
We document the age and extent of late Quaternary glaciofluvial processes in Garwood Valley, McMurdo Dry Valleys, Antarctica, using mapping, stratigraphy, geochronology, and geochemical analysis of sedimentary and ice deposits. Geomorphic and stratigraphic evidence indicates damming of the valley at its Ross Sea outlet by the expanded Ross Sea ice sheet during the Last Glacial Maximum. Damming resulted in development of a proglacial lake in Garwood Valley that persisted from late Pleistocene to mid-Holocene time, and in the formation of a multilevel delta complex that overlies intact, supraglacial till and buried glacier ice detached from the Ross Sea ice sheet. Radiocarbon dating of delta deposits and inferred relationships between paleolake level and Ross Sea ice sheet grounding line positions indicate that the Ross Sea ice sheet advanced north of Garwood Valley at ca. 21.5 ka and retreated south of the valley between 7.3 and 5.5 ka. Buried ice remaining in Garwood Valley has a similar geochemical fingerprint to grounded Ross Sea ice sheet material elsewhere in the southern Dry Valleys. The sedimentary sequence in Garwood Valley preserves evidence of glaciofluvial interactions and climate-driven hydrological activity from the end of the Pleistocene through the mid-Holocene, making it an unusually complete record of climate activity and paleoenvironmental conditions from the terrestrial Antarctic
Integrated analyses of the microbiological, immunological and ontological transitions in the calf ileum during early life
peer reviewedAberdeen Angus calves were sacrificed from immediately post-birth up to 96 days of age (DOA) and ileal samples were collected for microbial, histological and immunological analyses. Firmicutes bacteria were established immediately in the ileum of calves after birth and remained the dominant phyla at all time points from birth until 96 DOA. Temporal shifts in phyla reflected significantly increased Bacteroidetes at birth followed by temporal increases in Actinobacteria abundance over time. At a cellular level, a significant increase in cell density was detected in the ileal villi over time. The innate cell compartment at birth was composed primarily of eosinophils and macrophages with a low proportion of adaptive T lymphocytes; whereas an increase in the relative abundance of T cells (including those in the intra-epithelial layer) was observed over time. The ileal intestinal cells were immunologically competent as assessed by expression levels of genes encoding the inflammasome sensor NLRP3, and inflammatory cytokines IL1A, IL1B and IL33—all of which significantly increased from birth. In contrast, a temporal reduction in genes encoding anti-inflammatory cytokine IL10 was detected from birth. A preliminary study of genetic factors that influence susceptibility to bovine tuberculosis in the British cattle herd
Associations between specific host genes and susceptibility to Mycobacterial infections such as tuberculosis have been reported in several species. Bovine tuberculosis (bTB) impacts greatly the UK cattle industry, yet genetic predispositions have yet to be identified. We therefore used a candidate gene approach to study 384 cattle of which 160 had reacted positively to an antigenic skin test (‘reactors’). Our approach was unusual in that it used microsatellite markers, embraced high breed diversity and focused particularly on detecting genes showing heterozygote advantage, a mode of action often overlooked in SNP-based studies. A panel of neutral markers was used to control for population substructure and using a general linear model-based approach we were also able to control for age. We found that substructure was surprisingly weak and identified two genomic regions that were strongly associated with reactor status, identified by markers INRA111 and BMS2753. In general the strength of association detected tended to vary depending on whether age was included in the model. At INRA111 a single genotype appears strongly protective with an overall odds ratio of 2.2, the effect being consistent across nine diverse breeds. Our results suggest that breeding strategies could be devised that would appreciably increase genetic resistance of cattle to bTB (strictly, reduce the frequency of incidence of reactors) with implications for the current debate concerning badger-culling
Kinetics of ATP release following compression injury of a peripheral nerve trunk
Compression and/or contusion of a peripheral nerve trunk can result in painful sensations. It is possible that release of ATP into the extracellular space may contribute to this symptom. In the present study, we used real-time measurements of ATP-induced bioluminescence together with electrophysiological recordings of compound action potentials to follow changes in the extracellular ATP concentration of isolated rat spinal roots exposed to mechanical stimuli. Nerve compression for about 8 s resulted in an immediate release of ATP into the extracellular space and in a decrease in the amplitude of compound action potentials. On average, a rise in ATP to 60 nM was observed when nerve compression blocked 50% of the myelinated axons. After the compression, the extracellular concentration of ATP returned to the resting level within a few minutes. The importance of ecto-nucleotidases for the recovery period was determined by exposure of isolated spinal roots to high concentrations of ATP and by use of inhibitors of ecto-nucleotidases. It was observed that spinal roots have a high capacity for ATP hydrolysis which is only partially blocked by βγ-methylene ATP and ARL 67156. In conclusion, acute nerve compression produces an increase in the extracellular concentration of ATP and of its metabolites which may be sufficient for activation of purinergic P2 and/or P1 receptors on axons of nociceptive afferent neurons

International meta-analysis of PTSD genome-wide association studies identifies sex- and ancestry-specific genetic risk loci.
The risk of posttraumatic stress disorder (PTSD) following trauma is heritable, but robust common variants have yet to be identified. In a multi-ethnic cohort including over 30,000 PTSD cases and 170,000 controls we conduct a genome-wide association study of PTSD. We demonstrate SNP-based heritability estimates of 5-20%, varying by sex. Three genome-wide significant loci are identified, 2 in European and 1 in African-ancestry analyses. Analyses stratified by sex implicate 3 additional loci in men. Along with other novel genes and non-coding RNAs, a Parkinson's disease gene involved in dopamine regulation, PARK2, is associated with PTSD. Finally, we demonstrate that polygenic risk for PTSD is significantly predictive of re-experiencing symptoms in the Million Veteran Program dataset, although specific loci did not replicate. These results demonstrate the role of genetic variation in the biology of risk for PTSD and highlight the necessity of conducting sex-stratified analyses and expanding GWAS beyond European ancestry populations

El objetivo principal del presente trabajo fue efectuar un análisis de la relación que tiene la satisfacción global que puede obtener un consumidor con los factores que considera importantes en los establecimientos de comida rápida del centro comercial real plaza de Chiclayo. El análisis de datos se hizo sobre una muestra de 196 clientes encuestados que visitan el centro comercial real plaza de la ciudad de Chiclayo. El análisis puso de manifiesto que las dimensiones que guardaban un alto grado de correlación con la satisfacción percibida fueron servicios y conveniencia y calidad e imagen, mientras que el valor económico de la compra no mostró una correlación significativa. Adicionalmente se halló un alto grado de insatisfacción con respecto a los precios y promociones ofrecidas, lo que supone un análisis posterior para comprender mejor estos hallazgos.Tesi

A systematic approach to mapping recessive disease genes in individuals from outbred populations
The identification of recessive disease-causing genes by homozygosity mapping is often restricted by lack of suitable consanguineous families. To overcome these limitations, we apply homozygosity mapping to single affected individuals from outbred populations. In 72 individuals of 54 kindred ascertained worldwide with known homozygous mutations in 13 different recessive disease genes, we performed total genome homozygosity mapping using 250,000 SNP arrays. Likelihood ratio Z-scores (ZLR) were plotted across the genome to detect ZLR peaks that reflect segments of homozygosity by descent, which may harbor the mutated gene. In 93% of cases, the causative gene was positioned within a consistent ZLR peak of homozygosity. The number of peaks reflected the degree of inbreeding. We demonstrate that disease-causing homozygous mutations can be detected in single cases from outbred populations within a single ZLR peak of homozygosity as short as 2 Mb, containing an average of only 16 candidate genes. As many specialty clinics have access to cohorts of individuals from outbred populations, and as our approach will result in smaller genetic candidate regions, the new strategy of homozygosity mapping in single outbred individuals will strongly accelerate the discovery of novel recessive disease genes
