    Cre-Dependent Optogenetic Transgenic Mice Without Early Age-Related Hearing Loss

    With the advent of recent genetic technologies for mice, it is now feasible to investigate the circuit mechanisms of brain functions in an unprecedented manner. Although transgenic mice are commonly used on C57BL/6J (C57) background, hearing research has typically relied on different genetic backgrounds, such as CBA/Ca or CBA due to the genetic defect of C57 mice for early age-related hearing loss. This limits the utilization of available genetic resources for hearing research. Here we report congenic (>F10) Cre-dependent channelrhodopsin2 (ChR2) mice on CBA/Ca background. By crossing this line with Cre-driver mice on C57 background, F1 hybrids restored the hearing deficit of C57 mice. We also found a linear relationship between aging and hearing loss, with progression rates varied depending on genetic backgrounds (3.39 dB/month for C57; 0.82 dB/month for F1 hybrid). We further demonstrate that this approach allows to express ChR2 in a specific type of inhibitory neurons in the auditory cortex and that they can be identified within a simultaneously recorded population of neurons in awake mice. Thus, our Cre-dependent optogenetic transgenic mice on CBA/Ca background are a valuable tool to investigate the circuit mechanisms of hearing across lifespan

The legal status of servants andworkers on Danish estates in the 19th centuryThe focus of this article is the legal relation between landlords and their servants and rural workers on Danish estates in the 19th Century. It examines legal practice at the local court in Rougsø, Sønderhald and Øster Lisbjerg districts in the years 1830-35 and 1870-73 and shows how conflicts between landlords and servants or workers were handled in court.It pays special attention to legal practice in cases where the courts operated in household conlicts that had traditionally been a private matter between master and servant. It examines the specific areas where legislation entitled landlords to impose sanctions on their servants and workers, such as the right of chastisement and the master’s right to dismiss his servants without notice. The article argues that a central feature in these cases was that legislation demanded that servants and workers showed their landlords unspecified obedience. It shows how local courts in the 19th century protected the remains of the traditional inequality between landlords and the rural population.

    SIMPLE SUMMARY: This study evaluates the use of acoustic devices as a method to monitor wolves by analyzing different variables extracted from wolf howls. By analyzing the wolf howls, we focused on identifying individual wolves, subspecies. We analyzed 170 howls from 16 individuals from the three subspecies: Arctic wolves (Canis lupus arctos), Eurasian wolves (C.l. lupus), and Northwestern wolves (C.l. occidentalis). We assessed the potential for individual recognition and recognition of three subspecies: Arctic, Eurasian, and Northwestern wolves. ABSTRACT: Wolves (Canis lupus) are generally monitored by visual observations, camera traps, and DNA traces. In this study, we evaluated acoustic monitoring of wolf howls as a method for monitoring wolves, which may permit detection of wolves across longer distances than that permitted by camera traps. We analyzed acoustic data of wolves’ howls collected from both wild and captive ones. The analysis focused on individual and subspecies recognition. Furthermore, we aimed to determine the usefulness of acoustic monitoring in the field given the limited data for Eurasian wolves. We analyzed 170 howls from 16 individual wolves from 3 subspecies: Arctic (Canis lupus arctos), Eurasian (C. l. lupus), and Northwestern wolves (C. l. occidentalis). Variables from the fundamental frequency (f0) (lowest frequency band of a sound signal) were extracted and used in discriminant analysis, classification matrix, and pairwise post-hoc Hotelling test. The results indicated that Arctic and Eurasian wolves had subspecies identifiable calls, while Northwestern wolves did not, though this sample size was small. Identification on an individual level was successful for all subspecies. Individuals were correctly classified with 80%–100% accuracy, using discriminant function analysis. Our findings suggest acoustic monitoring could be a valuable and cost-effective tool that complements camera traps, by improving long-distance detection of wolves

    Influence of beam pruning techniques on LET and RBE in proton arc therapy

    Introduction: Proton arc therapy (PAT) is an emerging treatment modality that holds promise to improve target volume coverage and reduce linear energy transfer (LET) in organs at risk. We aimed to investigate if pruning the highest energy layers in each beam direction could increase the LET in the target and reduce LET in tissue and organs at risk (OAR) surrounding the target volume, thus reducing the relative biological effectiveness (RBE)-weighted dose and sparing healthy tissue. Methods: PAT plans for a germinoma, an ependymoma and a rhabdomyosarcoma patient were created in the Eclipse treatment planning system with a prescribed dose of 54 Gy(RBE) using a constant RBE of 1.1 (RBE1.1). The PAT plans was pruned for high energy spots, creating several PAT plans with different amounts of pruning while maintaining tumor coverage, denoted PX-PAT plans, where X represents the amount of pruning. All plans were recalculated in the FLUKA Monte Carlo software, and the LET, physical dose, and variable RBE-weighted dose from the phenomenological Rørvik (ROR) model and an LET weighted dose (LWD) model were evaluated. Results and discussion: For the germinoma case, all plans but the P6-PAT reduced the mean RBE-weighted dose to the surrounding healthy tissue compared to the PAT plan. The LET was increasingly higher within the PTV for each pruning iteration, where the mean LET from the P6-PAT plan was 1.5 keV/μm higher than for the PAT plan, while the P4- and P5-PAT plans provided an increase of 0.4 and 0.7 keV/μm, respectively. The other plans increased the LET by a smaller margin compared to the PAT plan. Likewise, the LET values to the healthy tissue were reduced for each degree of pruning. Similar results were found for the ependymoma and the rhabdomyosarcoma case. We demonstrated a PAT pruning technique that can increase both LET and RBE in the target volume and at the same time decreased values in healthy tissue, without affecting the target volume dose coverage.publishedVersio

    Non-canonical role for Lpar1-EGFP subplate neurons in early postnatal mouse somatosensory cortex

    Subplate neurons (SPNs) are thought to play a role in nascent sensory processing in neocortex. To better understand how heterogeneity within this population relates to emergent function, we investigated the synaptic connectivity of Lpar1-EGFP SPNs through the first postnatal week in whisker somatosensory cortex (S1BF). These SPNs comprise of two morphological subtypes: fusiform SPNs with local axons and pyramidal SPNs with axons that extend through the marginal zone. The former receive translaminar synaptic input up until the emergence of the whisker barrels, a timepoint coincident with significant cell death. In contrast, pyramidal SPNs receive local input from the subplate at early ages but then – during the later time window – acquire input from overlying cortex. Combined electrical and optogenetic activation of thalamic afferents identified that Lpar1-EGFP SPNs receive sparse thalamic innervation. These data reveal components of the postnatal network that interpret sparse thalamic input to direct the emergent columnar structure of S1BF
