    Isolation of Nuclei from Physarum flavicomum: Demonstration of Nuclear Cyclic Acid AMP Phosphodiesterase

    Cyclic AMP phosphodiesterase activity in the nucleus of the myxomycete Physarum flavicomum was demonstrated by cytochemical staining utilizing electron microscopy and by enzymatic assays with tritiated cyclic AMP as the substrate. Cytochemical staining showed Physarum\u27s plasmodial phosphodiesterase activity to be located in the nucleus, along the plasma membrane, in vesicles, and free in the cytoplasm. Nuclear phosphodiesterase, which may be cell cycle dependent, was primarily located in the nucleolus. Nuclei from three to five day old microplasmodial cultures were isolated by the method of Henney and Yee. Whole cells were collected through centrifugation and washed. Pellets were homogenized in a medium composed of 0.01 MTris-HC1 (pH 7.2 at 4 °C), 0.25 M sucrose, 0.01% Triton X-100, and 5mM CaC1₂. Nuclei were collected through double filtration and two 1.0 M sucrose density gradient centrifugations. After the nuclei were washed, microscopic examination revealed a purity of over 90%. Radioactive assays of the nuclear preparations demonstrated phosphodiesterase activity consistant with that indicated by cytochemical localization. The specific activity of the nuclear enzyme was 15 nMole of cyclic AMP hydrolyzed /min/mg. of protein

    The effect of caffeine mouth rinse on self-paced cycling performance

    The aim of the study was to determine whether caffeine mouth rinse would improve 30 min self-paced cycling trial. Twelve healthy active males (age 20.5±0.7 years, mass 87.4±18.3 kg) volunteered for the study. They attended the laboratory on 3 separate occasions performing a 30 min self-paced cycling trial. On one occasion water was given as a mouth rinse for 5 s (PLA), on another occasion a 6.4% maltodextrin (CHO) solution was given for 5 s and finally a caffeine solution (containing 32 mg of caffeine dissolved in 125 ml water; CAF) was given for 5 s. Distance cycled, heart rate, ratings of perceived exertion, cadence, speed and power output were recorded throughout all trials. Distance cycled during the CAF mouth rinse trial (16.2±2.8 km) was significantly greater compared to PLA trial (14.9±2.6 km). There was no difference between CHO and CAF trials (P=0.89). Cadence, power and velocity were significantly greater during the CAF trial compared to both PLA and CHO (P0.05).     Seawater cadmium in the Florida Straits over the Holocene and implications for Upper AMOC variability

Over the next century, AMOC is projected to weaken due to greenhouse gas warming, though projecting its future behavior is dependent on a better understanding of how AMOC changes are forced. Seeking to resolve an apparent contradiction of AMOC trends from paleorecords of the more recent past, we reconstruct seawater cadmium, a nutrient-like tracer, in the Florida Straits over the last ∼8,000 years, with emphasis on the last millennium. The gradual reduction in seawater Cd over the last 8,000 years could be due to a reduction in AMOC, consistent with cooling Northern Hemisphere temperatures and a southward shift of the Intertropical Convergence Zone. However, it is difficult to reconcile this finding with evidence for an increase in geostrophic flow through the Florida Straits over the same time period. We combine data from intermediate water depth sediment cores to extend this record into the Common Era at sufficient resolution to address the broad scale changes of this time period. There is a small decline in the Cd concentration in the Late Little Ice Age relative to the Medieval Climate Anomaly, but this change was much smaller than the changes observed over the Holocene and on the deglaciation.     The influence of air-sea exchange on the isotopic composition of oceanic carbon: Observations and modeling

    Although the carbon isotopic composition of ocean waters after they leave the surface ocean is determined by biological cycling, air-sea exchange affects the carbon isotopic composition of surface waters in two ways. The equilibrium fractionation between oceanic and atmospheric carbon increases with decreasing temperature. In Southern Ocean Surface Waters this isotopic equilibration enriches δ13C relative to the δ13C expected from uptake and release of carbon by biological processes alone. Similarly, surface waters in the subtropical gyres are depleted in δ13C due to extensive air-sea exchange at warm temperatures. Countering the tendency toward isotopic equilibration with the atmosphere (a relatively slow process), are the effects of the equilibration of CO2 itself (a much faster process). In regions where there is a net transfer of isotopically light CO2 from the ocean to the atmosphere (e.g., the equator) surface waters become enriched in 13C, whereas in regions where isotopically light CO2 is entering the ocean (e.g., the North Atlantic) surface waters become depleted in 13C. A compilation of high quality oceanic δ13C measurements along with experiments performed using a zonally averaged three-basin dynamic ocean model are used to explore these processes

    Mindfulness meditation targets transdiagnostic symptoms implicated in stress-related disorders: Understanding relationships between changes in mindfulness, sleep quality, and physical symptoms

    Mindfulness-Based Stress Reduction (MBSR) is an 8-week meditation program known to improve anxiety, depression, and psychological well-being. Other health-related effects, such as sleep quality, are less well established, as are the psychological processes associated with therapeutic change. This prospective, observational study (n=213) aimed to determine whether perseverative cognition, indicated by rumination and intrusive thoughts, and emotion regulation, measured by avoidance, thought suppression, emotion suppression, and cognitive reappraisal, partly accounted for the hypothesized relationship between changes in mindfulness and two health-related outcomes: sleep quality and stress-related physical symptoms. As expected, increased mindfulness following the MBSR program was directly correlated with decreased sleep disturbance (r=-0.21, p=0.004) and decreased stress-related physical symptoms (r=-0.38, p<0.001). Partial correlations revealed that pre-post changes in rumination, unwanted intrusive thoughts, thought suppression, experiential avoidance, emotion suppression, and cognitive reappraisal each uniquely accounted for up to 32% of the correlation between the change in mindfulness and change in sleep disturbance and up to 30% of the correlation between the change in mindfulness and change in stress-related physical symptoms. Results suggest that the stress-reducing effects of MBSR are due, in part, to improvements in perseverative cognition and emotion regulation, two “transdiagnostic” mental processes that cut across stress-related disorders

    High-frequency side-scan sonar fish reconnaissance by autonomous underwater vehicles

Moving high-frequency sources to depth using autonomous underwater vehicles (AUVs) mitigates this and also co-locates transducers with other AUV-mounted short-range sensors to allow a holistic approach to ecological surveys. This widely available tool with a pedigree for bottom mapping is not commonly applied to fish reconnaissance and requires the development of an interpretation of pelagic reflective features, revisitation of count methods, image-processing rather than wave-form recognition for automation, and an understanding of bias. In a series of AUV mission test cases, side-scan sonar (600 and 900 kHz) returns often resolved individual school members, spacing, size, behavior, and (infrequently) species from anatomical features and could be intuitively classified by ecologists — but also produced artifacts. Fish often followed the AUV and thus were videographed, but in doing so removed themselves from the sonar aperture. AUV-supported high-frequency side-scan holds particular promise for survey of scarce, large species or for synergistic investigation of predators and their prey because the spatial scale of observations may be similar to those of predators.AUV missions were funded by an Office of Naval Research grant to the Woods Hole Oceanographic Institution and Rutgers University.     Peripheral arterial disease affects ground reaction forces during walking

    Objective: Claudication is the most common manifestation of peripheral arterial disease (PAD), producing significant ambulatory compromise. The gait of claudicating patients has been evaluated using primarily temporal and spatial parameters. With the present study, we used advanced biomechanical measures to further delineate the ambulatory impairment of claudicating patients. We hypothesized that the claudicating legs of PAD patients have an altered kinetic gait pattern compared with normal legs from control subjects. Methods: Ambulation kinetics (ground reaction forces) were evaluated in claudicating patients and compared with age-matched healthy controls. Forces were analyzed in the vertical, anterior–posterior, and medial–lateral directions. Time from heel touch-down to toe-off (stance time) and time spent in double-limb support were also evaluated. Results: The study recruited 14 PAD patients (age, 58 ± 3.4 years; weight, 80.99 ± 15.64 kg) with femoropopliteal occlusive disease (ankle-brachial index [ABI], 0.56 ± 0.03) and five controls (age, 53 ± 3.4 years; weight, 87.38 ± 12.75 kg; ABI, ≥1.00). Vertical force curve evaluation demonstrated significant flattening in claudicating patients resulting in a lower and less fluctuant center of mass when ambulating. In the anterior–posterior direction, claudicating patients demonstrated significantly decreased propulsion forces. In the medial–lateral direction, they had significantly increased forces consistent with wider steps and an inability to swing their legs straight through. Claudicating patients demonstrated a greater stance time and time in double limb support compared with healthy controls. Most importantly, gait abnormalities were present before the onset of claudication, with gait worsening after the onset of claudication. Conclusion: Claudicating patients demonstrate significant gait impairments that are present even before they experience any limb discomfort. These alterations may make them feel more stable and secure while attempting to minimize use of the affected limb. Advanced biomechanical analysis, using ambulation kinetics, permits objective and quantitative evaluation of the gait of claudicating patients. Such evaluation may point to new rehabilitation strategies and provide objective measurement of functional outcomes after medical and surgical therapy
