    SUMMARY Schistosomiasis is a neglected disease with public health importance in tropical and subtropical regions. An alternative to the disease control is the use of molluscicides to eliminate or reduce the intermediate host snail population causing a reduction of transmission in endemic regions. In this study nine extracts from eight Piperaceae species were evaluated against Biomphalaria glabrata embryos at blastula stage. The extracts were evaluated in concentrations ranging from 100 to 10 mg/L. Piper crassinervium and Piper tuberculatum extracts were the most active (100% of mortality at 20 mg/L and 30 mg/L respectively).RESUMO A esquistossomose é uma doença negligenciada de importância para a saúde pública em regiões tropicais e subtropicais. Uma alternativa para o controle da doença é o uso de moluscicidas para eliminar ou reduzir a população de caramujo hospedeiro, acarretando uma redução da transmissão da doença nas regiões endemicas. Neste estudo, nove extratos vegetais provenientes de oito espécies de Piperaceae foram expostos a embriões de Biomphalaria glabrata no estágio de blástula. Os extratos foram avaliados em concentrações que variaram entre 100 e 10 mg/L, sendo Piper crassinervium e Piper tuberculatum os extratos mais ativos (100% de mortalidade a 20 mg/L e 30 mg/L respectivamente)

    Interaction Gardens and Butterfly Catalogues: a Joint Strategy to Promote Capacity Development in Protected Areas and Reduce the Extinction of Experience in Cities

    More than half of the world’s population live in cities. Increasing numbers of generations are now born and raised in urban landscapes with decreasing opportunities towards interacting with natural environments. This extinction of experience leads to environmental apathy and lack of bioliteracy, which is a central aspect to be tackled in conservation strategies. At the same time, people who live near or at non-urban settings are closer to natural habitats but do not have similar access to concepts of environmentalism and may lack incentives to lean towards nature conservation, instead of its exploitation or even illegal activities. We here propose that interaction gardens, that is, gardens with multiple trophic levels (primary producers, herbivores, predators, and parasitoids), can tackle these issues, especially if planned with incentives, such as butterfly catalogues, aimed as a common goal of the garden community. It can also bring benefits to human health and well-being, increase the survival chances of local biodiversity, and strengthen the front line of conservation by promoting income strategies to people who live near protected areas. Our specific aims are to 1) share a case study of capacity development at the Intervales State Park in the Atlantic Forest, Southeast Brazil; 2) present the butterfly catalogue of this protected area; 3) present guidelines for interaction gardens at both urban and non-urban settings; and 4) discuss alternative perspectives about Neotropical conservation. We provide a translated version of the text in Portuguese to encourage students, educators, NGOs and local communities of other protected areas to venture in our proposed joint strategy of interaction gardens with butterfly catalogues

    Plasticity in plant defense and the role of phytochemical dissimilarity in limiting specialist herbivory

    Phytochemical diversity is an effective plant defensive attribute, but much more research has focused on genetic and environmental controls of specific defensive compounds than phytochemical diversity per se. Documenting plasticity in phytochemical richness and plant chemical composition as opposed to individual compounds is important for understanding plant defense. This study outlines a multi-site transplant experiment in Cerrado gallery forests in central Brazil, utilizing Piper arboreum (Piperaceae), a prevalent and widespread neotropical shrub. Clones from four distinct populations were planted either at their origin site or in a different forest. Secondary metabolite composition varied between populations initially and then changed after transplanting. Interestingly, clones with chemical profiles that were distinct from the populations where they were introduced experienced reduced specialist chrysomelid herbivory compared to clones that were more chemically similar to the existing P. arboreum populations where they were planted. Specialist Lepidoptera herbivory also declined in clones transplanted to a new forest, but this change could not be ascribed to chemical profiles. In contrast, generalist herbivory was unaffected by chemical dissimilarity and transplanting. This research adds to the expanding body of evidence suggesting that phytochemical diversity is a dynamic trait exerting unique effects on different herbivore guilds

    A Araucaria angustifolia é uma conífera endêmica das regiões sul e sudeste do Brasil sendo considerada uma espécie em extinção devido ao extenso extrativismo madeireiro. Atualmente, existem inúmeros projetos visando o reflorestamento e o uso sustentável deste pinheiro. Em vista destes pontos, o estudo das propriedades dos componentes das folhas com o intuito da utilização destes com fins comerciais tornou-se de extrema importância. As suas folhas foram submetidas à extração com solventes e foram identificados seis biflavonóides majoritários, dentre estes a amentoflavona e a ginkgetina, que são apontados como agentes contra inflamações e artrites. A fração rica de biflavonóides (BFF) extraída da araucaria foi testada frente a sua atividade em proteger contra danos em biomoléculas provocadas por espécies reativas de oxigênio, capacidade em quelar metais e proteção contra raios UV. A capacidade do BFF em proteger contra danos provocados por espécies reativas de oxigênio foi comparado com compostos conhecidamente antioxidantes, como o α-tocoferol, Trolox®, quercetina, rutina e com padrões de biflavonóides, a amentoflavona e ginkgetina. O BFF demonstrou que possui uma constante de supressão do 1O2 (50 x 106 M-1s-1), superior ao da quercetina (9 x 106 M-1s-1) e foi o mais eficiente na proteção contra quebras de simples fita em DNA plasmidial, provocado por esta espécie reativa. Ainda em relação à proteção de DNA plasmidial o BFF foi capaz de proteger também contra estes danos provocados através da reação de Fenton, apesar de não demonstrar a mesma eficiência da quercetina que mostrou ser um potente protetor destes danos. O BFF protegeu contra lipoperoxidação em lipossomos de fosfatidilcolina induzida por raios UV e reação de Fenton. Em análises realizadas com espectrometria de massas foi observada a formação de complexos destes biflavonóides com íons metálicos como ferro, cobre e alumínio que possuem um papel importante na formação de radicais livres. Em relação à capacidade fotoprotetora do BFF, este inibiu a formação de dímeros de pirimidina que são apontados como causadores de câncer de pele induzidos, principalmente por radiação UV-B. Esta ação protetora foi superior àquela conferida ao p-metoxicinamato de octila, um conhecido fotoprotetor. Com o intuito de permitir a solubilização do BFF em soluções aquosas e assim, avaliar a ação do BFF em células, incorporou-se o BFF em ciclodextrina. Essa inclusão favoreceu a incorporação de BFF em células CV1-P na concentração aproximada de 0,4 µg/ml após 24 horas de incubação. Essa concentração incorporada não demonstrou ser tóxica para as células no teste com MTT. Assim, o BFF tem despertado grande interesse em relação ao seu potencial na utilização nas mais variadas áreas como cosmética, alimentos e fitoterápicos.Araucaria angustifolia is an endemic conifer in southern and southeastern Brazil endangered due the extensive loggings. Its needles contain six major amentoflavone-type biflavonoids, including amentoflavone, ginkgetin and tetra-O-methylamentoflavone, all reported to possess a variety of biological activities such as anti-inflammatory and antiarthritic activities. The organic fraction rich in biflavonoids (BFF) extracted from Araucaria was evaluated regarding its activity to protect against damage to biomolecules promoted by reactive oxygen species, its capacity to chelate ion metals and protection it affords against UV radiation. The ability of the BFF prevent oxidative damages was compared to antioxidants compounds, such as, α-tocopherol, Trolox®, quercetin, rutin and with standards of biflavonoids amentoflavone and ginkgetin. The BFF showed a higher 1O2 quenching rate (50 x 106 M-1s-1) than individual quercetin (9 x 106 M-1s-1). Accordingly, BFF was shown to strongly inhibit plasmid DNA single strand break (ssb) induced by 1O2 generated by NDPO2. On the other hand, BFF did not protect plasmid DNA against ssb triggered by the Fenton reaction as effectively as quercetin and rutin. BFF, quercetin and rutin were able to protect liposomes against peroxidative degradation caused by UV-irradiation. Since there is considerable evidence relating oxygen species with UV phospholipid degradation, the protective effect of quercetin and rutin is likely to result from their well-known scavenging activity against hydroxyl and peroxyl radicals and superoxide anion radicals. Using electrospray ionization mass spectrometry, metal (Fe3+, Cu2+ and Al3+) biflavonoids complexes were also detected and characterized. The sunlight UV region is believed to be largely responsible for the greatest damage to the skin including indution of the pirimidine dimers formation suggested to be implicated in skin cancer. BFF was able to diminish the formation of this photoproduct more efficiently tham octyl methoxycinnamate. Solubilization of BFF in aqueous solutions was performed using cyclodextrin, which allowed the incorporation of 0,4 g/ml of biflavonoids in CV1-P cells after 24 hours. In this conditions the BFF was not cytotoxic to CV1-P evaluated by the MTT assay.Altogether, these properties point BFF as an excellent candidate for successful employment as antioxidant compound in several systems

    The genus Eois Hübner (Geometridae: Larentiinae) comprises 254 valid species, 217 of which were described from the Neotropics and 31 of those having their type locality in Brazil. Since this species rich genus has never been revised, and may potentially include many cryptic undescribed species, Eois embodies a problematic taxonomic scenario. The actual diversity of Eois is greatly underestimated and the Brazilian fauna is poorly known, both because of inadequate sampling and because of the potential existence of cryptic species "hidden" within some nominal taxa. In this study we investigated the diversity within a cryptic species complexes associated to the E. pallidicosta and E. odatis clades. We describe three new species Eois oya Moraes & Montebello sp. nov., Eois ewa Moraes & Stanton sp. nov., and Eois oxum Moraes & Freitas sp. nov., in an integrative taxonomy approach, using morphology, host plant use and species delimitation tools

    Toxoplasmosis is a tropical and neglected disease caused by the parasitic protozoa Toxplasma gondii. Conventional treatment with sulfadiazine and pyrimethamine plus folinic acid, has some drawbacks, such as inefficacy in the chronic phase, toxic side effects, and potential cases of resistance have been observed. In this study, the activity of essential oils (EOs) from three Piper species and their main constituents, including α-Pinene (Piper lindbergii and P. cernuum), β-Pinene (P. cernuum), and dillapiole (P. aduncum), were evaluated against tachyzoites of T. gondii. α-Pinene was more active [(IC50 0.3265 (0.2958 to 0.3604) μg/mL)] against tachyzoites than P. lindbergii EO [0.8387 (0.6492 to 1.084) μg/mL]. Both α-Pinene and P. lindbergii EO exhibited low cytotoxicity against NHDF cells, with CC50 41.37 (37.64 to 45.09) µg/mL and 83.80 (75.42 to 91.34) µg/mL, respectively, suggesting they could be of potential use against toxoplasmosis

    Common bean (Phaseolus vulgaris) is present in the daily diet of various countries and, as for other legumes, has been investigated for its nutraceutical potential. Thus, 16 genotypes from different gene pools, representing seven types of seed coats and different responses to pathogens and pests, were selected to verify their isoflavone contents. The isoflavonoids daidzein and genistein and the flavonols kaempferol, myricetin, and quercetin were found. Grains of the black type showed the highest concentrations of isoflavonoids and were the only ones to exhibit daidzein. IAC Formoso, with high protein content and source of resistance to anthracnose, showed the greatest concentration of genistein, representing around 11% of the content present in soybean, as well as high levels of kaempferol. Arc 1, Raz 55, and IAC Una genotypes showed high content of coumestrol. The results suggest the use of IAC Formoso to increase the nutraceutical characteristics in common bean.629699-70

    Pipa carvalhoi skin secretion profiling: Absence of peptides and identification of kynurenic acid as the major constitutive component

    AbstractThe presence of peptides has been identified in all African pipid genera; nevertheless, little is known about skin secretion of South American frog genus Pipa. Skin secretion from captive and wild Pipa carvalhoi were obtained in the presence or absence of norepinephrine stimulation. The <10kDa fraction was analyzed by liquid chromatography and mass spectrometry, searching for peptides. Chromatographic profiles show the presence of a major component in this secretion, regardless of the stimulation method (norepinephrine or mechanical stimulation) and the origin of the animal (captivity or wild), as well as in the absence of any stimulus. The general mass distribution profile in P. carvalhoi skin secretion shows numerous components below 800Da. Moreover, no peptide could be identified, regardless of the chromatographic approach. The major component was purified and identified as kynurenic acid, an l-tryptophan derivative. P. carvalhoi does not secrete peptides as toxins in its skin. In addition, we here report that kynurenic acid is the main component of P. carvalhoi skin secretion. Although no biological activity was associated with kynurenic acid, we propose that this molecule is a pheromone that signals the presence of a co-specific in the shady environment in which this animal lives.
