    Strangers sharing space and lives : a qualitative study of the attitudes of single elderly persons living in shared accommodation in Hong Kong public housing estates = 「同居共住:長者對合住公共房屋的意見調查硏究」硏究報告

    In response to the perceived housing needs of single elderly people, the Housing for Senior Citizens Scheme has been developed by the Hong Kong Housing Authority since the mid-1980s to provide shared housing for unrelated elderly people. Under this scheme, they live together in an apartment unit with individual bedrooms, but sharing certain facilities. This group is fairly numerous and, in Hong Kong in 1998, there were about 110,000 elderly people living alone for various reasons,among whom 34,000 were tenants in public housing estates. Despite some advantages, shared living for any age group can however lead to potential problems. Using qualitative methodologies including focus groups and in-depth interviews, the research attempted to gain insights into the views of residents on the design and management of HSC projects, and their aspirations. Nine of the 38 HSC projects operating in 32 Hong Kong public housing estates in mid-1998 were studied. A total of 69 elderly participants (78% female, 22% male) attended the nine focus groups, each comprising 6 to 12 residents. Participants\u27 ages ranged from 62 to 90 years and their length of residence in an HSC from three months to 11 years. To residents, positive features of the HSC projects included the provision of shelter in an elderly person\u27s later life, the care and attention of wardens and the installation of emergency alarms. Design and facility improvements in the newer housing estates appeared to serve better the needs of the residents, with enhanced privacy and reduction in shared facilities. However, older blocks still suffered from problems of insufficient maintenance and repair, and perceived high charges. Most residents appeared to have tried hard to adjust to group life but would prefer to have their own kitchen and bathroom. Conflicts sometimes arose from personality clashes, different cleaning and living habits, shared facilities and bills for water and repairs. Nevertheless, there were some residents who enjoyed the informal caring relationships and the feeling of security. Another important feature found with respect to the elderly respondents was their low self-esteem and strong sense of powerlessness derived from their sharing of accommodation. Poor family relationships and previous living arrangements were also evident. The research suggests that integration of a caring role into housing provision is increasingly important. Autonomy in housing choice and in the management of life styles are also crucial given the diversity of personalities, needs and demands of the elderly population. The group living model should not be assumed to be a panacea for all single older persons in housing need

    Investigation of campylobacter concisus gastric epithelial pathogenicity using AGS cells

    Campylobacter concisus is an oral bacterium. Recent studies suggest that C. concisus may be involved in human gastric diseases. The mechanisms, however, by which C. concisus causes human gastric diseases have not been investigated. Here we examined the gastric epithelial pathogenicity of C. concisus using a cell culture model. Six C. concisus strains and the human gastric epithelial cell line AGS cells were used. IL-8 produced by AGS cells after incubation with C. concisus was measured using enzyme-linked immunosorbent assay (ELISA), and AGS cell apoptosis was determined by caspase 3/7 activities. The effects of C. concisus on actin arrangement in AGS cells was determined using fluorescence staining. The effects of C. concisus on global gene expression in AGS cells was determined by transcriptomic analysis and quantitative real-time PCR (qRT-PCR). The role of the upregulated CYP1A1 gene in gastric cancer survival was assessed using the Kaplan-Meier method. C. concisus induced production of IL-8 by AGS cells with strain variation. Significantly increased caspase 3/7 activities were observed in AGS cells incubated with C. concisus strains when compared to AGS cells without bacteria. C. concisus induced actin re-arrangement in AGS cells. C. concisus upregulated 30 genes in AGS cells and the upregulation of CYP1A1 gene was confirmed by qRT-PCR. The Kaplan-Meier analysis showed that upregulation of CYP1A1 gene is associated with worse survival in gastric cancer patients. Our findings suggest that C. concisus may play a role in gastric inflammation and the progression of gastric cancer. Further investigation in clinical studies is warranted

    Identification of Human Papillomavirus Type 58 Lineages and the Distribution Worldwide

    Background. Human papillomavirus type 58 (HPV-58) accounts for a much higher proportion of cervical cancers in East Asia than other types. A classification system of HPV-58, which is essential for molecular epidemiological study, is lacking. Methods and results. This study analyzed the sequences of 401 isolates collected from 15 countries and cities. The 268 unique concatenated E6-E7-E2-E5-L1-LCR sequences that comprised 57% of the whole HPV-58 genome showed 4 distinct clusters. L1 and LCR produced tree topologies that best resembled the concatenated sequences and thus are the most appropriate surrogate regions for lineage classification. Moreover, short fragments from L1 (nucleotides 6014–6539) and LCR (nucleotides 7257–7429 and 7540–52) were found to contain sequence signatures informative for lineage identification. Lineage A was the most prevalent lineage across all regions. Lineage C was more frequent in Africa than elsewhere, whereas lineage D was more prevalent in Africa than in Asia. Among lineage A variants, sublineage A2 dominated in Africa, the Americas, and Europe, but not in Asia. Sublineage A1, which represents the prototype that originated from a patient with cancer, was rare worldwide except in Asia. Conclusions. HPV-58 can be classified into 4 lineages that show some degree of ethnogeographic predilection in distribution. The evolutionary, epidemiological, and pathological characteristics of these lineages warrant further study

    Altered E-cadherin expression and p120 catenin localization in esophageal squamous cell carcinoma

    Background: E-cadherin is a well-known tumor suppressor and its dysregulated expression correlates with tumor differentiation, metastasis and survival in esophageal squamous cell carcinoma (ESCC). p120 catenin is an Armadillo protein normally bound to E-cadherin in the cadherin-catenin complex at the adherens junction. Dysregulated expression and mislocalization of p120ctn affect the protective function of the complex. The objective of the present study was to evaluate the clinical significance of E-cadherin and p120ctn expression in ESCC. Methods: Immunohistochemistry was performed to investigate the expression of E-cadherin and p120ctn proteins in 71 patients with ESCC. The relationships between protein expression and clinicopathological characteristics were analyzed. Results: Reduced E-cadherin and p120ctn expressions were observed in 42.3% and 8.5% of ESCC cases, respectively. Reduction of membranous p120ctn was observed in 33.8% of cases. Membranous E-cadherin was preserved when p120ctn co-localized on the membrane of tumor cells (72.3%, P = 0.001). High level E-cadherin expression and membranous p120ctn preservation positively correlated with tumor differentiation (P = 0.001 and P = 0.008, respectively). p120ctn expression was also significantly related to lymph node metastasis (P = 0.003). Heterogeneous expression of both E-cadherin and p120ctn was observed in dysplasia. Conclusions: Altered E-cadherin expression and p120ctn localization were related to tumor differentiation, indicating their important roles in the pathogenesis of ESCC. © 2007 The Society of Surgical Oncology, Inc.postprin

    The single-cell chromatin accessibility landscape in mouse perinatal testis development

    Spermatogenesis depends on an orchestrated series of developing events in germ cells and full maturation of the somatic microenvironment. To date, the majority of efforts to study cellular heterogeneity in testis has been focused on single-cell gene expression rather than the chromatin landscape shaping gene expression. To advance our understanding of the regulatory programs underlying testicular cell types, we analyzed single-cell chromatin accessibility profiles in more than 25,000 cells from mouse developing testis. We showed that single-cell sequencing assay for transposase-accessible chromatin (scATAC-Seq) allowed us to deconvolve distinct cell populations and identify cis-regulatory elements (CREs) underlying cell-type specification. We identified sets of transcription factors associated with cell type-specific accessibility, revealing novel regulators of cell fate specification and maintenance. Pseudotime reconstruction revealed detailed regulatory dynamics coordinating the sequential developmental progressions of germ cells and somatic cells. This high-resolution dataset also unveiled previously unreported subpopulations within both the Sertoli and Leydig cell groups. Further, we defined candidate target cell types and genes of several genome-wide association study (GWAS) signals, including those associated with testosterone levels and coronary artery disease. Collectively, our data provide a blueprint of the ‘regulon’ of the mouse male germline and supporting somatic cells
