    Anti-Ro52/TRIM21 antibodies are associated with aberrant inflammatory circuits in patients with systemic autoimmune rheumatic diseases

    INTRODUCTION: Anti-Ro52/TRIM21 antibodies are markers for several systemic autoimmune rheumatic diseases (SARD). OBJECTIVE: To assess whether anti-Ro52/TRIM21 antibodies are related to abnormalities in inflammatory circuits. METHODS: Cross-sectional study of consecutive outpatients with SARD. Anti-Ro52/TRIM21 antibodies and serum amyloid A protein were measured by ELISA; panels for 18 cytokines and nine chemokines were analyzed on a Luminex reading platform, while high-sensitivity C-reactive protein (hs-CRP) and complement were measured by nephelometry. RESULTS: Among 167 included patients, 143 had systemic lupus erythematosus (SLE), 16 had primary Sjögren's syndrome and eight had systemic sclerosis; 41 (24%) were positive for anti-Ro52/TRIM21 antibodies. Patients with anti-Ro52/TRIM21 antibodies had higher serum levels of IL-2, IL-4, IL-6, GM-CSF, IL-21, IL-22, hs-CRP and chemokines CCL4, CXCL8, CXCL10 and CXCL12, but lower levels of complement C4. Anti-Ro52/TRIM21 antibody titers were positively correlated with IL-2, IL-4, IL-6, IL-10, IL-21, IL-22, CXCL10, and hs-CRP, and negatively with complements C3 and C4. When only SLE patients were included, no association was identified between anti-Ro52/TRIM21 antibodies and disease activity or organ-specific involvement. CONCLUSIONS: Anti-Ro52/TRIM21 antibodies are associated with aberrant cytokine circuits and elevated levels of angiogenic molecules and neutrophil and monocyte chemoattractants, which suggests an active role for these antibodies in SARD.</p

    Coasting cosmologies with time dependent cosmological constant

    The effect of a time dependent cosmological constant is considered in a family of scalar tensor theories. Friedmann-Robertson-Walker cosmological models for vacumm and perfect fluid matter are found. They have a linear expansion factor, the so called coasting cosmology, the gravitational "constant" decreace inversely with time; this model satisfy the Dirac hipotesis.     Estágio profissional de intervenção em fisioterapia neurológica com pacientes adultos após aciente vascular cerebral

    Mestrado em Fisioterapia NeurológicaIntrodução: A Fisioterapia Neurológica é uma área de intervenção essencial na medida em que existem cada vez mais doentes com sequelas resultantes de lesões do sistema nervoso central a necessitar de cuidados de fisioterapia, nomeadamente doentes com Acidente Vascular Cerebral já que as taxas de incidência são altas e a mortalidade tem vindo a decrescer com os avanços da medicina. Por outro lado, os conhecimentos atuais da área da neurociência permitem que a intervenção do fisioterapeuta seja baseada na evidência. Objetivos: Aprofundar conhecimentos, capacidades e competências em Fisioterapia Neurológica em pacientes adultos; intervindo essencialmente, em pacientes com sequelas de Acidente Vascular Cerebral, desenvolvendo o modelo de raciocínio clínico e investigando a efetividade do conceito de Bobath na funcionalidade de pacientes pós Acidente Vascular Cerebral. Metodologia: O estágio articulou a vertente de investigação, a dimensão pedagógica e a prática clínica; decorreu no Hospital Curry Cabral (Centro Hospitalar Lisboa Central) com pacientes adultos a realizar fisioterapia, em regime de internamento ou ambulatório, devido a patologia neurológica e mais especificamente com pacientes com sequelas de AVC; teve a duração de 200h. Foram selecionadas três escalas de funcionalidade (o Indicador de Barthel Modificado, a Medida de Independência Funcional e a Motor Assessement Scale) para a avaliação dos casos clínicos acompanhados e a abordagem em fisioterapia foi baseada no Conceito de Bobath. Resultados: Este estágio favoreceu a aquisição e consolidação de conhecimentos na área da Fisioterapia Neurológica, nomeadamente sobre o Conceito de Bobath e a avaliação da funcionalidade. São apresentados três casos clínicos de pacientes com sequelas de AVC que permitiram treinar a aplicação das escalas de funcionalidade, desenvolver o raciocínio clínico, aplicar as estratégias selecionadas e refletir sobre a eficácia da intervenção. Discussão: Embora não exista evidência robusta da superioridade do Conceito de Bobath em relação a outras abordagens, a sua filosofia facilita a avaliação e a tomada de decisão numa perspetiva holística sem menosprezar a individualidade de cada paciente. Das três escalas de funcionalidade aplicadas a Motor Assessement Scale é a única a levar em consideração a qualidade do movimento, o que a torna mais sensível a pequenas evoluções facilitando a mensuração adequada dos objetivos da intervenção. Conclusão: A realização deste estágio foi um período de aprendizagem importante e uma experiência enriquecedora no aspeto clínico, científico e académico.ABSTRACT - Introduction: Neurological Physiotherapy is an essential intervention area as there are more and more patients with sequelae resulting from injuries to the central nervous system requiring physiotherapy care, namely patients with stroke as the incidence rates are high and mortality has been decreasing with medical advances. On the other hand, current knowledge in the field of neuroscience allows the intervention of the physiotherapist to be based on evidence Objectives: To deepen knowledge, skills, and competencies in Neurological Physiotherapy in adult patients; essentially intervening in patients with sequelae of stroke, developing the model of clinical reasoning and investigating the effectiveness of the Bobath Concept in the functionality of patients after stroke. Methodology: The internship articulated the research aspect, the pedagogical dimension, and the clinical practice; it took place at Hospital Curry Cabral (Centro Hospitalar Lisboa Central) with adult patients undergoing physical therapy, inpatient or outpatient, due to neurological pathology and more specifically with patients with stroke sequelae; lasted 200 hours. Three scales of functionality were selected (the Barthel modified indicator the Functional Independence Measure and the Motor Assessment Scale) for the evaluation of the clinical cases followed and the approach in physiotherapy was based on the Bobath Concept. Results: This internship allowed the acquisition and consolidation of knowledge in the area of Neurological Physiotherapy, namely on the Bobath Concept and the evaluation of functionality. Three clinical cases of patients with stroke sequelae are presented, which allowed to train the application of the selected functionality scales, develop clinical reasoning and reflect on the effectiveness of the intervention. Discussion: While there is no robust evidence of the superiority of the Bobath Concept over other approaches, its philosophy facilitates assessment and decision-making from a holistic perspective without underestimating the individuality of each patient. Of the three scales of functionality applied the Motor Assessment Scale, is the only one to consider the quality of the movement, which makes it more sensitive to small developments, facilitating the adequate measurement of the intervention goals. Conclusion: This internship was an important learning period and an enriching experience in the clinical, scientific, and academic aspects.N/

    Taxonomic variations in the gut microbiome of gout patients with and without tophi might have a functional impact on urate metabolism

    [Objective] To evaluate the taxonomic composition of the gut microbiome in gout patients with and without tophi formation, and predict bacterial functions that might have an impact on urate metabolism.[Methods] Hypervariable V3–V4 regions of the bacterial 16S rRNA gene from fecal samples of gout patients with and without tophi (n = 33 and n = 25, respectively) were sequenced and compared to fecal samples from 53 healthy controls. We explored predictive functional profiles using bioinformatics in order to identify differences in taxonomy and metabolic pathways. [Results] We identified a microbiome characterized by the lowest richness and a higher abundance of Phascolarctobacterium, Bacteroides, Akkermansia, and Ruminococcus_gnavus_group genera in patients with gout without tophi when compared to controls. The Proteobacteria phylum and the Escherichia-Shigella genus were more abundant in patients with tophaceous gout than in controls. Fold change analysis detected nine genera enriched in healthy controls compared to gout groups (Bifidobacterium, Butyricicoccus, Oscillobacter, Ruminococcaceae_UCG_010, Lachnospiraceae_ND2007_group, Haemophilus, Ruminococcus_1, Clostridium_sensu_stricto_1, and Ruminococcaceae_UGC_013). We found that the core microbiota of both gout groups shared Bacteroides caccae, Bacteroides stercoris ATCC 43183, and Bacteroides coprocola DSM 17136. These bacteria might perform functions linked to one-carbon metabolism, nucleotide binding, amino acid biosynthesis, and purine biosynthesis. Finally, we observed differences in key bacterial enzymes involved in urate synthesis, degradation, and elimination.     Avaliação da incidência de antracnose, do desempenho e estado nutricional de variedades de mangueira, para cultivo orgânico, na região centro-norte do Estado de São Paulo.

    A mudança do perfil do consumidor, aliada aos riscos da contaminação por agrotóxicos, tem levado à busca de alternativas ecologicamente apropriadas para produção de frutas. Os objetivos deste trabalho foram avaliar a incidência de antracnose, o desempenho e estado nutricional de variedades de mangueira conduzidas organicamente na região de Pindorama-SP. Foram utilizadas 17 variedades de mangueira. O experimento foi instalado em delineamento experimental em blocos completos ao acaso, com 17 tratamentos (variedades) e seis repetições. Foi avaliada a severidade de antracnose nas folhas, através de uma escala diagramática, atribuindo-se notas aos sintomas. Foram avaliados o crescimento e o desenvolvimento (altura da planta, perímetro do tronco e da copa) e o estado nutricional, mediante análise foliar, das diferentes variedades utilizadas. Através dos resultados obtidos, podem-se considerar como muito suscetíveis à antracnose as variedades Bourbon, Rocha e Rosa; e resistentes, as variedades IAC 111, Alfa, Beta e Parvin; as variedades de manga apresentaram o mesmo padrão de crescimento; as maiores alturas da planta corresponderam aos maiores diâmetros do tronco e da copa; a variedade Parvin apresentou o melhor desempenho dentre as variedades estudadas, com relação à resistência à antracnose, altura e diâmetro do caule e da copa, podendo ser recomendada ao cultivo orgânico. As variedades Omega e Alfa também apresentaram bom crescimento, podendo ser indicadas para esse cultivo, pelo menos nessa fase inicial; as variedades Surpresa e Rosa não apresentaram bom desempenho, no campo, em relação às demais, não devendo ser recomendadas para o cultivo orgânico, principalmente a variedade Rosa, bastante suscetível à antracnose. As concentrações de N, P e K foram elevadas na fase vegetativa das plantas, comparadas à baixa concentração de Ca; houve carência de Boro em todas as variedades estudadas. A manga Rosa, provavelmente, sofreu toxicidade ao excesso de manganês, ocasionando diminuição em seu desenvolvimento

    A silviculture-oriented spatio-temporal model for germination in Pinus pinea L. in the Spanish Northern Plateau based on a direct seeding experiment

    Natural regeneration in Pinus pinea stands commonly fails throughout the Spanish Northern Plateau under current intensive regeneration treatments. As a result, extensive direct seeding is commonly conducted to guarantee regeneration occurrence. In a period of rationalization of the resources devoted to forest management, this kind of techniques may become unaffordable. Given that the climatic and stand factors driving germination remain unknown, tools are required to understand the process and temper the use of direct seeding. In this study, the spatio-temporal pattern of germination of P. pinea was modelled with those purposes. The resulting findings will allow us to (1) determine the main ecological variables involved in germination in the species and (2) infer adequate silvicultural alternatives. The modelling approach focuses on covariates which are readily available to forest managers. A two-step nonlinear mixed model was fitted to predict germination occurrence and abundance in P. pinea under varying climatic, environmental and stand conditions, based on a germination data set covering a 5-year period. The results obtained reveal that the process is primarily driven by climate variables. Favourable conditions for germination commonly occur in fall although the optimum window is often narrow and may not occur at all in some years. At spatial level, it would appear that germination is facilitated by high stand densities, suggesting that current felling intensity should be reduced. In accordance with other studies on P. pinea dispersal, it seems that denser stands during the regeneration period will reduce the present dependence on direct seeding
