    Strategic knowledge for the identification and construction of goal states

    There is widespread recognition that goal recognition strategies, in the context of structural analysis and cognitive (user) models, represent a major field of contemporary research into discourse understanding. This thesis reports a goal interpretation paradigm that embraces both a novel goal structure formalism and strategic knowledge. The goal interpretation processes involve the identification of goal primitives and the construction of goal states. The mechanisms developed for goal interpretation rely on explicit goal recognition (selection) and confirmation of feasibility. A goal state contains all the information required by the planner. By constructing a goal state, the chance of failure in planning is greatly reduced and the efficiency of the planning system is vastly improved. These mechanisms are not limited to inference. Other mechanisms are reported include goal structure processing, goal primitives identification and searching strategies, extended heuristic classification method and a new conceptual graph operation (i.e. SPLIT)

    Genomic and Transcriptomic Alterations Associated with STAT3 Activation in Head and Neck Cancer.

    BackgroundHyperactivation of STAT3 via constitutive phosphorylation of tyrosine 705 (Y705) is common in most human cancers, including head and neck squamous carcinoma (HNSCC). STAT3 is rarely mutated in cancer and the (epi)genetic alterations that lead to STAT3 activation are incompletely understood. Here we used an unbiased approach to identify genomic and epigenomic changes associated with pSTAT3(Y705) expression using data generated by The Cancer Genome Atlas (TCGA).Methods and findingsMutation, mRNA expression, promoter methylation, and copy number alteration data were extracted from TCGA and examined in the context of pSTAT3(Y705) protein expression. mRNA expression levels of 1279 genes were found to be associated with pSTAT3(705) expression. Association of pSTAT3(Y705) expression with caspase-8 mRNA expression was validated by immunoblot analysis in HNSCC cells. Mutation, promoter hypermethylation, and copy number alteration of any gene were not significantly associated with increased pSTAT3(Y705) protein expression.ConclusionsThese cumulative results suggest that unbiased approaches may be useful in identifying the molecular underpinnings of oncogenic signaling, including STAT3 activation, in HNSCC. Larger datasets will likely be necessary to elucidate signaling consequences of infrequent alterations

    A simple decision market model

    Economic modeling of decision markets has mainly considered the market scoring rule setup. Literature has made reference to the alternative, joint elicitation type decision market, but no in depth analysis of it appears to have been published. This paper develops a simple decision market model of the joint elicitation type, that provides a specific decision market nomenclature on which to base future analysis.A generally accepted prediction market model is modified, by introducing two additional concepts: “proper information market” and “relevant information”. Our work then provides original contributions to the theoretical discourse on information markets, including finding the sufficient and necessary condition for convergence to the best possible prediction. It is shown in our new prediction market model that “all agents express relevant information” is a sufficient and necessary condition for convergence to the direct communication equilibrium in a proper information (prediction) market.Our new prediction market model is used to formulate a simple decision market model of the joint elicitation market type. It is shown that our decision market will select the best decision if a specific selection and payout rule is defined. Importantly, our decision market model does not need to delay payment of any contracts to the observation of the desired outcome. Therefore, when dealing with long-term outcome projects, our decision market does not need to be a long running market. Future work will test for the statistical significance of relevant information (identified as important in our idealized decision market model) in laboratory and real world settings

    An AP-1/clathrin coat plays a novel and essential role in forming the Weibel-Palade bodies of endothelial cells

    Clathrin provides an external scaffold to form small 50–100-nm transport vesicles. In contrast, formation of much larger dense-cored secretory granules is driven by selective aggregation of internal cargo at the trans-Golgi network; the only known role of clathrin in dense-cored secretory granules formation is to remove missorted proteins by small, coated vesicles during maturation of these spherical organelles. The formation of Weibel-Palade bodies (WPBs) is also cargo driven, but these are cigar-shaped organelles up to 5 μm long. We hypothesized that a cytoplasmic coat might be required to make these very different structures, and we found that new and forming WPBs are extensively, sometimes completely, coated. Overexpression of an AP-180 truncation mutant that prevents clathrin coat formation or reduced AP-1 expression by small interfering RNA both block WPB formation. We propose that, in contrast to other secretory granules, cargo aggregation alone is not sufficient to form immature WPBs and that an external scaffold that contains AP-1 and clathrin is essential

    Process Evaluation of the Tier 1 Program of the Project P.A.T.H.S.

    To understand the implementation quality of the Tier 1 Program of the Project P.A.T.H.S., two observers carried out process evaluation in six schools randomly selected from the participating schools in the form of systematic observations of 12 units. Results showed that the overall level of program adherence was generally high, ranging from 50% to 95%, with an average of 84.5%. High implementation quality of the program in the areas of student interest, student participation and involvement, classroom control, use of interactive delivery method, use of strategies to enhance student motivation, instructors familiarity with the students, opportunity for reflection, degree of achievement of the objectives, quality of preparation, overall implementation quality, and success of implementation was also observed. The findings provide support for the implementation quality of the program

    Exploring providers’ perspectives of a community based TB approach in Southern Ethiopia: implication for community based approaches

    Abstract Background There is increasing interest in the role of close-to-community providers in supporting universal health coverage, but questions remain about the best approaches to supporting and motivating these providers, and the optimal package they can deliver indifferent contexts and support required. We report on the experiences of different health providers involved in a community based intervention to support access to tuberculosis diagnosis and treatment in Southern Ethiopia. Methods The aim of the study is to explore the experiences of health providers in delivering a community-based tuberculosis package in southern Ethiopia and to draw lessons for community-based programmes. A qualitative methodology was used. Methods included in-depth interviews (IDIs, n= 37) with all health provider groups: Community health promoters (CHPs), health extension workers (HEWs), district supervisors and laboratory technicians were undertaken to obtain a detailed understanding of the experiences of providers in the community based tuberculosis package. These were complemented with cadre specific focus group discussions (n= 3). We used the framework approach for qualitative analysis. Results The key theme that emerged was the positive impact the community-based intervention had on vulnerable groups’ access to diagnosis, care and treatment for tuberculosis. Providers found the positive feedback from, and visible impact on, communities very motivating. Other themes related to motivation and performance included supervision and support; learning new skills; team problem solving/ addressing challenges and incentives. Against the backdrop of the Ethiopian Health Extension Programme (HEP), HEWs were successfully able to take on new tasks (collecting sputum and preparing smears) with additional training and appropriate support from supervisors, laboratory technicians and CHPs. Conclusion All categories of providers were motivated by the high visible impact of the community-based intervention on poor and vulnerable communities and households. HEWs role in the community-based intervention was supported and facilitated through the structures and processes established within the community-based intervention and the broader nation-wide Health Extension Programme. Within community based approaches there is need to develop context embedded strategies to support, sustain and motivate this critical cadre who play a pivotal role in linking health systems and rural communities

    Orthopaedic sport biomechanics: a new paradigm

    This article proposes a new paradigm, "Orthopaedic sport biomechanics", for the understanding of the role of biomechanics in preventing and managing sports injury. Biomechanics has three main roles in this paradigm: (1) injury prevention, (2) immediate evaluation of treatment, and (3) long-term outcome evaluation. Related previous studies showing the approach in preventing and managing anterior cruciate ligament rupture and anterior talofibular ligament tear are highlighted. Orthopaedics and biomechanics specialists are encouraged to understand what they could contribute to the current and future practice of sports medicine

    The use of motion analysis to measure pain-related behaviour in a rat model of degenerative tendon injuries

    Chronic tendinopathy is characterized with longstanding activity-related pain with degenerative tendon injuries. An objective tool to measure painful responses in animal models is essential for the development of effective treatment for tendinopathy. Gait analysis has been developed to monitor the inflammatory pain in small animals. We reported the use of motion analysis to monitor gait changes in a rat model of degenerative tendon injury. Intratendinous injection of collagenase into the left patellar tendon of Sprague Dawley rat was used to induce degenerative tendon injury, while an equal volume of saline was injected in the control groups. Motion analyses with a high speed video camera were performed on all rats at pre-injury, 2, 4, 8, 12 or 16 weeks post injection. In the end-point study, the rats were sacrificed to obtain tendon samples for histological examination after motion analyses. In the follow-up study, repeated motion analyses were performed on another group of collagenase-treated and saline-treated rats. The results showed that rats with injured patellar tendon exhibited altered walking gait as compared to the controls. The change in double stance duration in the collagenase-treated rats was reversible by administration of buprenorphrine (p = 0.029), it suggested that the detected gait changes were associated with pain. Comparisons of end-point and follow-up studies revealed the confounding effects of training, which led to higher gait velocities and probably a different adaptive response to tendon pain in the trained rats. The results showed that motion analysis could be used to measure activity-related chronic tendon pain. © 2009 Elsevier B.V. All rights reserved

    A critical review on the principles, applications, and challenges of waste-to-hydrogen technologies

    Hydrogen sourced from energy recovery processes and conversion of waste materials is a method of providing both a clean fuel and a sustainable waste management alternative to landfill and incineration. The question is whether waste-to–hydrogen can become part of the zero-carbon future energy mix and serve as one of the cleaner hydrogen sources which is economically viable and environmentally friendly. This work critically assessed the potential of waste as a source of hydrogen production via various thermochemical (gasification and pyrolysis) and biochemical (fermentation and photolysis) processes. Research has shown hydrogen production yields of 33.6 mol/kg and hydrogen concentrations of 82% from mixed waste feedstock gasification. Biochemical methods such as fermentation can produce hydrogen up to 418.6 mL/g. Factors including feedstock quality, process requirements and technology availability were reviewed to guide technology selection and system design. Current technology status and bottlenecks were discussed to shape future development priorities. These bottlenecks include expensive production and operation processes, heterogeneous feedstock, low process efficiencies, inadequate management and logistics, and lack of policy support. Improvements to hydrogen yields and production rates are related to feedstock processing and advanced energy efficiency processes such as torrefaction of feedstock which has shown thermal efficiency of gasification up to 4 MJ/kg. This will affect the economic feasibility and concerns around required improvements to bring the costs down to allow waste to viewed as a serious competitor for hydrogen production. Recommendations were also made for financially competitive waste-to-hydrogen development to be part of a combined solution for future energy needs
