Tato diplomová práce se zabývá řešením nejzávažnějších vnitřních vad odlitků ze slitin hliníku litých do pískových forem ve společnosti Slévárna a modelárna Nové Ransko, s.r.o. Řešení problematiky obsahuje popis problémových odlitků s šetřením struktury a fází a následný detailní rozbor jejich výroby. V práci je zkoumán vliv Fe a Mn na možný vznik vnitřní porezity provedením zkušebních taveb se zhodnocením změny vnitřní struktury. Řešením je přísnější kontrola chemického složení taveniny s ohledem na poměr Mn/Fe přítomných ve slitině, nákup odplyňovacího zařízení ke snížení naplynění taveniny a také zdokonalení kontroly naplynění taveniny nákupem vhodného zařízení pro stanovení indexu hustoty.This thesis deals with solving the most serious internal defects in castings made of aluminum alloys which are cast in sand molds in Slévárna a modelárna Nové Ransko Ltd. The problem solving contains a description of the problematic castings with the investigation of the structure and phases and subsequent detailed analysis of their production. The Author of this thesis also examines the influence of Fe and Mn on a possible formation of the internal porosity with the help of test meltings with the evaluation of the internal structural change. The Author’s design solution is a close checking of chemical composition with regard to the ratio Mn / Fe present in the alloy, the purchase of Foundry Degassing Unit to reduce gassiness of melted metals and better control of gassiness with the help of the appropriate equipment for the density Dichte index.

Obraz života a díla Elizabeth Bishop

The aim of the thesis is straightforward: to provide readers with a glimpse into the life of Elizabeth Bishop but not to put emphasis on biographical details. The focus lies on her two most formative relationships with her fellow poets, namely Marianne Moore and Robert Lowell. The thesis aims to present five selected poems and to read them with acknowledging the mutual influences and, at the same time, it strives to provide specific instances of such influences. The first part of the thesis is dedicated to the relationship with Marianne Moore. The two poets met during Bishop's Vassar years and their friendship lasted until Moore's death in 1972. From the teacher - mentored paradigm, their friendship evolved into an affectionate companionship. The thesis introduces their relationship while using selected letters, interviews and, to illustrate the matters more clearly, two of Bishop's poems, "The Roosters" and "Invitation to Miss Marianne Moore." The first poem captures the breaking free from Moore's direct influence and additionally serves as a link to the second part of the thesis. The later poem is used to illustrate their reconciliation and to present the milestone that marks the shift of paradigms. The second part of the thesis presents Robert Lowell, who is the already mentioned link between...Bakalářská práce si klade zdánlivě jednoduchý cíl, a tím je čtení vybraných básní Elizabeth Bishopové skrze optiku dvou vztahů, které byly v jejím životě nejvíce formativní. Jedná se o vztah s modernistickou básnířkou Marianne Moorovou a Bishopovým současníkem Robertem Lowellem. První část práce je věnovaná vztahu s Marianne Mooreovou, kterou Bishopová potkala při svých studiích na Vassar College a která ji provázela životem až do své smrti v roce 1972. Od statusu učitelka-žačka se básnířky posunuly na rovinu vzájemně se respektujících přítelkyň. Bakalářská práce představuje jejich vztah pomocí vybraných pasáží z dopisů a rozhovorů a pro lepší ilustraci si na pomoc bere dvě básně: "Roosters" a "Invitation to Miss Marianne Moore". První báseň ukazuje, jakým způsobem se Bishopová vymanila z vlivu Moore a také předkládá tento důležitý krok jako spojnici k druhé části bakalářské práce. Druhá báseň představuje jakési usmíření a urovnání jejich přátelství a také milník, který značící konec paradigmatu učitelka - žačka a který nastoluje novou fázi jejich vztahu. Analysis of Historical Pharmaceutical Preparations of Caffeine and Santonin.

Historické lékové formy kofeinu a santoninu byly analyzovány hmotnostní spektrometrií, vzorek kofeinu byl kvantifikován spektrofotometricky, vzorek santoninu byl kvantifikován acidobazickou titrací. V analyzované historické injekční formě "Coffeinum natriosalicylicum" (datované přibližně 1945) bylo stanoveno 110 ± 1 % deklarovaného množství kofeinu a 94,33 ± 0,02 % deklarovaného množství salicylanu sodného. Nebyly pozorovány žádné rozpadové produkty kofeinu ani salicylanu sodného. Obsah santoninu v historickém vzorku (datovaném asi 1918) byl stanoven titračně na 104 ± 4 %, jednalo se o čistý santonin bez příměsí. Nebyly nalezeny žádné rozpadové produkty santoninu. Analyzované vzorky byly stabilní i po více jak sedmdesáti, resp. devadesáti, letech skladování.Historical pharmaceutical preparations of caffeine and santonin were analyzed by mass spectrometry, the sample of caffeine was quantified by spectrophotometry, the sample of santonin was quantified by acid-base titration. In the analyzed historical injection solution "Coffeinum natriosalicylicum" (dated circa 1945) was determined 110 ± 1 % of the declared amount of caffeine and 94.33 ± 0.02 % of the declared amount of sodium salicylate. There were observed neither degradation products of caffeine nor sodium salicylate. The content of santonin in the historical sample (dated prior 1918) was determined by titration to 104 ± 4 %, sample was pure santotnin with no additions. No degradation products of santonin was observed. Biologist and Philosopher Zdeněk Neubauer. Studies on Intellectual Biography.

Dissertation abstract Biologist a philosopher Zdeněk Neubauer Studies on Intellectual Biography The subject of the dissertation research is the personality and work of the Czech biologist and philosopher Zdeněk Neubauer in contemporary scientific and social contexts, in selected aspects, concentrating mainly on the period from the beginning of his scientific activity to the 1990s. It focuses on his intellectual biography in general, with an attempt to capture, describe and informatively complete those aspects of his work and professional activities where the necessary coherent information has not been available so far. The basic methodology of the thesis is qualitative research with as much critical objectivity as possible. The necessary evaluation and interpretation procedures are subordinated to this basic approach and objectives. The text of the dissertation is divided into ten chapters, which structure Neubauer's life into thematic units. The chapters are partly arranged chronologically, but most of the themes are interwoven throughout the work, so this structure is only ancillary. The breakdown of the chapters is as follows: Childhood, Naples Stay, Serratia Marcescens, Letter on Biology, Paul Ricoeur and the Rehabilitation of the Subject, Spirit and Nature, Clash of Paradigms, and The Science Wars,...disertační práce Biolog a filosof Zdeněk Neubauer Studie k intelektuální biografii Předmětem výzkumu disertační práce je osobnost a dílo českého biologa a filosofa Zdeňka Neubauera v dobových vědeckých a společenských kontextech, ve vybraných aspektech, soustředěný převážně na období od počátku jeho vědecké činnosti do devadesátých let. Zaměřuje se obecně na jeho intelektuální biografii se snahou postihnout, popsat a informačně ucelit ty stránky jeho díla a odborných aktivit, kde dosud potřebné souvislé informace nebyly k dispozici. Základní metodologií práce je kvalitativní výzkum s co největší možnou kritickou objektivitou. Dílčí nezbytné hodnotící a interpretační postupy jsou podřízeny tomuto základnímu přístupu a cílům. Text disertace je rozdělen do deseti kapitol, jež strukturují Neubauerův život do tematických celků. Kapitoly jsou částečně uspořádány chronologicky, většina témat se však prolíná celým dílem, proto má tato struktura pouze pomocný charakter. Recommendations on the Improvements for Marketing for the VMT Ecopack s.r.o.

Bakalářská práce je orientována na oblast průmyslového trhu, kde jsem se snažila navrhnout patřičná opatření pro zlepšení marketingových aktivit společnosti VMT Ecopack s.r.o. V první části vysvětluji rozdíly mezi spotřebitelským a B2B marketingem, odlišnou komunikaci se zákazníky, segmentaci trhu, řízení vztahu se zákazníky atd. V dalším úseku se věnuji popisu společnosti VMT Ecopack s.r.o., jeho předmět podnikání, organizační strukturu, financování a další důležité informace k osvětlení problematiky. Co se týče návrhové části, pomocí různých metod a analýz jsem sestavila SWOT analýzu podniku, provedla průzkum uvnitř podniku a zaměřila se na zlepšení například podpory prodeje nebo komunikace.Bachelor’s thesis is oriented in industrial market area. I have tried to suggest some important measures to facilitate the improvement of marketing activities in VMT Ecopack company. In the first part I have explained the fundamental differences between B2C and B2B marketing, different communication with customers, targeting the market, customer relationship mananagement etc. In the subsequent parts I describe the VMT Ecopack company, it’s scope of business, organizational structure, funding indside the company and another important informations for the knowledge of matters. To conclude, in the last part of thesis I conducted a SWOT analysis on the company, market research and focused on methods of improvement with regard to sales support and communication with industrial customers.

The Role of iNKT Cells in the Pathogenesis of Infectious And Autoimmune Diseases

iNTK buňky jsou thymus dependentní populace nekonvenčních T lymfocytů. Exprimují konstantní TCR řetězec Vα24Jα18 párovaný s omezeným spektrem β řetězců a rozpoznávají glykolipidové antigeny prezentované neklasickými MHC I. molekulami - CD1d - na povrchu APC. Tato malá subpopulace lymfocytů se podílí na imunitní odpovědi proti mnohým infekčním agens vč. virových infekcí a to i přesto, že viry neexprimují vlastní glykolipidové antigeny. iNKT lymfocyty odlišuje od klasických T lymfocytů i rychlost jejich odpovědi. Velmi záhy po své stimulaci produkují velké množství cytokinů a to Th1, Th2 a Th17 profilu. V mé dizertační práci popisuji chování iNKT lymfocytů během infekce lidskými herpetickými viry. Popisuji signifikantní snížení počtu iNKT u pacientů, kteří nekontrolují latentní VZV infekci a prodělávají časté reaktivace ve formě pásového oparu. Zjištěny a popsány jsou i fenotypové změny iNKT - zvýšená exprese NKG2D receptoru, downregulace NKG2A u skupiny pacientů během akutní infekce HSV-1, kvantitativní změna produkce TNF- α ve smyslu jeho zvýšení. V závěru dizertační práce se zamýšlím nad obecnou pozicí iNKT buněk v imunitní odpovědi a nad jejich evolučním významem. Naše výsledky naznačují podíl iNKT buněk na kontrole herpetických infekci a napodobují více vrozenou imunitní rekci než adaptivní. U...iNKT (invariant natural killer T cell) are thymus-dependent unconventional T cells. They express highly restricted TCR - Vα24Jα18 - chain paired with restricted set of β chains. iNKT cells recognize lipid antigens. These antigens are presented by CD1d molecule on the surface of APC. This small population of cells is involved in the immune responses against various infection agents including viral infection even though viruses do not express their own glycolipid antigens. iNKT cells differ from clasical T lymfocytes in the promtly production of cytokines of Th1, Th2 and Th17 profile. Here I describe their behavior during reactivation of human herpes virus and their role in human defense against the varicella zoster virus. We observed decreased number of iNKT in patients who underwent reactivations of varicella zoster virus because they failed to control latent infection of VZV. Phenotypic changes of iNKT cells were also found and described - increased expression of the NKG2D receptor and downregulation of NKG2A in a group of patients during acute herpes simplex virus infection. iNKT cells enhance their capacity to produce TNF-α. Analysis of Historical Pharmaceutical Preparations of Naloxone, Adrenaline and Ephedrine.

Cílem diplomové práce byla analýza historických léčivých přípravků, zahrnující stanovení obsahu léčivé látky a identifikaci jejích případných degradačních produktů. Historický léčivý přípravek naloxonu byl analyzován hmotnostní spektrometrií. Historické léčivé přípravky adrenalinu a efedrinu byly analyzovány pomocí UHPLC-MS a kvantifikovány metodou kalibrační přímky. V historické injekční formě naloxonu, "NARCAN", datované kolem roku 1980, nebyly pozorovány žádné degradační produkty a naměřené hmotnostní i UV spektrum odpovídalo spektrům pro naloxon. Analyzovaný vzorek naloxonu byl stabilní i po 35 letech skladování. V analyzované historické injekční formě adrenalinu, "Adrenalin Hydrochlor., Dr. Heisler" (datované mezi léty 1917 a 1938) bylo stanoveno 5,26 ± 0,11 % deklarovaného množství adrenalinu. V naměřených spektrech byly patrné degradační produkty, které však zatím nebyly v literatuře popsány a jejich identifikace byla nad rámec této práce. Analyzovaný vzorek adrenalinu byl v průběhu cca devadesáti let téměř kompletně degradován. Stabilitní test prováděný se čtyřmi roztoky standardu adrenalinu prokázal vliv kyslíku, světla, teploty i času na degradaci adrenalinu. V historické injekční formě efedrinu, "Ephedrin" (datovaném mezi léty 1945 a 1948), bylo stanoveno 107,58 ± 0,39 % deklarovaného množství...The aim of the thesis was to analyze the historical pharmaceutical preparations, including the determination of the active substance and identify theirs possible degradation products. A historical pharmaceutical preparation of naloxone was analyzed by mass spectrometry. Historical pharmaceutical preparations of adrenaline and ephedrine were analyzed by UHPLC-MS and were quantified using a calibration curve. In the historical injection solution of naloxone, "NARCAN", dated around 1980, there were no significant degradation products and the measured mass and UV spectrum was consistent with the spectrum of naloxone. The analyzed sample of naloxone was stable even after 35 years of storage. In the analyzed historical injection solution of adrenaline, "Adrenalin Hydrochlor., Dr. Heisler" (dated between 1917 and 1938) was determined 5.26 ± 0.11 % of the declared amount of adrenaline. In the measured spectras were noticeable degradation products, which have not been described in the literature yet and their identification was beyond the scope of this paper. The analyzed sample of adrenaline was almost completely degraded during about ninety years. The stability test carried out with four standard solutions of adrenaline proved influence of oxygen, light, temperature and time on the degradation of adrenaline. Komunitní centrum Buňka - Kutná Hora

V klidné oblasti historického jádra Kutní hory vzniká návrh Komunitního centra Buňka. Buňka je útočištěm pro aktivní děti a mládež, ve kterém se mohou zapojit do množství zájmových a vzdělávacích aktivit. V uličním pohledu hmoty respektují tradiční tvarosloví maloměstské zástavby. Linie hmot se sedlovou střechou je rozdělena do tří na sebe navazujících dudov částečně ustupujících za původní mohutnou opukovou zeď lemují uliční čáru. Tradiční vyzdívané fasády domů jsou narušeny moderním jádrem s kopilitovou fasádou, které slouží jako centrální vstup. Ve dvoře komunitního centra je ukrytý jiný svět. Do výrazného svahu jsou zapuštěny dva železobetonové skelety pohlcené v zeleni.The Buňka Community Center is being designed in a quiet area of the historic centre of Kutná Hora. The centre is a refuge for active children and youth, in which they can participate in several engaging and educational activities.In the street view, the masses respect the traditional morphology of small-town buildings. The line of masses with a gable roof is divided into three subsequent buildings, partially receding behind the original massive marl wall following the street line. The traditional plaster facades of the houses are disturbed by a modern core with a channel glass facade, which serves as a central entrance.Another world is hidden in the courtyard of the community centre. The Use of Methods of the Project Management for Real Company

Diplomová práce je orientovaná na oblast obalového průmyslu. Prostřednictvím aplikací metod projektového managementu je navrženo řešení šicí dílny pro společnost VMT Ecopack s.r.o. Úvodní část je zaměřena na popis základních pojmů a všech fází projektu, zejména pak využívaných nástrojů v sektoru plánování. Analytický úsek práce uvádí první informace o firmě včetně provedeného rozboru vnějšího a vnitřního okolí. V poslední fázi je poté vypracován vlastní návrh na zavedení nového střediska pomocí využití účinných metod projektového řízení.The diploma thesis is focused on the packaging industry. Through the application of project management methods it proposed a solution a sewing workshop for the VMT Ecopack s.r.o. The Introductory part is aimed at the description of basic concepts and all subsequent stages of the project, focusing especially on the methodologies in the planning part. The Analytical section primarily states information about the company including conducted analysis of the internal and external area. The last part has been designed and developed my own proposal to establish, encompass and develop a new section of the company through the use of effective project management tools and approaches.
