NMR-based plant metabolomics protocols: a step-by-step guide.
Metabolomics is one of the “omics” sciences that can reveal the metabolic phenotype of organisms. This capability makes it a valuable tool for plant investigation, as plants present a vast chemical diversity. From the analytical point of view, two main techniques are frequently used in metabolomics and are often complementary: Mass spectrometry (MS) and Nuclear Magnetic Resonance (NMR) spectroscopy. Here, we describe NMR and its applications in plant metabolomics. We start by contextualizing the research field to then explore study design, sample collection, sample preparation, NMR data acquisition, and data analysis, showing the key features for achieving quality and relevant results. Within these topics, the most common databases used for plant metabolites identification and assignments are listed, as these help to shorten the laborious task of metabolomics investigation of natural products. Concerning NMR parameters, we discuss the key pulse sequences, recommend acquisition parameters, and examine the data each sequence can provide. Similarly, we delve into data analysis, highlighting the most commonly used chemometric methods and how to achieve high-quality results. Evaluation of basil extract ( Ocimum basilicum L.) on oxidative, anti-genotoxic and anti-inflammatory effects in human leukocytes cell cultures exposed to challenging agents
ABSTRACT Ocimum is one of the most important genera of the Lamiaceae family. Several studies about basil and its popular use reveal many characteristics of the herb, including its use as antioxidant, anti-aging, anti-inflammatory, anti-carcinogenic, anti-microbial, and cardiovascular agents, among others. In this paper, we evaluated genotoxic, oxidative, and anti-inflammatory parameters from the extract of Ocimum basilicum in different concentrations, using human leukocytes cultures exposed to challenging agents. Our results confirm that the O. basilicum extract acts as an antioxidant and effectively reverts or subjugates the effects of high oxidizing agents such as hydrogen peroxide. These actions are attributed to its composition, which is rich in polyphenols and flavonoids as well as compounds such as rosmarinic acid, all of which have well-known antioxidant activity. We also show that our basil extract presents anti-inflammatory properties, the mechanism of which is a composed interaction between the inhibition of pro-inflammatory mediator and the stimulation of anti-inflammatory cytokines. Although pharmacodynamics studies are necessary to evaluate the activities in vivo, our results demonstrated that basil could act as an antioxidant and anti-inflammatory and a possible alternative for medicinal treatment
Avaliação Ecológica Rápida de Qualidade de Água e Bioindicadores Bentônicos no Parque Nacional da Serra do Gandarela, Minas Gerais
Durante uma disciplina de pós-graduação, realizamos uma Avaliação Ecológica Rápida sobre qualidade de água e biodiversidade de bioindicadores bentônicos no Parque Nacional (PARNA) da Serra do Gandarela, MG. Os objetivos foram: (a) capacitar profissionais na utilização de metodologias atuais em Rapid Assessment Protocols (RAP) para estudos de diagnóstico e monitoramento de integridade ecológica de ecossistemas aquáticos em regiões de cabeceira de bacias hidrográficas; (b) contribuir ao conhecimento da biodiversidade aquática em corpos d’água no PARNA Serra do Gandarela; (c) fomentar intercâmbio e colaboração entre mestrandos, doutorandos, guarda-parques, visitantes, membros de comitês de bacia, gestores ICMBio, moradores e interessados na conservação de biodiversidade no Quadrilátero Ferrífero; (d) gerar informações de base sobre qualidade de água, biodiversidade de macroinvertebrados bentônicos bioindicadores de qualidade de água no PARNA Serra do Gandarela. Os principais resultados revelam que o PARNA Serra do Gandarela é guardião de riachos de cabeceira em condições de referência, com elevada diversidade de hábitats aquáticos e ótima qualidade de água, onde vivem organismos bentônicos sensíveis, tolerantes e resistentes à poluição. Sendo a Serra do Gandarela responsável pela segurança hídrica de grande parte dos municípios do Quadrilátero Ferrífero de Minas Gerais, é extremamente importante que estes riachos em condições de referência sejam conservados e utilizados em futuros estudos de avaliação de impactos ambientais e programas de monitoramento de condições ecológicas de longo prazo. As condições de alta preservação devem servir como baliza de referência para tomadores de decisão em processos de licenciamento ambiental de empreendimentos potencialmente causadores de impactos ambientais e riscos à biodiversidade
