    Effect of acetone extract of Rumex japonicas Houtt on hydrogen peroxide-induced apoptosis in rat myocardial cells

    Purpose: To investigate the protective effect of the acetone extract of Rumex japonicas Houtt. (AER) on rat myocardial cells.Methods: R. japonicas was extracted with 75 % aqueous ethanol by reflux to afford total extract (TER). TER was suspended in water and then extracted with acetone to afford acetone fraction of R. japonicas (AER). High performance liquid chromatography (HPLC) combined with standard substances was carried out to analyze the major constituents of AER. Apoptosis in myocardial H9c2 cell line was induced by H2O2 (100 μmol/L). The cells were treated with AER (50, 100 and 200 μg/mL, and cell viability was evaluated by the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) assay, while oxidative stress level in H9c2 cells was evaluated by determining levels of lactate dehydrogenase (LDH), malondialdehyde (MDA), creatinine kinase (CK), superoxide dismutase (SOD), and catalase (CAT). Furthermore, apoptotic proteins (caspase-3, Bax and Bcl-2) in H9c2 cells were analyzed by using western blot assay.Results: Results revealed that the main components of AER are aloe-emodin, rhein, emodin, chrysophanol and physcion. AER (50, 100 and 200 μg/mL) inhibited the cell viability reduction of the H9c2 cells induced by H2O2 (p < 0.05, p < 0.01, p < 0.01, respectively). AER (50, 100 and 200 μg/mL) decreased LDH and CK contents of H9c2 cells (p < 0.01). The levels of SOD (p<0.01) and CAT (p < 0.01) were increased by AER treatments (100 and 200 μg/mL); in addition, AER (50, 100 and 200 μg/mL) decreased MDA levels (p < 0.01). Besides, the present results also revealed that AER could down-regulate caspase-3 and Bax, but up-regulated Bcl-2.Conclusion: AER alleviates apoptosis induced by H2O2 in myocardial H9c2 cells via inhibition of oxidative stress and mitochondria-mediated apoptosis.     High-efficiency single-photon source above the loss-tolerant threshold for efficient linear optical quantum computing

    Photon loss is the biggest enemy for scalable photonic quantum information processing. This problem can be tackled by using quantum error correction, provided that the overall photon loss is below a threshold of 1/3. However, all reported on-demand and indistinguishable single-photon sources still fall short of this threshold. Here, by using tailor shaped laser pulse excitation on a high-quantum efficiency single quantum dot deterministically coupled to a tunable open microcavity, we demonstrate a high-performance source with a single-photon purity of 0.9795(6), photon indistinguishability of 0.9856(13), and an overall system efficiency of 0.712(18), simultaneously. This source for the first time reaches the efficiency threshold for scalable photonic quantum computing. With this source, we further demonstrate 1.89(14) dB intensity squeezing, and consecutive 40-photon events with 1.67 mHz count rate

    Prodorom platonske antropologije, kršćanstvom još od 2. stoljeća počinje dominirati dualizam, nepomućen ponešto holističkijim tomizmom, a dodatno osnažen kartezijanizmom koji je kršćansku teologiju i dušu odgurnuo još dalje od tijela. Tek se šezdesetih godina 20. stoljeća teolozi počinju osvješćivati o daleko pozitivnijem i inkluzivnijem stavu Biblije prema tijelu. No, stoljeće prije toga, adventistički se pokret rađa u kondicionalizmu kakvog je iznio Hobbes u Levijatanu (XLIV). Čovjek nema dušu, on jest »živa duša« – tijelo oživljeno »dahom života« (Post 2,7). Bez tijela nema života, pa tako ni vječnog pakla. Ovome adventisti pridružuju i filozofiju zdravstvene reforme u kojoj njega tijela ima ključnu ulogu i o čemu ovisi intelektualni i duhovni napredak čovjeka, te na tom temelju grade obilnu zdravstvenu i obrazovnu praksu. Ova fizikalistička inačica kršćanske antropologije jedinstven je svjetonazorski doprinos filozofiji tijela i tema vrijedna akademske pažnje.Following the spread of Platonic anthropology, Christianity has started, already since the 2nd century A.D., to be dominated by dualism – a trend undisturbed by somewhat more holistic Thomism, and further strengthened by Cartesianism, which distanced Christian theology and soul even further away from the body. During the 1960s, theologians have become aware of the far more positive and inclusive attitude that the Bible has towards the body. Yet, a century before, the Adventist movement was born in conditionalism such as presented by Hobbes in Leviathan (XLIV). Man does not have a soul; he is a “living soul” – a body vivified by the “breath of life” (Gen 2:7). Without the body, there is no life, nor, consequentially, eternal hell. To this Adventists have also conjoined a philosophy of health reform in which the care of the body has a key role, and upon which depends man’s intellectual and spiritual wellbeing. On this foundation, they have built a rich healthcare and educational practice. This physicalist version of Christian anthropology is a unique worldview contribution to philosophy of the body and a subject worthy of academic attention

    The Bone-Protecting Efficiency of Chinese Medicines Compared With Western Medicines in Rheumatoid Arthritis: A Systematic Review and Meta-Analysis of Comparative Studies

    Background: Rheumatoid Arthritis (RA) is a systemic autoimmune disease leading to joint destruction. The prevention of bone and cartilage destruction has received increased attention in recent years.Objective: To evaluate the current evidences regarding the bone-protecting efficacy of Chinese medicine or the combination of Chinese medicine and Western medicine for RA.Methods: We comprehensively searched PubMed, Embase, the Cochrane Library (www.thecochranelibrary.com), the China National Knowledge Infrastructure (CNKI), the Database for Chinese Technical Periodicals (VIP), and SinoMed. We then performed a systematic review and cumulative meta-analysis of all randomized controlled trials (RCTs) assessing the two therapy methods.Results: Sixteen studies including 1,171 patients were included in the final analysis. The results showed that Chinese medicine could significantly improve the bone mineral density (BMD) (mean difference [MD] = 0.05 /g·cm−2, 95% CI [0.03, 0.08], P < 0.00001), and decrease the serum matrix metalloproteinase 3 (MMP-3) ([SMD] = −2.84, 95% CI [−4.22, −1.47], P < 0.0001).Conclusions: Chinese medicine may provide an efficiently alternative choice for the treatment of RA in terms of the bone-protecting efficiency. Given the inherent limitations of the included studies, future well-designed RCTs are required to confirm and update the findings of this analysis

    Management of granulomatous lobular mastitis: an international multidisciplinary consensus (2021 edition)

    Granulomatous lobular mastitis (GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence. Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions, etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology. The approach to treatment of GLM should be applied according to the different clinical stage of GLM. This evidence-based consensus would be valuable to assist front-line surgeons and medical specialists in the optimal management of GLM.Improving the Ability of Diagnosis and Treatment of Difficult Disease

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data
