Examining the Reasons for Student Responses to Threatening Behaviors on a College Campus
Underreporting criminal activity to authorities can pose significant challenges, particularly within college campuses. Crime prevention teams have recognized the importance of reporting potentially concerning behaviors that may precede violent acts. However, reasons for reporting preincident behaviors have been understudied among college samples and failed to account for informal responses, such as talking to third parties or changing personal security features. The present study surveyed 1,075 students from a midwestern state university and evaluated their awareness of threatening or concerning behaviors on campus, response behaviors, and reasons for either acting on or failing to report preincident behaviors. Findings reflected reporting rates (12.3%) consistent with the scientific literature but revealed informal responding as the most common reaction to preincident behaviors (44.6%). Reasons for not taking action included a desire to be uninvolved and perceiving the situation as unlikely to be immediately dangerous. By contrast, those who reported preincident behaviors appraised the situation as immediately dangerous and likely to result in harm. Evidence for Circumstellar Disks around Young Brown Dwarfs in the Trapezium Cluster
We report the results of deep infrared observations of brown dwarf candidates
in the Trapezium cluster in Orion. Analysis of the JHK color-color diagram
indicates that a large fraction (approx 65% +/- 15%) of the observed sources
exhibit infrared excess emission. This suggests the extreme youth of these
objects and in turn, provides strong independent confirmation of the existence
of a large population of substellar objects in the cluster. Moreover, this
suggests that the majority of these substellar objects are presently surrounded
by circumstellar disks similar to the situation for the stellar population of
the cluster. This evidence for a high initial disk frequency (> 50%) around
cluster members of all masses combined with the smooth continuity of the
cluster's initial mass function across the hydrogen burning limit suggests that
a single physical mechanism is likely responsible for producing the entire
cluster mass spectrum down to near the deuterium burning limit. The results may
also indicate that even substellar objects are capable of forming with systems

Synergistic drug combinations from electronic health records and gene expression.
ObjectiveUsing electronic health records (EHRs) and biomolecular data, we sought to discover drug pairs with synergistic repurposing potential. EHRs provide real-world treatment and outcome patterns, while complementary biomolecular data, including disease-specific gene expression and drug-protein interactions, provide mechanistic understanding.MethodWe applied Group Lasso INTERaction NETwork (glinternet), an overlap group lasso penalty on a logistic regression model, with pairwise interactions to identify variables and interacting drug pairs associated with reduced 5-year mortality using EHRs of 9945 breast cancer patients. We identified differentially expressed genes from 14 case-control human breast cancer gene expression datasets and integrated them with drug-protein networks. Drugs in the network were scored according to their association with breast cancer individually or in pairs. Lastly, we determined whether synergistic drug pairs found in the EHRs were enriched among synergistic drug pairs from gene-expression data using a method similar to gene set enrichment analysis.ResultsFrom EHRs, we discovered 3 drug-class pairs associated with lower mortality: anti-inflammatories and hormone antagonists, anti-inflammatories and lipid modifiers, and lipid modifiers and obstructive airway drugs. The first 2 pairs were also enriched among pairs discovered using gene expression data and are supported by molecular interactions in drug-protein networks and preclinical and epidemiologic evidence.ConclusionsThis is a proof-of-concept study demonstrating that a combination of complementary data sources, such as EHRs and gene expression, can corroborate discoveries and provide mechanistic insight into drug synergism for repurposing

De novo assembly of the cattle reference genome with single-molecule sequencing.
BackgroundMajor advances in selection progress for cattle have been made following the introduction of genomic tools over the past 10-12 years. These tools depend upon the Bos taurus reference genome (UMD3.1.1), which was created using now-outdated technologies and is hindered by a variety of deficiencies and inaccuracies.ResultsWe present the new reference genome for cattle, ARS-UCD1.2, based on the same animal as the original to facilitate transfer and interpretation of results obtained from the earlier version, but applying a combination of modern technologies in a de novo assembly to increase continuity, accuracy, and completeness. The assembly includes 2.7 Gb and is >250Ă more continuous than the original assembly, with contig N50 >25 Mb and L50 of 32. We also greatly expanded supporting RNA-based data for annotation that identifies 30,396 total genes (21,039 protein coding). The new reference assembly is accessible in annotated form for public use.ConclusionsWe demonstrate that improved continuity of assembled sequence warrants the adoption of ARS-UCD1.2 as the new cattle reference genome and that increased assembly accuracy will benefit future research on this species
Moving forward during major goal blockage: situational goal adjustment in women facing infertility
Individuals confronting chronic medical conditions often face profound challenges to cherished life goals. The primary aim of this study was to examine the associations of goal adjustment with psychological adjustment in the context of infertility. At study entry (T1; n = 97) and 6 months later (T2; n = 47), women in fertility treatment completed measures of goal blockage, goal adjustment ability, and psychological adjustment. At T1, greater perceived and actual goal blockage were related to negative psychological adjustment. Ability to disengage from the goal of biological parenthood was associated with less infertility-specific thought intrusion, whereas engagement with other goals was related to fewer depressive symptoms and greater positive states of mind. Greater general goal engagement was protective against the negative relationships between low goal disengagement and the dependent variables. Promoting letting go of the unattainable and investing in the possible may be a useful intervention to foster well-being among individuals experiencing profound goal blockage
Prevalence of and factors associated with late diagnosis of HIV in Malawi, Zambia, and Zimbabwe: Results from population-based nationally representative surveys
Introduction
Late diagnosis of HIV (LD) increases the risk of morbidity, mortality, and HIV transmission. We used nationally representative data from population-based HIV impact assessment (PHIA) surveys in Malawi, Zambia, and Zimbabwe (2015â2016) to characterize adults at risk of LD and to examine associations between LD and presumed HIV transmission to cohabiting sexual partners.
Methods
We estimated the prevalence of LD, defined as CD4 count <350 cells/ÎŒL, among adults newly diagnosed with HIV during the surveys and odds ratios for associated factors. We linked newly diagnosed adults (index cases) to their household sexual partners and calculated adjusted odds ratios for associations between LD of the index case, viral load of the index case, and duration of HIV exposure in the relationship, and the HIV status of the household sexual partner.
Results
Of 1,804 adults who were newly diagnosed with HIV in the surveys, 49% (882) were diagnosed late. LD was associated with male sex, older age, and almost five times the odds of having an HIV-positive household sexual partner (adjusted odds ratio [aOR], 4.65 [95% confidence interval: 2.56â8.45]). Longer duration of HIV exposure in a relationship and higher viral load of the index case were both independently associated with higher odds of having HIV-positive household sexual partners. Individuals with HIV exposure of more than 5 years had more than three times (aOR 3.42 [95% CI: 1.63â7.18]) higher odds of being HIV positive than those with less than 2 years HIV exposure. The odds of being HIV positive were increased in individuals who were in a relationship with an index case with a viral load of 400â3499 copies/mL (aOR 4.06 [95% CI 0.45â36.46]), 3,500â9,999 copies/mL (aOR 11.32 [95% CI: 4.08â31.39]), 10,000â49,999 copies/mL (aOR 17.07 [95% CI: 9.18â31.72]), and â„50,000 copies/mL (aOR 28.41 [95% CI: 12.18â66.28]) compared to individuals who were in a relationship with an index case with a viral load of <400 copies/mL.
Conclusions
LD remains a challenge in Southern Africa and is strongly associated with presumed HIV transmission to household sexual partners. Our study underscores the need for earlier HIV diagnosis, particularly among men and older adults, and the importance of index testing
Prevalence of nonsuppressed viral load and associated factors among HIV-positive adults receiving antiretroviral therapy in Eswatini, Lesotho, Malawi, Zambia and Zimbabwe (2015 to 2017): results from population-based nationally representative surveys.
INTRODUCTION
The global target for 2020 is that â„90% of people living with HIV (PLHIV) receiving antiretroviral therapy (ART) will achieve viral load suppression (VLS). We examined VLS and its determinants among adults receiving ART for at least four months.
METHODS
We analysed data from the population-based HIV impact assessment (PHIA) surveys in Eswatini, Lesotho, Malawi, Zambia and Zimbabwe (2015 to 2017). PHIA surveys are nationally representative, cross-sectional household surveys. Data collection included structured interviews, home-based HIV testing and laboratory testing. Blood samples from PLHIV were analysed for HIV RNA, CD4 counts and recent exposure to antiretroviral drugs (ARVs). We calculated representative estimates for the prevalence of VLS (viral load <1000Â copies/mL), nonsuppressed viral load (NVL; viral load â„1000Â copies/mL), virologic failure (VF; ARVs present and viral load â„1000Â copies/mL), interrupted ART (ARVs absent and viral load â„1000Â copies/mL) and rates of switching to second-line ART (protease inhibitors present) among PLHIV aged 15 to 59Â years who participated in the PHIA surveys in Eswatini, Lesotho, Malawi, Zambia and Zimbabwe, initiated ART at least four months before the survey and were receiving ART at the time of the survey (according to self-report or ARV testing). We calculated odds ratios and incidence rate ratios for factors associated with NVL, VF, interrupted ART, and switching to second-line ART.
RESULTS
We included 9200 adults receiving ART of whom 88.8% had VLS and 11.2% had NVL including 8.2% who experienced VF and 3.0% who interrupted ART. Younger age, male sex, less education, suboptimal adherence, receiving nevirapine, HIV non-disclosure, never having married and residing in Zimbabwe, Lesotho or Zambia were associated with higher odds of NVL. Among people with NVL, marriage, female sex, shorter ART duration, higher CD4 count and alcohol use were associated with lower odds for VF and higher odds for interrupted ART. Many people with VF (44.8%) had CD4 counts <200 cells/”L, but few (0.31% per year) switched to second-line ART.
CONCLUSIONS
Countries are approaching global VLS targets for adults. Treatment support, in particular for younger adults, and people with higher CD4 counts, and switching of people to protease inhibitor- or integrase inhibitor-based regimens may further reduce NVL prevalence
