Tests comparatifs d'utilisabilité des plateformes Swissbib.ch et Data.bnf.fr
Lâapparition des plateformes en Linked Open Data, permet dâentrevoir de nouvelles possibilitĂ©s dâaccĂšs Ă lâinformation et offre de nouvelles opportunitĂ©s en termes dâutilisabilitĂ©. Ainsi, ce travail est rĂ©alisĂ© dans le cadre du projet linked.swissbib.ch qui vise Ă enrichir le catalogue swissbib.ch grĂące au web sĂ©mantique. Le but de ce travail de bachelor est par le biais de tests comparatifs dâĂ©valuer la satisfaction des utilisateurs vis-Ă -vis de ce nouveau type dâoutil de recherche et la maniĂšre dont ils sâapproprient ce dernier, face Ă lâutilisation dâun catalogue « standard » de recherche. Ainsi, ces tests se focalisent sur la plateforme data.bnf.fr dĂ©veloppĂ©e en Linked Open Data et le mĂ©ta catalogue swissbib.ch. Lâobjectif de cette mĂ©thode est de recueillir des donnĂ©es permettant la formulation de recommandations pour le dĂ©veloppement de fonctionnalitĂ©s en Linked Open Data pour la plateforme swissbib.ch. A travers ce mĂ©moire, le processus qui a permis dâĂ©laborer ce test dans lâobjectif de rĂ©pondre aux besoins du mandat est prĂ©sentĂ© de mĂȘme que les rĂ©sultats obtenus. Ces derniers mettent en avant le potentiel dâune plateforme en Linked Open Data face Ă lâutilisation dâun catalogue standard de recherche. En effet, il apparaĂźt que la plateforme data.bnf.fr de par sa structure et ses fonctionnalitĂ©s semble faciliter le processus de prise en main de lâoutil et a un impact positif sur la satisfaction des utilisateurs par rapport Ă lâutilisation du mĂ©ta catalogue swissbib.ch. Par consĂ©quent, la plateforme data.bnf.fr semble ĂȘtre un modĂšle intĂ©ressant Ă suivre dans le cadre du dĂ©veloppement de fonctionnalitĂ©s en web sĂ©mantique pour swissbib.ch. Toutefois, les tests ont rĂ©vĂ©lĂ© Ă©galement des Ă©lĂ©ments auxquels il faut ĂȘtre attentif dans une logique de structure telle quâon peut la trouver sur data.bnf.fr. Enfin, ce travail a esquissĂ© le potentiel de prise en main dâune plateforme LOD, notamment pour les participants nâĂ©tant pas rodĂ©s Ă lâutilisation des catalogues de bibliothĂšques. Cet aspect met en avant le potentiel du Linked Open Data afin de rendre les catalogues de bibliothĂšques davantage « user friendly »
Role of Phosphorylation in Moesin Interactions with PIP 2 -Containing Biomimetic Membranes
International audienceMoesin, a protein of the ezrin, radixin, and moesin family, which links the plasma membrane to the cytoskeleton, is involved in multiple physiological and pathological processes, including viral budding and infection. Its interaction with the plasma membrane occurs via a key phosphoinositide, the phosphatidyl(4,5)inositol-bisphosphate (PIP2), and phosphorylation of residue T558, which has been shown to contribute, in cellulo, to a conformationally open protein. We study the impact of a double phosphomimetic mutation of moesin (T235D, T558D), which mimics the phosphorylation state of the protein, on protein/PIP2/microtubule interactions. Analytical ultracentrifugation in the micromolar range showed moesin in the monomer and dimer forms, with wild-type (WT) moesin containing a slightly larger fraction (âŒ30%) of dimers than DD moesin (10-20%). Only DD moesin was responsive to PIP2 in its micellar form. Quantitative cosedimentation assays using large unilamellar vesicles and quartz crystal microbalance on supported lipid bilayers containing PIP2 reveal a specific cooperative interaction for DD moesin with an ability to bind two PIP2 molecules simultaneously, whereas WT moesin was able to bind only one. In addition, DD moesin could subsequently interact with microtubules, whereas WT moesin was unable to do so. Altogether, our results point to an important role of these two phosphorylation sites in the opening of moesin: since DD moesin is intrinsically in a more open conformation than WT moesin, this intermolecular interaction is reinforced by its binding to PIP2. We also highlight important differences between moesin and ezrin, which appear to be finely regulated and to exhibit distinct molecular behaviors
From Tomato Pomaces Biorefinery to Biobased Shape-Memory Semicrystalline Polyester Networks
The
development of new biobased polyester materials is of great
interest. Seed-free and pulp-free peels provided by the biorefinery
of industrial tomato wastes, i.e., pomaces, are rich in cutin, a polyester
of hydroxy fatty acids. In this study, these fatty acids were recovered
after alkaline hydrolysis, resulting in a crude brown-colored extract
due to the coextracted phenolic compounds. Further purification drastically
reduced the amount of these phenolic compounds, resulting in a pale
yellow product containing about 95% (9/10)-16-dihydroxy hexadecanoic
acid and 5% dicarboxylic fatty acids. Polymerization of the purified
extract at 150 °C results in a weakly cross-linked polyester
network with an estimated average number of 80 repeating units between
nodes. Interestingly, the mechanical behavior of this material at
25 °C is determined by its semicrystalline structure. A necking
phenomenon is observed during tensile tests, resulting in an apparent
plastic deformation of 286% and a Young modulus of 72 MPa. In addition,
this biobased polyester exhibits shape-memory properties with the
ability to be hot or cold-programmed. This work highlights the significant
impact of minor compounds related to the biochemical heterogeneity
of agro-industrial waste products on the polyester properties and
the potential of the biorefinery process to modulate the properties
of biopolymers
