    We live in a world where resources are limited and how we invest them has an impact on the citizens’ wellbeing. The goal of this thesis is to provide, through the tools of economic analysis, some insights for the optimal allocation of our resources in three different areas: economics of crime, economics of education and economics of labour. First, societies aim at lowering crime rates and this is why a great amount of resources is spent in punishing offenders. How effective is punishment in lowering crime rates is still unclear: what are the forms of custody that deter lawbreakers from resuming their life of crime? Through a fuzzy regression discontinuity design, we show that keeping young offenders separate from their older peers and far from an overcrowded environment is beneficial only when rehabilitation is offered. Second, empowering women and enhancing children’s early childhood development are two important objectives that are often pursued by independent policy initiatives in developing countries. Understanding the consequences of exploiting potentially beneficial complementarities in pursuing both aims together can be relevant. Through a quasi-natural experiment we evaluate a program implemented in Quito, Ecuador, that targets both. We find that women who are involved in the education of their children are empowered in different dimensions, as reflected in their higher likelihood to find full-time employment in the formalsector and in their greater independence in intra-household decision-making. Children’s dropout rates decrease, while school grades and scores on cognitive tests increase, particularly for girls. Finally, governments can introduce and raise minimum wage levels in order to protect their workers. We want to understand the implications of minimum wages on informal markets in developing countries. By exploiting relative variation in minimum wages across labour market groups within countries we show that a higher minimum wage is associated with a larger selfemployment share

    Tough on young offenders : harmful or helpful?

    How harshly should society punish young lawbreakers in order to prevent or reduce their criminal activity in the future? Through a fuzzy regression discontinuity design, we shed light on the question by exploiting two quasi-natural experiments stemming to compare outcomes from relatively harsh or rehabilitative criminal incarceration practices involving young offenders in the 1980’s in England and Wales. According to our local linear regression estimates, young offenders exposed to the harsher youth facilities are 20.7 percent more likely to recidivate in the nine years subsequent to their custody, and they commit on average 2.84 offences more than offenders who experienced prison. Moreover, they are more likely to commit violent offences, thefts, burglaries and robberies. On the contrary, offenders who were sent to the more rehabilitative youth facilities are less likely to reoffend in the future when compared to offenders sent to prison. We conclude that it is effective to keep young offenders separate from their older peers in prison, but only when they are held in institutions that are not solely focused on punishment

    CD271 downregulation promotes melanoma progression and invasion in 3-dimensional models and in zebrafish

    CD271 is a neurotrophin receptor variably expressed in melanoma. While contradictory data are reported on its role as a marker of tumor initiating cells, little is known on its function in tumor progression. CD271 expression was higher in spheroids derived from freshly isolated cells of primary melanomas and in primary WM115 and WM793-B cell lines, while it decreased during progression to advanced stages in cells isolated from metastatic melanomas and in metastatic WM266-4 and 1205Lu cell lines. Moreover, CD271 was scarcely detected in the highly invasive spheroids (SKMEL28 and 1205Lu). CD271, originally expressed in the epidermis of skin reconstructs, disappeared when melanoma started to invade the dermis. SKMEL8 CD271(-) cells showed greater proliferation and invasiveness in vitro, and were associated with a higher number of metastases in zebrafish, as compared to CD271(+) cells. CD271 silencing in WM115 induced a more aggressive phenotype in vitro and in vivo. On the contrary, CD271 overexpression in SKMEL28 cells reduced invasion in vitro, and CD271 overexpressing 1205Lu cells was associated with a lower percentage of metastases in zebrafish. A reduced cell-cell adhesion was also observed in absence of CD271. Taken together, these results indicate that CD271 loss is critical for melanoma progression and metastasis

    Autothermal biochar production and characterization at pilot scale

    The present work reports on the results from the validation campaign of an autothermal pilot carbonization unit (CarbON) and on the characterization of the produced biochar and pyrolysis vapors. The proposed pilot plant leverages the simplicity and effectiveness of autothermal operations together with open top, downdraft design, to bring to the small scale the performance of larger installations. In autothermal operation, heat for the process is internally provided by combusting part of the feedstock and evolved volatiles inside the reactor, the so called “oxidative pyrolysis”.     Development of a Desmocollin-3 Active Mouse Model Recapitulating Human Atypical Pemphigus

    Pemphigus vulgaris (PV) is a life-threatening mucocutaneous autoimmune blistering disease. It is often associated with autoantibodies to the desmosomal adhesion proteins Desmoglein 3 (DSG3) and Desmoglein 1 (DSG1). Recently, auto-antigens, such as desmocollins and others have been described in PV and in atypical pemphigus forms such as Pemphigus Herpetiformis (PH), Pemphigus Vegetans (PVeg), and Paraneoplastic Pemphigus (PP). Desmocollins belong to a cadherin subfamily that provides structure to the desmosomes and play an important role in cell-to-cell adhesion. In order to verify the pathogenic activity of anti-Desmocollin 3 (DSC3) antibodies, we developed an active disease model of pemphigus expressing anti-DSC3 autoantibodies or antiDSC3 and anti-DSG3 antibodies. This approach included the adoptive transfer of DSC3 and/or DSG3 lymphocytes to Rag2(-/-) immunodeficient mice that express DSC3 and DSG3. Our results show that the presence of anti-DSC3 auto-antibodies is sufficient to determine the appearance of a pathological phenotype relatable to pemphigus, but with features not completely super-imposable to those observed in the DSG3 active model, suggesting that the DSC3 active model might mimic the atypical pemphigus. Moreover, the presence of both anti-DSC3 and anti-DSG3 antibodies determines a more severe phenotype and a slower response to prednisolone. In conclusion, we have developed an adult DSC3 pemphigus mouse model that differs from the DSG3 model and supports the concept that antigens other than desmogleins may be responsible for different phenotypes in human pemphigus

    Broadband Dielectric Spectroscopy Study of Biobased Poly(alkylene 2,5-furanoate)s’ Molecular Dynamics

    Abstract Poly(2,5-alkylene furanoate)s are bio-based, smart, and innovative polymers that are considered the most promising materials to replace oil-based plastics. These polymers can be synthesized using ecofriendly approaches, starting from renewable sources, and result into final products with properties comparable and even better than those presented by their terephthalic counterparts. In this work, we present the molecular dynamics of four 100% bio-based poly(alkylene 2,5-furanoate)s, using broadband dielectric spectroscopy measurements that covered a wide temperature and frequency range. We unveiled complex local relaxations, characterized by the simultaneous presence of two components, which were dependent on thermal treatment. The segmental relaxation showed relaxation times and strengths depending on the glycolic subunit length, which were furthermore confirmed by high-frequency experiments in the molten region of the polymers. Our results allowed determining structure–property relations that are able to provide further understanding about the excellent barrier properties of poly(alkylene 2,5-furanoate)s.     Antimicrobial resistance in Enterococcus strains isolated from healthy domestic dogs

    Enterococci are opportunistic bacteria that cause severe infections in animals and humans, capable to acquire, express, and transfer antimicrobial resistance. Susceptibility to 21 antimicrobial agents was tested by the disk diffusion method in 222 Enterococcus spp. strains isolated from the fecal samples of 287 healthy domestic dogs. Vancomycin and ampicillin minimum inhibitory concentrations (MICs) and high-level aminoglycoside resistance (HLAR) tests were also performed. Isolates showed resistance mainly to streptomycin (88.7%), neomycin (80.6%), and tetracycline (69.4%). Forty-two (18.9%) isolates showed an HLAR to streptomycin and 15 (6.7%) to gentamicin. Vancomycin and ampicillin MIC values showed 1 and 18 resistant strains, respectively. One hundred and thirty-six (61.2%) strains were classified as multidrug resistant and six (2.7%) strains as possibly extensively drug-resistant bacteria. Enterococcus faecium and Enterococcus faecalis were the most prevalent antimicrobial resistant species. Companion animals, which often live in close contact with their owners and share the same environment, represent a serious source of enterococci resistant to several antibiotics; for this reason, they may be a hazard for public health by providing a conduit for the entrance of resistance genes into the community
