    B cell activation and proliferation increase intracellular zinc levels

    Zinc ions serve as second messengers in major cellular pathways, including the regulation pathways of proliferation and their proper regulation is necessary for homeostasis and a healthy organism. Accordingly, expression of zinc transporters can be altered in various cancer cell lines and is often involved in producing elevated intracellular zinc levels. In this study, human B cells were infected with Epstein–Barr virus (EBV) to generate immortalized cells, which revealed traits of tumor cells, such as high proliferation rates and an extended lifespan. These cells showed differentially altered zinc transporter expression with ZIP7 RNA and protein expression being especially increased as well as a corresponding increased phosphorylation of ZIP7 in EBV-transformed B cells. Accordingly, free zinc levels were elevated within these cells. To prove whether the observed changes resulted from immortalization or rather high proliferation, free zinc levels in in vitro activated B cells and in freshly isolated B cells expressing the activation marker CD69 were determined. Here, comparatively increased zinc levels were found, suggesting that activation and proliferation, but not immortalization, act as crucial factors for the elevation of intracellular free zinc

    Оптимизация условий экстрагирования меланинов из лузги подсолнечника

    Определены оптимальные значения параметров экстрагирования меланинов из лузги подсолнечника: частота вибраций 24 Гц, концентрация раствора гидроксида натрия 0,36 М, 3 ступени экстрагирования, продолжительность экстрагирования на каждой ступени 20 минут. Выход меланинов при данных условиях составил 13,09%. Полученные данные могут быть использованы при разработке технологии получения меланина из альтернативного сырья - лузги подсолнечника.Defined are the optimal values of the parameters extraction of melanin from the husks of sunflower: the vibration frequency 24 Hz, the concentration of sodium hydroxide solution 0,36 M, 3 stages of extraction, duration of extraction at each stage in 20 minutes. The degree of extraction of melanin under these conditions were 13,09%. The results obtained determine the possibility of development of technology for production melanines from alternative raw materials - sunflower husk

    В работе представлены результаты исследования каталитической активности металлических порошков ВК6 используемых в роли катализатора в синтезе Фишера-Тропша. Испытания были проведены на лабораторной установке, разработанной и изготовленной ЗАО «Катакон», г.Новосибирск. Для получения тонкодисперсных порошков использовался метод электроискрового диспергирования в жидкой среде. Для изучения физико-химических свойств полученных порошков использовались: определение величины удельной поверхности частиц порошков по методу БЭТ, методы растровой и сканирующей электронной микроскопии, а также рентгенофазового анализа.The research effort presents the results of a studing catalytic activity of metal powders VK6 used as catalyst in the synthesis of Fischer-Tropsch. Tests were conducted on a laboratory installation, which was designed and manufactured by CJSC "Caticon", Novosibirsk. The method of electrospark dispersion in liquid medium was applied during obtaining a fine powder. To study the physico-chemical properties of the obtained powders were used for determination of the specific surface of particles of powders by BET method, methods of raster and scanning electron microscopy, and X-ray phase analysis

    S-thioallylation of proteins by the garlic defence substance allicin and its biological effects

    A single clove of edible garlic (Allium sativum L.) of about 10 g produces up to 5 mg of allicin (diallylthiosulfinate), a thiol-reactive sulfur-containing defence substance that gives injured garlic tissue its characteristic smell. Allicin induces apoptosis or necrosis in a dose-dependent manner but biocompatible doses influence cellular metabolism and signalling cascades. Oxidation of protein thiols and depletion of the glutathione pool are thought to be responsible for allicin's physiological effects. Here, we studied the effect of allicin on post-translational thiol-modification in human Jurkat T-cells using shotgun LC-MS/MS analyses. We identified 332 proteins that were modified by S-thioallylation in the Jurkat cell proteome which causes a mass shift of 72 Da on cysteines. Many S-thioallylated proteins are highly abundant proteins, including cytoskeletal proteins tubulin, actin, cofilin, filamin and plastin-2, the heat shock chaperones HSP90 and HSPA4, the glycolytic enzymes GAPDH, ALDOA, PKM as well the protein translation factor EEF2. Allicin disrupted the actin cytoskeleton in murine L929 fibroblasts. Allicin stimulated the immune response by causing Zn2+ release from proteins and increasing the Zn2+-dependent IL-1-triggered production of IL-2 in murine EL-4 T-cells. Furthermore, allicin caused inhibition of enolase activity, an enzyme considered a cancer therapy target. In conclusion, our study revealed the widespread extent of S-thioallylation in the human Jurkat cell proteome and showed effects of allicin exposure on essential cellular functions of selected targets, many of which are targets for cancer therapy

    An individual’s zinc status has a significant impact on the immune system, and zinc deficiency, as well as supplementation, modulates immune function. To investigate the effects of zinc on different leukocyte subsets, we used microarray technology to analyze and compare the changes in mRNA expression in cell culture models of monocytes (THP-1), T cells (Jurkat), and B cells (Raji), in response to supplementation for 40 h with 50 μM zinc or 2.5 μM of the membrane-permeant zinc chelator TPEN [N,N,N′,N′-tetrakis-(2-pyridyl-methyl)ethylenediamine], respectively. In each cell type, several hundred genes were identified to be zinc sensitive, but only a total of seven genes were commonly regulated in all three cell lines. The majority of those genes were involved in zinc homeostasis, and none in immune function. Nevertheless, further analysis revealed that zinc affects entire functional networks of genes that are related to proinflammatory cytokines and cellular survival. Although the zinc-regulated activities are similar throughout the gene networks, the specific genes that are affected vary significantly between different cell types, a situation that helps to elucidate the disparity of the effects that zinc has on different leukocyte populations
