Inventory extension at the Nuclear Materials Storage Facility
The planned renovation of the Nuclear Material Storage Facility (NMSF) at Los Alamos National Laboratory will be a significant addition to the plutonium storage capacity of the nuclear weapons complex. However, the utility of the facility may be impaired by an overly conservative approach to performing inventories of material in storage. This report examines options for taking advantage of provisions in Department of Energy orders to extend the time between inventories. These extensions are based on a combination of modern surveillance technology, facility design features, and revised operational procedures. The report also addresses the possibility that NMSF could be the site of some form of international inspection as part of the US arms control and nonproliferation policy

Geochemistry of Background Sediment Samples at Technical Area 39, Los Alamos National Laboratory
This report presents results of chemical analyses of 24 analytes in 16 background sediment samples collected from Ancho Canyon and Indio Canyon at Technical Area (TA) 39, Los Alamos National Laboratory. Preliminary upper tolerance limits (UTLS) for sediments are calculated from this data set but, because of the small sample size, these UTLs exceed the maximum values in the data set by up to 50'ZO and will require revision as more background sediment data are obtained

Migration of Sr-20, Cs-137, and Pu-239/240 in Canyon below Los Alamos outfall
Technical Area-21 (TA-21) of Los Alamos National Laboratory (LANL) is on a mesa bordered by two canyons DP Canyon and Los Alamos (LA) Canyon. DP Canyon is a small semiarid watershed with a well defined channel system where the stream flow is ephemeral. TA-21 has had a complex history of waste disposal as research to determine the chemical and metallurgical properties of nuclear materials occurred here from 1945-1978. Due to these operations, the TA-21 mesa top and bordering canyons have been monitored and characterized by the LANL Environmental Restoration Program. Results identify radionuclide values at outfall. 21-011 (k) which exceed Screening Action Levels, and points along DP Canyon which exceed regional background levels. The radiocontaminants considered in this study are strontium-90, cesium-137, and plutonium-239. This research examines sediment transport and speciation of radionuclide contaminant migration from a source term named SWMU 21-011 (k) down DP Canyon. Three dimensional surface plots of data from 1977-1994 are used to portray the transport and redistribution of radioactive contaminants in an alluvial stream channel. An overall decrease in contamination concentration since 1983 has been observed which could be due to more stringent laboratory controls and also to the removal of main plutonium processing laboratories to another site

Recent observation of beta decay of 115-In to the first excited level of
115-Sn with an extremely low Q_beta value (Q_beta ~ 1 keV) could be used to set
a limit on neutrino mass. To give restriction potentially competitive with
those extracted from experiments with 3-H (~2 eV) and 187-Re (~15 eV), atomic
mass difference between 115-In and 115-Sn and energy of the first 115-Sn level

Inventory extension considerations for long-term storage at the nuclear materials storage facility
Los Alamos National Laboratory is in the process of modifying its nuclear materials storage facility to a long-term storage configuration. In support of this effort, we examined technical and administrative means to extend periods between physical inventories. Both the frequency and sample size during a physical inventory could significantly impact required sizing of the non-destructive assay (NDA) laboratory as well as material handling capabilities. Several options are being considered, including (1) treating each storage location as a separate vault, (2) minimizing the number of items returned for quantitative analysis by optimizing the use of in situ confirmatory measurements, and (3) utilizing advanced monitoring technologies. Careful consideration of these parameters should allow us to achieve and demonstrate safe and secure storage while minimizing the impact on facility operations and without having to increase the size of the NDA laboratory beyond that required for anticipated shipping and receiving activities
Epithelial immunomodulation by aerosolized Toll-like receptor agonists prevents allergic inflammation in airway mucosa in mice
Allergic asthma is a chronic inflammatory respiratory disease associated with eosinophilic infiltration, increased mucus production, airway hyperresponsiveness, and airway remodeling. Epidemiologic data reveal that the prevalence of allergic sensitization and associated diseases has increased in the twentieth century. This has been hypothesized to be partly due to reduced contact with microbial organisms (the hygiene hypothesis) in industrialized society. Airway epithelial cells, once considered a static physical barrier between the body and the external world, are now widely recognized as immunologically active cells that can initiate, maintain, and restrain inflammatory responses, such as those that mediate allergic disease. Airway epithelial cells can sense allergens via expression of myriad Toll-like receptors (TLRs) and other pattern-recognition receptors. We sought to determine whether the innate immune response stimulated by a combination of Pam2CSK4 (“Pam2”, TLR2/6 ligand) and a class C oligodeoxynucleotide ODN362 (“ODN”, TLR9 ligand), when delivered together by aerosol (“Pam2ODN”), can modulate the allergic immune response to allergens. Treatment with Pam2ODN 7 days before sensitization to House Dust Mite (HDM) extract resulted in a strong reduction in eosinophilic and lymphocytic inflammation. This Pam2ODN immunomodulatory effect was also seen using Ovalbumin (OVA) and A. oryzae (Ao) mouse models. The immunomodulatory effect was observed as much as 30 days before sensitization to HDM, but ineffective just 2 days after sensitization, suggesting that Pam2ODN immunomodulation lowers the allergic responsiveness of the lung, and reduces the likelihood of inappropriate sensitization to aeroallergens. Furthermore, Pam2 and ODN cooperated synergistically suggesting that this treatment is superior to any single agonist in the setting of allergen immunotherapy

Erosion and deposition on the Pajarito Plateau, New Mexico, and implications for geomorphic responses to late Quaternary climatic changes
The Pajarito Plateau of northern New Mexico contains a rich and diverse record of late Quaternary landscape changes in a variety of geomorphic settings that include gently-sloping mesa tops, steep canyon walls, and canyon bottoms. A broad range of investigations during the past decade, motivated by environmental and seismic hazard concerns, have resulted in examination of the characteristics, stratigraphy, and age of sediments and soils at numerous locations throughout the Plateau. Geochronologic control is provided by >140 radiocarbon dates supplemented by soil characterization and tephrochronology. In this paper we first summarize some of the results of recent and ongoing work on late Quaternary deposits on the Pajarito Plateau, illustrating both the complexity of the geomorphic record and some common elements that have been observed in multiple locations. We then use these observations, in combination with other work in the Southwest, to make some inferences about the local geomorphic response to regional climatic changes. Because the geomorphic and paleoclimatic records are fragmentary, and because the relations between large scale climate changes and local variations in precipitation, vegetation, and geomorphic processes are not fully understood, many uncertainties exist concerning the response of the local landscape to past climatic fluctuations. In addition, variations in local landscape sensitivity related to prior erosional history and spatial variations in vegetation, and the localized nature of many storms, probably contribute to the complexity of the geomorphic record. Nevertheless, the work discussed in this paper suggests a strong relation between regional climatic changes and local geomorphic history, and provides a framework for considering relations between modem processes, the record of past landscape changes, and future erosion and deposition on the Plateau and in surrounding areas

Florbetapir F 18 amyloid PET and 36-month cognitive decline:a prospective multicenter study
This study was designed to evaluate whether subjects with amyloid beta (Aβ) pathology, detected using florbetapir positron emission tomorgraphy (PET), demonstrated greater cognitive decline than subjects without Aβ pathology. Sixty-nine cognitively normal (CN) controls, 52 with recently diagnosed mild cognitive impairment (MCI) and 31 with probable Alzheimer's disease (AD) dementia were included in the study. PET images obtained in these subjects were visually rated as positive (Aβ+) or negative (Aβ−), blind to diagnosis. Fourteen percent (10/69) of CN, 37% (19/52) of MCI and 68% (21/31) of AD were Aβ+. The primary outcome was change in ADAS-Cog score in MCI subjects after 36 months; however, additional outcomes included change on measures of cognition, function and diagnostic status. Aβ+ MCI subjects demonstrated greater worsening compared with Aβ− subjects on the ADAS-Cog over 36 months (5.66±1.47 vs −0.71±1.09, P=0.0014) as well as on the mini-mental state exam (MMSE), digit symbol substitution (DSS) test, and a verbal fluency test (P<0.05). Similar to MCI subjects, Aβ+ CN subjects showed greater decline on the ADAS-Cog, digit-symbol-substitution test and verbal fluency (P<0.05), whereas Aβ+ AD patients showed greater declines in verbal fluency and the MMSE (P<0.05). Aβ+ subjects in all diagnostic groups also showed greater decline on the CDR-SB (P<0.04), a global clinical assessment. Aβ+ subjects did not show significantly greater declines on the ADCS-ADL or Wechsler Memory Scale. Overall, these findings suggest that in CN, MCI and AD subjects, florbetapir PET Aβ+ subjects show greater cognitive and global deterioration over a 3-year follow-up than Aβ− subjects do
Chronic mucocutaneous candidiasis in APECED or thymoma patients correlates with autoimmunity to Th17-associated cytokines
Chronic mucocutaneous candidiasis (CMC) is frequently associated with T cell immunodeficiencies. Specifically, the proinflammatory IL-17A–producing Th17 subset is implicated in protection against fungi at epithelial surfaces. In autoimmune polyendocrinopathy candidiasis ectodermal dystrophy (APECED, or autoimmune polyendocrine syndrome 1), CMC is often the first sign, but the underlying immunodeficiency is a long-standing puzzle. In contrast, the subsequent endocrine features are clearly autoimmune, resulting from defects in thymic self-tolerance induction caused by mutations in the autoimmune regulator (AIRE). We report severely reduced IL-17F and IL-22 responses to both Candida albicans antigens and polyclonal stimulation in APECED patients with CMC. Surprisingly, these reductions are strongly associated with neutralizing autoantibodies to IL-17F and IL-22, whereas responses were normal and autoantibodies infrequent in APECED patients without CMC. Our multicenter survey revealed neutralizing autoantibodies against IL-17A (41%), IL-17F (75%), and/ or IL-22 (91%) in >150 APECED patients, especially those with CMC. We independently found autoantibodies against these Th17-produced cytokines in rare thymoma patients with CMC. The autoantibodies preceded the CMC in all informative cases. We conclude that IL-22 and IL-17F are key natural defenders against CMC and that the immunodeficiency underlying CMC in both patient groups has an autoimmune basis
