    Import 23/08/2017Cílem této bakalářské práce je popsat absolvování odborné praxe ve firmě HS Interactive s.r.o. Praxe byla zaměřena na vývoj mobilní aplikace pro operační systém Android. Aplikace je mobilním klientem pro sociální síť MatchToMe. V úvodu popisuji důvody, které vedly k výběru odborné praxe. Dále se věnuji úkolům, které mi byly zadány s jejich implementací a postupem řešení problémů, které se objevily při vývoji. Závěr práce je věnován zhodnocení získaných zkušeností a dosažených výsledků.Purpose of this bachelor thesis is to describe a professional practice in company HS Interactive s.r.o. Practice was focused on the development of mobile application for the operating system Android. The application is a mobile client for social network MatchToMe. In the introduction I describe reasons that led to the selection of professional practice. Then I describe tasks that I have been awarded with their implementations and process of solution issues that have emerged during development.     Dynamic microRNAome profiles in the developing porcine liver

    <p>Increasing evidence suggests that micro (mi)RNAs play important roles in various biological process. To evaluate the roles of miRNA in the porcine liver, we investigated the dynamic profiles of microRNAomes using liver tissue from pigs during the embryonic period (embryonic day 90), weaning stage (postnatal day 30), and adult stage (7 years old). A total of 186 unique miRNAs were differentially expressed during liver development. We also identified that 17, 13, and 6 miRNAs were specifically abundant at embryonic day 90, postnatal day 30, and at 7 years, respectively. Besides regulating basic cellular roles in development, miRNAs expressed at the three developmental stages also participated in regulating “embryonic liver development,” “early hepatic growth and generating a functioning liver,” and “energy metabolic processes,” respectively. Our study indicates that miRNAs are extensively involved in liver development, and provides a valuable resource for the further elucidation of miRNA regulatory roles during liver development.</p> <p>miRNAs are extensively involved in liver development, especially for these stage-enriched miRNAs.</p

    Bergmann’s rule states that within a species of endotherms smaller individuals are found in warmer conditions, which is consistent for nearly all endotherms, while in ectotherms body size patterns are less consistent. As ectothermic vertebrates, the morphology of amphibians is likely impacted by climatic conditions. Here, we examined latitudinal variation in body size in the ranid frog, <i>Fejervarya limnocharis</i>, based on literature and our own data on mean body size of 3637 individuals from 50 populations and average age of 2873 individuals from 40 populations in China. The results showed that body size was positively correlated with environmental temperature, but not with precipitation. Body size was negatively correlated with latitude among populations in this species, which supported the inverse of Bergmann’s rule. Our findings suggest that a larger body size in low-latitude populations is associated with a longer growing season related to the higher environmental temperatur

    Self-Powered Nanocomposites under an External Rotating Magnetic Field for Noninvasive External Power Supply Electrical Stimulation

    Electrical stimulation in biology and gene expression has attracted considerable attention in recent years. However, it is inconvenient that the electric stimulation needs to be supplied an implanted power-transported wire connecting the external power supply. Here, we fabricated a self-powered composite nanofiber (CNF) and developed an electric generating system to realize electrical stimulation based on the electromagnetic induction effect under an external rotating magnetic field. The self-powered CNFs generating an electric signal consist of modified MWNTs (m-MWNTs) coated Fe<sub>3</sub>O<sub>4</sub>/PCL fibers. Moreover, the output current of the nanocomposites can be increased due to the presence of the magnetic nanoparticles during an external magnetic field is applied. In this paper, these CNFs were employed to replace a bullfrog’s sciatic nerve and to realize the effective functional electrical stimulation. The cytotoxicity assays and animal tests of the nanocomposites were also used to evaluate the biocompatibility and tissue integration. These results demonstrated that this self-powered CNF not only plays a role as power source but also can act as an external power supply under an external rotating magnetic field for noninvasive the replacement of injured nerve

    The expression of P450 genes is regulated by trans-regulatory factors or cis-regulatory elements and influences how endogenous or xenobiotic substances are metabolized in an organism’s tissues. In this study, we showed that overexpression of the cytochrome P450 gene, CYP6CY22, led to resistance to cyantraniliprole in Aphis gossypii. The expression of CYP6CY22 increased in the midgut and remaining carcass of the CyR strain, and after repressing the expression of CYP6CY22, the mortality of cotton aphids increased 2.08-fold after exposure to cyantraniliprole. Drosophila ectopically expressing CYP6CY22 exhibited tolerance to cyantraniliprole and cross-tolerance to xanthotoxin, quercetin, 2-tridecanone, tannic acid, and nicotine. Moreover, transcription factor CF2–II (XM_027994540.2) is transcribed only as the splicing variant isoform CF2–II-AS, which was found to be 504 nucleotides shorter than CF2–II in A. gossypii. RNAi and yeast one-hybrid (Y1H) results indicated that CF2–II-AS positively regulates CYP6CY22 and binds to cis-acting element p (−851/-842) of CYP6CY22 to regulate its overexpression. The above results indicated that CYP6CY22 was regulated by the splicing isoform CF2-II-AS, which will help us further understand the mechanism of transcriptional adaption of cross-tolerance between synthetic insecticides and plant secondary metabolites mediated by P450s
