    Basal metabolic rate as a potential determinant in risk sensitive foraging.

    In foraging, there are many possible determinants of an individual’s sensitivity to risk, such as an animal’s learning or various biological imperatives. However, few studies on risk sensitivity incorporate information about an animal’s metabolism. One measure of metabolic rate is basal metabolic rate (BMR), or the minimum rate at which metabolism must produce energy to maintain homeostasis, which is highly plastic. Therefore, we investigated a potential relationship between risk sensitivity and BMR. This metaanalysis revealed little to no relationship between BMR and risk sensitivity, perhaps because of the high intraspecific variation of each of these traits

    Semi-automated high-throughput fluorescent intercalator displacement-based discovery of cytotoxic DNA binding agents from a large compound library

    High-throughput fluorescent intercalator displacement (HT–FID) was adapted to the semi-automated screening of a commercial compound library containing 60,000 molecules resulting in the discovery of cytotoxic DNA-targeted agents. Although commercial libraries are routinely screened in drug discovery efforts, the DNA binding potential of the compounds they contain has largely been overlooked. HT–FID led to the rapid identification of a number of compounds for which DNA binding properties were validated through demonstration of concentration-dependent DNA binding and increased thermal melting of A/T- or G/C-rich DNA sequences. Selected compounds were assayed further for cell proliferation inhibition in glioblastoma cells. Seven distinct compounds emerged from this screening procedure that represent structures unknown previously to be capable of targeting DNA leading to cell death. These agents may represent structures worthy of further modification to optimally explore their potential as cytotoxic anti-cancer agents. In addition, the general screening strategy described may find broader impact toward the rapid discovery of DNA targeted agents with biological activity

    Stability of the human faecal microbiome in a cohort of adult men

    Characterizing the stability of the gut microbiome is important to exploit it as a therapeutic target and diagnostic biomarker. We metagenomically and metatranscriptomically sequenced the faecal microbiomes of 308 participants in the Health Professionals Follow-Up Study. Participants provided four stool samples—one pair collected 24–72 h apart and a second pair ~6 months later. Within-person taxonomic and functional variation was consistently lower than between-person variation over time. In contrast, metatranscriptomic profiles were comparably variable within and between subjects due to higher within-subject longitudinal variation. Metagenomic instability accounted for ~74% of corresponding metatranscriptomic instability. The rest was probably attributable to sources such as regulation. Among the pathways that were differentially regulated, most were consistently over- or under-transcribed at each time point. Together, these results suggest that a single measurement of the faecal microbiome can provide long-term information regarding organismal composition and functional potential, but repeated or short-term measures may be necessary for dynamic features identified by metatranscriptomics

    High temperature/pressure MAS-NMR for the study of dynamic processes in mixed phase systems

    A new MAS-NMR rotor (the WHiMS rotor) has been developed which can reach pressures of 400 bar at 20 °C or 225 bar at 250 °C. These rotors are ideal for mixed phase systems such as a reaction using a solid catalyst with a liquid/supercritical solvent topped with high pressure gas in the head space. After solid and liquid portions of the sample are loaded, the rotor is capped with an o-ring equipped polymer bushing that snaps into a mating groove in the rotor. The bushings incorporate a check valve into the sealing mechanism which allows for pressurization without mechanical manipulation – they will allow gas to flow in but not out. This WHiMS rotor design has enabled experiments on a wide variety of biotic and abiotic mixed-phase systems. Geochemical systems have also been studied, for example, adsorption and confinement studies of supercritical methane/CO2 in clays and other minerals which display pressure dependent 13C chemical shifts. Example data from other mixed-phase chemical and microbial systems are reported. These include monitoring metabolite conversion of extremophilic bacteria found in subsurface systems at elevated pressures and real-time operando reactions in catalysis systems - with liquid-quality resolution for 1H and 13C NMR spectra

    A Catalog of Spectroscopically Confirmed White Dwarfs from the Sloan Digital Sky Survey Data Release 4

    We present a catalog of 9316 spectroscopically confirmed white dwarfs from the Sloan Digital Sky Survey Data Release 4. We have selected the stars through photometric cuts and spectroscopic modeling, backed up by a set of visual inspections. Roughly 6000 of the stars are new discoveries, roughly doubling the number of spectroscopically confirmed white dwarfs. We analyze the stars by performing temperature and surface gravity fits to grids of pure hydrogen and helium atmospheres. Among the rare outliers are a set of presumed helium-core DA white dwarfs with estimated masses below 0.3 Msun, including two candidates that may be the lowest masses yet found.     Thalamic GABA levels and Occupational Manganese Neurotoxicity: Association with Exposure Levels and Brain MRI

    Excessive occupational exposure to Manganese (Mn) has been associated with clinical symptoms resembling idiopathic Parkinson’s disease (IPD), impairing cognitive and motor functions. Several studies point towards an involvement of the brain neurotransmitter system in Mn intoxication, which is hypothesized to be disturbed prior to onset of symptoms. Edited Magnetic Resonance Spectroscopy (MRS) offers the unique possibility to measure γ-amminobutyric acid (GABA) and other neurometabolites in vivo non-invasively in workers exposed to Mn. In addition, the property of Mn as Magnetic Resonance Imaging (MRI) contrast agent may be used to study Mn deposition in the human brain. In this study, using MRI, MRS, personal air sampling at the working place, work history questionnaires, and neurological assessment (UPDRS-III), the effects of chronic Mn exposure on the thalamic GABAergic system was studied in a group of welders (N = 39) with exposure to Mn fumes in a typical occupational setting. Two subgroups of welders with different exposure levels (Low: N = 26; mean air Mn = 0.13 ± 0.1 mg/m3; High: N = 13; mean air Mn = 0.23 ± 0.18 mg/m3), as well as unexposed control workers (N = 22, mean air Mn = 0.002 ± 0.001 mg/m3) were recruited. The group of welders with higher exposure showed a significant increase of thalamic GABA levels by 45% (p < 0.01, F(1,33) = 9.55), as well as significantly worse performance in general motor function (p < 0.01, F(1,33) = 11.35). However, welders with lower exposure did not differ from the controls in GABA levels or motor performance. Further, in welders the thalamic GABA levels were best predicted by past-12-months exposure levels and were influenced by the Mn deposition in the substantia nigra and globus pallidus. Importantly, both thalamic GABA levels and motor function displayed a non-linear pattern of response to Mn exposure, suggesting a threshold effect

    Discovery of New Ultracool White Dwarfs in the Sloan Digital Sky Survey

    We report the discovery of five very cool white dwarfs in the Sloan Digital Sky Survey (SDSS). Four are ultracool, exhibiting strong collision induced absorption (CIA) from molecular hydrogen and are similar in color to the three previously known coolest white dwarfs, SDSS J1337+00, LHS 3250 and LHS 1402. The fifth, an ultracool white dwarf candidate, shows milder CIA flux suppression and has a color and spectral shape similar to WD 0346+246. All five new white dwarfs are faint (g > 18.9) and have significant proper motions. One of the new ultracool white dwarfs, SDSS J0947, appears to be in a binary system with a slightly warmer (T_{eff} ~ 5000K) white dwarf companion.Comment: 15 pages, 3 figures, submitted to ApJL.     New Experimental Limits on Macroscopic Forces Below 100 Microns

    Results of an experimental search for new macroscopic forces with Yukawa range between 5 and 500 microns are presented. The experiment uses 1 kHz mechanical oscillators as test masses with a stiff conducting shield between them to suppress backgrounds. No signal is observed above the instrumental thermal noise after 22 hours of integration time. These results provide the strongest limits to date between 10 and 100 microns, improve on previous limits by as much as three orders of magnitude, and rule out half of the remaining parameter space for predictions of string-inspired models with low-energy supersymmetry breaking. New forces of four times gravitational strength or greater are excluded at the 95% confidence level for interaction ranges between 200 and 500 microns.Comment: 25 Pages, 7 Figures: Minor Correction

    Our current research focuses on addressing four specific questions. Has the dominant style of volcanic expression within the quadrangle varied in a systematic fashion over time? Does the tectonic deformation within the quadrangle record significant regional patterns that vary spatially or temporally, and if so what are the scales, orientations and sources of the stress fields driving this deformation? If mantle upwelling and downwelling have played a significant role in the formation of Atla Regio and Atalanta Planitia as has been proposed, does the geology of Ganiki Planitia record evidence of northwest-directed lateral mantle flow connecting the two sites? Finally, can integration of the tectonic and volcanic histories preserved within the quadrangle help constrain competing resurfacing models for Venus
