    Hydrogen peroxide metabolism during peroxisome proliferation by fenofibrate

    AbstractFenofibrate, the hypolipidemic drug and peroxisome proliferator, was given to mice (0.23% w/w in the diet) during 1–3 weeks and enzyme activities, H2O2 concentration, and H2O2 production rate were determined. A maximal increase of 150% in liver/body weight ratio was observed after 3 weeks of treatment. Acyl-CoA oxidase, catalase and uricase activities were increased by 712%, 506% and 41% respectively by treatment with fenofibrate. Se- and non Se-glutathione peroxidase and Mn-superoxide dismutase activities were increased by 331 %, 188% and 130% respectively in the liver of 2 weeks-treated mice. CuZn superoxide dismutase activity was not affected by fenofibrate treatment. H2O2 steady-state concentration showed an increase of 89% after 2 weeks of treatment. H2O2 production rates, and the steady-state concentrations of the intermediates HO , R and ROO , calculated using experimental data, were higher in the liver of fenofibrate-treated mice than in control animals. According to our findings, the imbalance between H2O2 production and its degradation by its metabolizing enzymes during peroxisome proliferation, would result in an increased level of H2O2 steady-state concentration, with the resulting oxidative stress which may lead to the generation of oxidative damage and to the induction of liver carcinogenesis

    Cardiovascular autonomic dysfunction and oxidative stress induced by fructose overload in an experimental model of hypertension and menopause

    Background: Metabolic syndrome is characterized by the association of 3 or more risk factors, including: abdominal obesity associated with an excess of abdominal fat, insulin resistance, type 2 diabetes, dyslipidemia and hypertension. Moreover, the prevalence of hypertension and metabolic dysfunctions sharply increases after the menopause. However, the mechanisms involved in these changes are not well understood. Thus, the aim of this study was to assess the effects of fructose overload on cardiovascular autonomic modulation, inflammation and cardiac oxidative stress in an experimental model of hypertension and menopause. Methods: Female SHR rats were divided into (n = 8/group): hypertensive (H), hypertensive ovariectomized (HO) and hypertensive ovariectomized undergoing fructose overload (100 g/L in drinking water) (FHO). Arterial pressure (AP) signals were directly recorded. Cardiac autonomic modulation was evaluated by spectral analysis. Oxidative stress was evaluated in cardiac tissue. Results: AP was higher in the FHO group when compared to the other groups. Fructose overload promoted an increase in body and fat weight, triglyceride concentration and a reduction in insulin sensitivity. IL-10 was reduced in the FHO group when compared to the H group. TNF-α was higher in the FHO when compared to all other groups. Lipoperoxidation was higher and glutathione redox balance was reduced in the FHO group when compared to other groups, an indication of increased oxidative stress. A negative correlation was found between IL-10 and adipose tissue. Conclusion: Fructose overload promoted an impairment in cardiac autonomic modulation associated with inflammation and oxidative stress in hypertensive rats undergoing ovarian hormone deprivation.Fil: Conti, Filipe Fernandes.     Antioxidant levels from different Antarctic fish caught around South Georgia Island and Shag Rocks

    Antarctic fish have been isolated for over several million years in an environment with a very low and constant temperature and high oxygen concentration. In such conditions the oxidative stress might be an important factor affecting their metabolic adaptive strategies. Activity of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx), vitamin E levels and total antioxidant capacity (TRAP) were measured in liver, gill, heart and muscle homogenates of red-blooded (Nototheniidae) and white-blooded (Channichthyidae) Antarctic fish. SOD activity was also measured in blood samples. Gill SOD activity was threefold higher in channichthyids than in nototheniids while CAT and GPx were significantly higher in the gills of channichthyids. The increased SOD activity of channichthyids probably reflects the large PO2 gradient across their gills. The H2O2 produced seems to be preferentially eliminated by diffusion, according to the low levels of CAT and GPx found in the gills of these species. In contrast, blood SOD was about fivefold higher in the latter group, which possesses erythrocytes and thus a much higher oxygen-carrying capacity. CAT activity was always higher in nototheniids except in muscle. However, vitamin E did not show clear differences between families except for the pattern observed in muscle. The higher content of vitamin E in this tissue shown in channichthyids is related to the higher volume density of mitochondria reported for this group, since vitamin E is responsible for preventing membrane lipid peroxidation. Accordingly, TRAP (representative of hydrosoluble antioxidant capacity) was also higher in muscle of channichthyids.     Influence of estimated training status on anti and pro-oxidant activity, nitrite concentration, and blood pressure in middle-aged and older women

    The purpose of this study was to compare the association between anti and pro-oxidant activity, nitrite concentration, and blood pressure (BP) in middle-aged and older women with different levels of estimated training status (TS). The sample consisted of 155 females (50-84 years) who were submitted to a physical examination to evaluate estimated TS through the "Functional Fitness Battery Test," BP measurements, and plasma blood samples to evaluate pro-oxidant and antioxidant activity and nitrite concentrations. Participants were separated by age into a middle-aged group (< 65 years) and an older (≥65 years) group and then subdivided in each group according to TS. Blood biochemistry was similar between groups. On the other hand, protein oxidation was lower in participants with higher TS, independent of age. Older females with higher TS presented higher nitrite concentrations, lower lipoperoxidation, and lower values of BP compared with those with lower TS. Lower GPx activity was observed in participants with higher TS compared with middle-aged with lower TS. Thus, our results suggest that good levels of TS may be associated with lower oxidative stress and higher nitrite concentration and may contribute to maintain normal or reduced blood pressure values.Fil: Jacomini, André M..     Antioxidant status in the aqueous humour of patients with glaucoma associated with exfoliation syndrome

    PURPOSE: To establish the antioxidant status of the aqueous humour in glaucoma associated with exfoliation syndrome (XFG) and to compare it to primary open-angle glaucoma (POAG) and cataract patients. METHODS: Patients were diagnosed with POAG, XFG, or cataract (n=25 for each group). Total reactive antioxidant potential (TRAP) was measured by chemiluminescence. Ascorbic acid levels and the activities of catalase, glutathione peroxidase (GPx), and superoxide dismutase (SOD) were measured spectrophotometrically.ResultsTRAP value was lower in XFG (28+/-2 microM Trolox) than in POAG (55+/-8 microM Trolox; P<0.001). TRAP values in both glaucomas were lower than the cataract value (124+/-5 microM Trolox; P<0.001). A decrease in ascorbic acid was measured in XFG (230+/-20 microM) compared with POAG (415+/-17 microM; P<0.001). Ascorbic acid in both glaucomas was lower than in cataract (720+/-30 microM; P<0.001). A significant increase in GPx was found in XFG (30+/-2 U/ml) compared with POAG (16+/-3 U/ml). GPx activity in both glaucomas was increased when compared with cataracts (6+/-2 U/ml; P<0.001). A significant increase of 67% in SOD activity was observed in the glaucoma group vscataract group (27+/-3 U/ml; P<0.001), but no changes were found between both glaucomas. CONCLUSIONS: The antioxidant status of the aqueous humour may play a role in the pathophysiology of both glaucomas.Fil: Ferreira, Sandra María.
