    A Category Classification Based Safety Risk Assessment Method for Railway Wagon Loading Status

    The identification and control of safety risks in the loading state of goods wagon is one of the important tasks to ensure the safety of goods in transit. In view of the problem that the current risk assessment of transportation schemes is mainly based on manual experience and cannot be quantified, which makes it difficult to accurately determine the safety risk of transportation on the way, a risk assessment method for loading status of goods wagon based on scenario classification was proposed. Firstly, based on a detailed analysis of the safety risk points in various stages of railway freight operations, a SHEL influencing factor model based on scenario classification was constructed. Then, considering the characteristics of railway freight transportation, a fuzzy accident tree model (FTA) of goods wagon loading state risk was constructed, and the fault tree was transformed into a Bayesian network structure according to the mapping algorithm of fuzzy fault tree and Bayesian. Furthermore, a triangular fuzzy membership function was introduced to describe the fault probability of nodes, and a BN based fuzzy fault tree inference algorithm was proposed. Finally, taking a railway station and route transporting coil steel goods in China as an example, this paper explained how to integrate expert knowledge through fault tree and Bayesian network to support railway freight scheme designers in conducting risk quantification assessment of freight wagon loading status

    Attitudes, practices and information needs regarding novel influenza A (H7N9) among employees of food production and operation in Guangzhou, Southern China: a cross-sectional study

    BACKGROUND: As of 30 May 2013, 132 human infections with avian influenza A (H7N9) had been reported in 10 Chinese cities. On 17 May 2013, because a chicken infection with H7 subtype avian influenza virus was detected in Guanzhou, Guangzhou became the 11th city to conduct emergency response operations. The goal of this study was to identify attitudes, practices and information needs among employees of food production and operation in Guangzhou. METHODS: A cross-sectional survey of face-to-face interviews was used during 17–24 June 2013. All adults seeking health examination in Guangzhou Center for Disease Control and Prevention who had lived in Guangzhou for at least 3 months, were engaged in food production and operation, and agreed to participate were interviewed. RESULTS: Of 1,450 participants, 69.72% worried about being infected with the A/H7N9 and 74.41% stated that they had searched for information about A/H7N9. The internet (76.92%), television (67.56%), and newspapers (56.26%) were the main methods of obtaining information; the use of these methods differed significantly by various demographic variables (P < 0.05). More than one-fifth of participants complained that the information was not timely enough (20.28%) and was intentionally concealed by the government (20.76%). Nearly one-third (32.35%) did not believe that the government could control the A/H7N9 epidemic. Most participants (80.76%) reported washing hands more frequently than before, while over one-third (37.17%) stated no longer buying poultry. A total of 84.00% indicated a willingness to receive an A/H7N9 vaccine, and the primary reason for not being willing was concern about safety (58.19%). A history of influenza vaccination and worry about being infected with the A/H7N9 were significantly associated with intention to receive an A/H7N9 vaccine (P < 0.05). CONCLUSIONS: Our findings provide insight into the attitudes and practices of employees of food production and operation 3 months after the first human A/H7N9 case reported in China, and 1 month after infected chickens were identified in Guangzhou. Distrust in the health department should be addressed, and more effort should be made to improve compliance of proper preventive measures to reduce panic among the public. The information needs should be taken into account in the next step of health education

    Identification of CD8+ cytotoxic T lymphocyte epitopes from porcine reproductive and respiratory syndrome virus matrix protein in BALB/c mice

    Twenty-seven nanopeptides derived from the matrix (M) protein of porcine reproductive and respiratory syndrome virus (PRRSV) were screened for their ability to elicit a recall interferon-γ (IFN-γ) response from the splenocytes of BALB/c mice following DNA vaccination and a booster vaccination with recombinant vaccinia virus rWR-PRRSV-M. We identified two peptides (amino acid residues K93FITSRCRL and F57GYMTFVHF) as CD8+ cytotoxic T lymphocyte (CTL) epitopes. These peptides elicited significant numbers of IFN-γ secreting cells, compared with other M nonapeptides and one irrelevant nonapeptide. Bioinformatics analysis showed that the former is an H-2Kd-restricted CTL epitope, and the latter is an H-2Dd-restricted CTL epitope. Multiple amino acid sequence alignment among different PRRSV M sequences submitted to GenBank indicated that these two CTL epitopes are strongly conserved, and they should therefore be considered for further research on the mechanisms of cellular immune responses to PRRSV

    Relating the composition of continental margin surface sediments from the Ross Sea to the Amundsen Sea, West Antarctica, to modern environmental conditions

    Investigating the multiple proxies involving productivity, organic geochemistry, and trace element (TE) enrichment in surface sediments could be used as paleoenvironment archives to gain insights into past and future environmental conditions changes. We present redox-sensitive TEs (Mn, Ni, Cu, U, P, Mo, Co, V, Zn, and Cd), productivity-related proxies (total organic carbon and opal), and total nitrogen and CaCO3 contents of bulk surface sediments of this area. The productivity proxies from the shelf and coastal regions of the Ross and the Amundsen seas showed that higher productivity was affiliated with an area of nutrient-rich deep water upwelling. The upwelling of weakly corrosive deep water may be beneficial for preserving CaCO3, while highly corrosive dense water, if it forms on the shelf near the coastal region (coastal polynya), could limit the preservation of CaCO3 in modern conditions. There were no oxic or anoxic conditions in the study area, as indicated by the enrichment factors of redox-sensitive TEs (Mn, Co, and U). The enrichment factor of Cd, which is redox-sensitive, indicated suboxic redox conditions in sediment environments because of high primary productivity and organic matter preservation/decomposition. The enrichment factors of other redox-sensitive TEs (P, Ni, Cu, V, and Zn) and the correlations between the element/Ti ratio with productivity and nutrient proxies indicated that the organic matter decomposed, and there was massive burial of phytoplankton biomass. There was variation in the enrichment, such that sediments were enriched in P, Mo, and Zn, but depleted in Ni, Cu, and V

    RNAi-directed downregulation of OsBADH2 results in aroma (2-acetyl-1-pyrroline) production in rice (Oryza sativa L.)

    <p>Abstract</p> <p>Background</p> <p>Aromatic rice is popular worldwide because of its characteristic fragrance. Genetic studies and physical fine mapping reveal that a candidate gene (<it>fgr</it>/<it>OsBADH2</it>) homologous to <it>betaine aldehyde dehydrogenase </it>is responsible for aroma metabolism in fragrant rice varieties, but the direct evidence demonstrating the functions of <it>OsBADH2 </it>is lacking. To elucidate the physiological roles of <it>OsBADH2</it>, sequencing approach and RNA interference (RNAi) technique were employed to analyze allelic variation and functions of <it>OsBADH2 </it>gene in aroma production. Semi-quantitative, real-time reverse transcription-polymerase chain reaction (RT-PCR), as well as gas chromatography-mass spectrometry (GC-MS) were conducted to determine the expression levels of <it>OsBADH2 </it>and the fragrant compound in wild type and transgenic <it>OsBADH2</it>-RNAi repression lines, respectively.</p> <p>Results</p> <p>The results showed that multiple mutations identical to <it>fgr </it>allele occur in the 13 fragrant rice accessions across China; <it>OsBADH2 </it>is expressed constitutively, with less expression abundance in mature roots; the disrupted <it>OsBADH2 </it>by RNA interference leads to significantly increased 2-acetyl-1-pyrroline production.</p> <p>Conclusion</p> <p>We have found that the altered expression levels of <it>OsBADH2 </it>gene influence aroma accumulation, and the prevalent aromatic allele probably has a single evolutionary origin.</p

    Antibody Dynamics of 2009 Influenza A (H1N1) Virus in Infected Patients and Vaccinated People in China

    BACKGROUND: To evaluate the risk of the recurrence and the efficiency of the vaccination, we followed-up antibody responses in patients with the 2009 pandemic H1N1 influenza and persons who received the pandemic H1N1 vaccine in Guangzhou China. METHODS: We collected serum samples from 129 patients and 86 vaccinated persons at day 0, 15, 30, 180 after the disease onset or the vaccination, respectively. Antibody titers in these serum samples were determined by haemagglutination inhibition (HI) assay using a local isolated virus strain A/Guangdong Liwan/SWL1538/2009(H1N1). RESULTS: HI antibody positive rate of the patients increased significantly from 0% to 60% at day 15 (χ(2) = 78, P<0.001) and 100% at day 30 (χ(2) = 23, P<0.001), but decreased significantly to 52% at day 180 (χ(2) = 38, P<0.001), while that of vaccinated subjects increased from 0% to 78% at day 15 (χ(2) = 110, P<0.001) and 81% at day 30 (χ(2) = 0.32, P = 0.57), but decreased significantly to 34% at day 180 (χ(2) = 39, P<0.001). Geometric mean titers (GMT) of HI antibodies in positive samples from the patients did not change significantly between day 15 and day 30 (T = 0.92, P = 0.36), but it decreased significantly from 80 at day 30 to 52 at day 180 (T = 4.5, P<0.001). GMT of vaccinated persons increased significantly from 100 at day 15 to 193 at day 30 (T = 4.5, P<0.001), but deceased significantly to 74 at day 180 (T = 5.1, P<0.001). Compared to the patients, the vaccinated subjects showed lower seroconversion rate (χ(2) = 11, P<0.001; χ(2) = 5.9, P = 0.015), but higher GMT (T = 6.0, P<0.001; T = 3.6, P = 0.001) at day 30 and day 180, respectively. CONCLUSION: Vaccination of 2009 influenza A (H1N1) was effective. However, about half or more recovered patients and vaccinated persons might have lost sufficient immunity against the recurrence of the viral infection after half a year. Vaccination or re-vaccination may be necessary for prevention of the recurrence

    Asian dust input in the western Philippine Sea: Evidence from radiogenic Sr and Nd isotopes

    The radiogenic strontium (Sr) and neodymium (Nd) isotope compositions of the detrital fraction of surface and subsurface sediments have been determined to trace sediment provenance and contributions from Asian dust off the east coast of Luzon Islands in the western Philippine Sea. The Sr and Nd isotope compositions have been very homogenous near the east coast of the Luzon Islands during the latest Quaternary yielding relatively least radiogenic Sr (87Sr/86Sr = 0.70453 to 0.70491) and more radiogenic Nd isotope compositions (εNd(0) = +5.3 to +5.5). These isotope compositions are similar to Luzon rocks and show that these sediments were mainly derived from the Luzon Islands. In contrast, the Sr and Nd isotope compositions of sediments on the Benham Rise and in the Philippine Basin are markedly different in that they are characterized by overall more variable and more radiogenic Sr isotope compositions (87Sr/86Sr = 0.70452 to 0.70723) and less radiogenic Nd isotope compositions (εNd(0) = −5.3 to +2.4). The Sr isotope composition in the Huatung Basin is intermediate between those of the east coast of Luzon and Benham Rise, but shows the least radiogenic Nd isotope compositions. The data are consistent with a two end-member mixing relationship between Luzon volcanic rocks and eolian dust from the Asian continent, which is characterized by highly radiogenic Sr and unradiogenic Nd isotope compositions. The results show that Asian continental dust contributes about 10–50% of the detrital fraction of the sediments on Benham Rise in the western Philippine Sea, which offers the potentials to reconstruct the climatic evolution of eastern Asia from these sediments and compare this information to the records from the central and northern Pacific
