Inhibition of PaCaMKII-E isoform in the dorsal unpaired median neurosecretory cells of cockroach reduces nicotine- and clothianidin-induced currents
Cellular responses to Ca(2+) require intermediary proteins such as calcium/calmodulin-dependent protein kinase II (CaMKII), which transduces the signal into downstream effects. We recently demonstrated that the cockroach genome encodes five different CaMKII isoforms, and only PaCaMKII-E isoform is specifically expressed in the dorsal unpaired median neurosecretory cells. In the present study, using antisense oligonucleotides, we demonstrated that PaCaMKII-E isoform inhibition reduced nicotine-induced currents through alpha-bungarotoxin-sensitive and -insensitive nicotinic acetylcholine receptor subtypes. Specifically, PaCaMKII-E isoform is sufficient to repress nicotinic current amplitudes as a result of its depression by antisense oligonucleotides. Similar results were found using the neonicotinoid insecticide clothianidin, which acted as a full agonist of dorsal unpaired median neuron nicotinic acetylcholine receptors. Clothianidin current amplitudes are strongly reduced under bath application of PaCaMKII-E antisense oligonucleotides but no significant results are found with alpha-bungarotoxin co-applied, demonstrating that CaMKII-E isoform affects nicotine currents through alpha-bungarotoxin-sensitive and -insensitive receptor subtypes whereas clothianidin currents are reduced via alpha-bungarotoxin-insensitive receptors. In addition, we found that intracellular calcium increase induced by nicotine and clothianidin were reduced by PaCaMKII-E antisense oligonucleotides, demonstrating that intracellular calcium increase induced by nicotine and clothianidin are affected by PaCaMKII-E inhibition. Gene expression of key regulators of mitochondrial biogenesis is sex dependent in mice with growth hormone receptor deletion in liver
Mitochondrial biogenesis is an essential process for cell viability. Mice with disruption of the growth hormone receptor (GHR) gene (Ghr gene) in the liver (LiGHRKO), in contrast to long-lived mice with global deletion of the Ghr gene (GHRKO), are characterized by lack of improved insulin sensitivity and severe hepatic steatosis. Tissue-specific disruption of the GHR in liver results in a mouse model with dramatically altered GH/IGF1 axis. We have previously shown increased levels of key regulators of mitochondrial biogenesis in insulin-sensitive GHRKO mice. The aim of the present study is to assess, using real-time PCR, the gene expression of key regulators of mitochondrial biogenesis (Pgc1 alpha, Ampk, Sirt1, Nrf2 and Mfn2) and a marker of mitochondrial activity (CoxIV) in brains, kidneys and livers of male and female LiGHRKO and wild-type (WT) mice. There were significant differences between males and females. In the brain, expression of Pgc1 alpha, Ampk, Sirt1, Nrf2 and Mfn2 was lower in pooled females compared to pooled males. In the kidneys, expression of Ampk and Sirt1 was also lower in female mice. In the liver, no differences between males and females were observed. Sexual dimorphism may play an important role in regulating the biogenesis of mitochondria

Orbital properties of an unusually low-mass sdB star in a close binary system with a white dwarf
We have used 605 days of photometric data from the Kepler spacecraft to study KIC 6614501, a close binary system with an orbital period of 0.157 497 47(25) days (3.779 939 h), that consists of a low-mass subdwarf B (sdB) star and a white dwarf (WD). As seen in many other similar systems, the gravitational field of the WD produces an ellipsoidal deformation of the sdB which appears in the light curve as a modulation at two times the orbital frequency. The ellipsoidal deformation of the sdB implies that the system has a maximum inclination of ∼40°, with i ≈ 20° being the most likely. The orbital radial velocity (RV) of the sdB star is high enough to produce a Doppler beaming effect with an amplitude of 432 ± 5 ppm, clearly visible in the folded light curve. The photometric amplitude that we obtain, K1 = 85.8 km s-1, is ∼12 per cent less than the spectroscopic RV amplitude of 97.2 ± 2.0 km s-1. The discrepancy is due to the photometric contamination from a close object at about 5 arcsec north-west of KIC 6614501, which is difficult to remove. The atmospheric parameters of the sdB star, Teff = 23 700 ± 500 K and log g = 5.70 ± 0.10, imply that it is a rare object below the extreme horizontal branch (EHB), similar to HD 188112. The comparison with different evolutionary tracks suggests a mass between ∼0.18 and ∼0.25 M⊙, too low to sustain core helium burning. If the mass was close to 0.18-0.19 M⊙, the star could be already on the final He-core WD cooling track. A higher mass, up to ∼0.25 M⊙, would be compatible with a He-core WD progenitor undergoing a cooling phase in a H-shell flash loop. A third possibility, with a mass between ∼0.32 and ∼0.40 M⊙, cannot be excluded and would imply that the sdB is a ‘normal\u27 (but with an unusually low mass) EHB star burning He in its core. In all these different scenarios, the system is expected to merge in less than 3.1 Gyr due to gravitational wave radiation
A pulsation zoo in the hot subdwarf B star KIC 10139564 observed by Kepler
We present our analyses of 15 months of Kepler data on KIC 10139564. We detected 57 periodicities with a variety of properties not previously observed all together in one pulsating subdwarf B (sdB) star. Ten of the periodicities were found in the low-frequency region, and we associate them with nonradial g modes. The other periodicities were found in the high-frequency region, which are likely p modes. We discovered that most of the periodicities are components of multiplets with a common spacing. Assuming that multiplets are caused by rotation, we derive a rotation period of 25.6 ± 1.8 d. The multiplets also allow us to identify the pulsations to an unprecedented extent for this class of pulsator. We also detect l ≥ 2 multiplets, which are sensitive to the pulsation inclination and can constrain limb darkening via geometric cancellation factors. While most periodicities are stable, we detected several regions that show complex patterns. Detailed analyses showed that these regions are complicated by several factors. Two are combination frequencies that originate in the super-Nyquist region and were found to be reflected below the Nyquist frequency. The Fourier peaks are clear in the super-Nyquist region, but the orbital motion of Kepler smears the Nyquist frequency in the barycentric reference frame and this effect is passed on to the sub-Nyquist reflections. Others are likely multiplets but unstable in amplitudes and/or frequencies. The density of periodicities also makes KIC 10139564 challenging to explain using published models. This menagerie of properties should provide tight constraints on structural models, making this sdB star the most promising for applying asteroseismology.
To support our photometric analysis, we have obtained spectroscopic radial-velocity measurements of KIC 10139564 using low-resolution spectra in the Balmer-line region. We did not find any radial-velocity variation. We used our high signal-to-noise average spectrum to improve the atmospheric parameters of the sdB star, deriving Teff = 31 859 K and log g = 5.673 dex
Expansion of the Tetragonal Magnetic Phase with Pressure in the Iron Arsenide Superconductor Ba₁₋ₓKₓFe₂As₂
In the temperature-concentration phase diagram of most iron-based superconductors, antiferromagnetic order is gradually suppressed to zero at a critical point, and a dome of superconductivity forms around that point. The nature of the magnetic phase and its fluctuations is of fundamental importance for elucidating the pairing mechanism. In Ba1-xKxFe2As2 and Ba1-xNaxFe2As2, it has recently become clear that the usual stripelike magnetic phase, of orthorhombic symmetry, gives way to a second magnetic phase, of tetragonal symmetry, near the critical point, in the range from x = 0.24 to x=0.28 for Ba1-xKxFe2As2. In a prior study, an unidentified phase was discovered for x \u3c 0.24 but under applied pressure, whose onset was detected as a sharp anomaly in the resistivity. Here we report measurements of the electrical resistivity of Ba1-xKxFe2As2 under applied hydrostatic pressures up to 2.75 GPa, for x = 0.22, 0.24, and 0.28. The critical pressure above which the unidentified phase appears is seen to decrease with increasing x and vanish at x = 0.24, thereby linking the pressure-induced phase to the tetragonal magnetic phase observed at ambient pressure. In the temperature-concentration phase diagram of Ba1-xKxFe2As2, we find that pressure greatly expands the tetragonal magnetic phase, while the stripelike phase shrinks. This reveals that pressure may be a powerful tuning parameter with which to explore the interplay between magnetism and superconductivity in this material
Split or Steal? Cooperative Behavior When the Stakes Are Large
We examine cooperative behavior when large sums of money are at stake, using data from the television game show Golden Balls. At the end of each episode, contestants play a variant on the classic prisoner's dilemma for large and widely ranging stakes averaging over $20,000. Cooperation is surprisingly high for amounts that would normally be considered consequential but look tiny in their current context, what we call a “big peanuts” phenomenon. Utilizing the prior interaction among contestants, we find evidence that people have reciprocal preferences. Surprisingly, there is little support for conditional cooperation in our sample. That is, players do not seem to be more likely to cooperate if their opponent might be expected to cooperate. Further, we replicate earlier findings that males are less cooperative than females, but this gender effect reverses for older contestants because men become increasingly cooperative as their age increases
