Application of hybrid binomial Langevin-multiple mapping conditioning method to reacting jet flow
The hybrid binomial Langevin-MMC (Multiple Mapping Conditioning) method combines the advantages of the binomial Langevin and MMC models in a consistent manner to overcome difficulties in each. The binomial Langevin method provides joint velocity-scalar statistics, but the treatment of scalars is complex since specification of the bounds is not trivial. The MMC method is capable of dealing with the mixing of any number of scalars, but it can be difficult to specify coefficients involving averages of the scalars and the introduced reference space. The difficulties are overcome by using the velocity statistics from the binomial Langevin model to obtain the reference variable for MMC and, subsequently, the mixing of MMC scalars is performed in a manner that minimises the difference between the mixture fractions for each submodel. The current work expands past studies of NO conversion in a mixing layer to include a study of the Sandia D Flame in preparation for the application to more complex combustion phenomena. Chemical Mutagenesis for the Modulation and Study of Protein Function and Dysfunction within Alzheimer’s Disease
Chemical mutagenesis is defined as the post-expression interconversion of amino-acid (AA) sidechains. This approach enabled the first ever point-mutations in proteins and has seen a resurgence of interest over the last two decades. This thesis reports on two disparate applications of chemical mutagenesis within the context of Alzheimer’s disease (AD). First, exploring the capabilities of utilizing chemical mutagenesis to enable a structure-activity relationship study on a single-domain antibody (SdAb) beyond the 20 canonical AAs. The starting SdAb was designed to target the Amyloid-b peptide (Ab) and has a nascent ability to inhibit its aggregation, a key pathological process in AD. Utilizing the synthetically versatile non-canonical AA dehydroalanine (Dha), we explore diverse sidechains at key residues along the binding loop of the SdAb and identify a non-canonical sidechain that potently enhances the inhibition of Ab’s primary nucleation. The second application focuses on the other major protein implicated in the pathology of AD: Tau. In AD, tau aggregates into ordered amyloid fibrils and contains a plethora of post-translational modifications (PTMs), many of which are speculated to have an impact on the pathogenesis of AD as well as other so-called tauopathies. In order to facilitate the accurate study of tau PTMs, an easily accessible method for their site- selective installation is needed. To this end, we again explore the utilisation of Dha to install accurate PTM mimetics for phosphorylation, lysine acetylation, and lysine dimethylation at diverse sites along tau. This approach produced homogenous products that accurately recapitulated known behaviours and allowed for the initial characterisation of poorly investigated PTMs. The characterisation of S199 phosphorylation, a known AD associated modification, through this approach provides the first evidence for its potential role in regulating tau’s primary function of microtubule polymerisation and stabilisation
Urheilijan markkina-arvon kehittäminen Twitterissä
Tämän opinnäytetyön tarkoituksena on tutkia suomalaisten pääsarjatason urheilijoiden Twitter-käyttäytymistä. Tavoitteena on selvittää naisten ja miesten sekä urheilulajien välisiä eroja. Tutkimuskohteena ovat jalkapallon, jääkiekon, koripallon, lentopallon, pesäpallon ja salibandyn naisten ja miesten pääsarjatasojen urheilijat.
Aineistoa varten kerättiin 12 pääsarjatason kaikkien Twitterissä olevien urheilijoiden tviitit vuoden ajalta Futusome Oy:n avulla. Lopullista aineistoa varten tehtiin harkittu otos, johon valittiin neljä viikon jaksoa jokaiselta vuodenajalta, jotta jokaisella urheilulajilla olisi tasapuoliset lähtökohdat. Näiden viikkojen sisällä tehdyt tviitit luokiteltiin STAR-mallin mukaan. STAR-mallia muokattiin suomalaisen some-kulttuurin mukaiseksi. STAR-malli koostui neljästä eri tekstin lajityypistä, eli tarinankerronnasta, keskustelun herättäjistä, viihteestä ja reagoinnista.
Salibandyn tviittimäärät olivat muita urheilulajeja paljon suuremmat, mutta siinä missä salibandy eroaa muista positiivisella tavalla, lentopallo taas eroaa huomattavan vähäisellä tviittimäärällään. Myös sarjakohtaisesti oli suuria eroja, jotkin joukkueet ovat toisia paljon aktiivisempia, kuten esimerkiksi naisten koripalloliigan Espoo United tuotti 85 % koko sarjan tviiteistä. Lajityyppejä verrattaessa reagointi oli sekä naisten että miesten eniten käytetty lajityyppi. Tämä johtui pitkälti siitä, että vastaaminen toisen henkilön tviittiin laskettiin pääasiassa reagointiin. Naiset käyttivät enemmän tarinankerronta-lajityyppiä kuin miehet, kun taas miehet käyttivät naisia enemmän viihde-lajityyppiä.
Suomalaiset urheilijat pystyisivät pienilläkin muutoksilla parantamaan markkina-arvoaan Twitterissä, sillä monipuolisella tviittaamisella ja jokaista tekstin lajityyppiä käyttämällä urheilija pystyisi helposti nostamaan markkina-arvoaan. Urheiluseurojen pitäisi omalta osaltaan myös panostaa enemmän urheilijoidensa sosiaalisten medioiden käyttöön. Jotta Suomessa päästäisiin muualla maailmassa nähtävään fanituskulttuuriin ja siihen reagointi-malliin, joka ulkomaisilla urheilijoilla sosiaalisissa medioissa on, voitaisiin urheilulajien sekä urheilijoiden näkyvyyttä ja tunnettuutta parantaa monipuolisilla maksuttomien tv-kanavien urheilulähetyksillä ja niihin panostamisella.The purpose of this thesis is to find how Finnish athletes use Twitter. The aim is to find differences between genders and also between sports. The subjects in this study are the athletes of Finnish men and women leagues of basketball, Finnish baseball, floorball, football, ice hockey and volleyball.
For the material Futusome Oy gathered tweets from the athletes of the 12 leagues between October 2015 and October 2016. For the final material four weeks were selected and all of them were picked from different seasons so that every sport had equal starting point. All tweets were categorized by the STAR model. STAR model was edited so that it is suitable for Finnish culture. STAR model includes four types of writing, storytelling, trigger, amusement and reaction.
Based on the findings, floorball players tweet much more than any other athletes, on the other hand volleyball players tweets much less than sports in the middle. Espoo United tweets 85 % of the women´s basketball league, so there are major differences in the activity of tweeting in leagues. According to the STAR model categorization reaction was the most used type of writing. After reaction women used storytelling more than men and on the other hand men used amusement more than women.
Finnish athletes could easily improve their market value by using diverse tweeting and using all types of writing from the STAR model. Sports clubs should invest in their athletes´ social media more. In order to reach a similar fan culture than for example in the United States, Finnish television sports selection could be upgraded and the team sports and their athletes highlighted more
