    Oral mucosal progenitor cell clones resist In Vitro myogenic differentiation

    Abstract Progenitor cells derived from the oral mucosa lamina propria (OMLP-PCs) demonstrate an ability to differentiate into tissue lineages removed from their anatomical origin. This clonally derived population of neural-crest cells have demonstrated potential to differentiate along mesenchymal and neuronal cell lineages. Objective Significant efforts are being made to generate functioning muscle constructs for use in research and clinical tissue engineering. In this study we aimed to determine the myogenic properties of clonal populations of expanded OMLP-PCs. Design PCs were subject to several in vitro culture conditions in an attempt to drive myogenic conversion. Methodologies include use of demethylation gene-modifying reagents, mechanical conditioning of tissue culture substrates, tuneable polyacrylamide gels and a 3-dimensional construct as well as published myogenic media compositions.     Routine clinical motion analysis : comparison of a bespoke real-time protocol to current clinical methods

    Motion analysis (MA) hardware has recently become more accessible; however, protocols have not developed in conjunction. Routine clinical assessment mostly relies on unreliable observational methods. This study aimed to develop an MA protocol for routine clinical use and compare kinematics and reliability to the gold-standard. Ten participants completed 10 over ground walks with a comprehensive marker set (bespoke and gold-standard). Inter/intra-assessor reliability was also compared. Results demonstrated comparable kinematics. Reliability of the bespoke model was lower than the gold standard but higher than observational methods. The bespoke model can be recommended for routine clinical use to assess patient progress and function

    How Different Are Threshold and Other Specified Feeding and Eating Disorders? Comparing Severity and Treatment Outcome

    Background: Other Specified Feeding and Eating Disorders (OSFED) are characterized by less frequent symptoms or symptoms that do not meet full criteria for another eating disorder. Despite its high prevalence, limited research has examined differences in severity and treatment outcome among patients with OSFED compared to threshold EDs [Anorexia Nervosa (AN), Bulimia Nervosa (BN), and Binge Eating Disorder (BED)]. The purpose of the current study was to examine differences in clinical presentation and treatment outcome between a heterogenous group of patients with OSFED or threshold EDs. Method: Patients with threshold EDs (AN = 42, BN = 50, BED = 14) or OSFED (n = 66) presenting for eating disorder treatment completed self-report questionnaires at intake and discharge to assess eating disorder symptoms, depression symptoms, impairment, and self-esteem. Results: At intake, OSFED patients showed lower eating concerns compared to patients with BN, but similar levels compared to AN and BED. The OSFED group showed higher restraint symptoms compared to BED, and similar restraint to AN and BN. Global symptoms as well as shape and weight concerns were similar between OSFED and threshold ED groups. There were no differences between diagnostic groups in self-esteem, depression scores, or symptom change from intake to discharge. Discussion: Our findings suggest that individuals with OSFED showed largely similar ED psychopathology and similar decreases in symptoms across treatment as individuals diagnosed with threshold EDs. Taken together, findings challenge the idea that OSFED is less severe and more resistant to treatment than threshold EDs

    Red nuggets grow inside-out: evidence from gravitational lensing

    We present a new sample of strong gravitational lens systems where both the foreground lenses and background sources are early-type galaxies. Using imaging from HST/ACS and Keck/NIRC2, we model the surface brightness distributions and show that the sources form a distinct population of massive, compact galaxies at redshifts 0.4≲z≲0.70.4 \lesssim z \lesssim 0.7, lying systematically below the size-mass relation of the global elliptical galaxy population at those redshifts. These may therefore represent relics of high-redshift red nuggets or their partly-evolved descendants. We exploit the magnifying effect of lensing to investigate the structural properties, stellar masses and stellar populations of these objects with a view to understanding their evolution. We model these objects parametrically and find that they generally require two S\'ersic components to properly describe their light profiles, with one more spheroidal component alongside a more envelope-like component, which is slightly more extended though still compact. This is consistent with the hypothesis of the inside-out growth of these objects via minor mergers. We also find that the sources can be characterised by red-to-blue colour gradients as a function of radius which are stronger at low redshift -- indicative of ongoing accretion -- but that their environments generally appear consistent with that of the general elliptical galaxy population, contrary to recent suggestions that these objects are predominantly associated with clusters.Comment: 21 pages; accepted for publication in MNRA
