    More Americans are eating hamburgers more well-done than in the past, according to national surveys. This change reduced the risk of E. coli O157:H7 infection by an estimated 4.6 percent and reduced associated medical costs and productivity losses by an estimated $7.4 million annually. In a 1996 survey, respondents who were more concerned about the risk of foodborne illness cooked and ordered hamburgers more well-done than those who were less concerned.     Distinct sub-second dopamine signaling in dorsolateral striatum measured by a genetically-encoded fluorescent sensor

    The development of genetically encoded dopamine sensors such as dLight has provided a new approach to measuring slow and fast dopamine dynamics both in brain slices and in vivo, possibly enabling dopamine measurements in areas like the dorsolateral striatum (DLS) where previously such recordings with fast-scan cyclic voltammetry (FSCV) were difficult. To test this, we first evaluated dLight photometry in mouse brain slices with simultaneous FSCV and found that both techniques yielded comparable results, but notable differences in responses to dopamine transporter inhibitors, including cocaine. We then used in vivo fiber photometry with dLight in mice to examine responses to cocaine in DLS. We also compared dopamine responses during Pavlovian conditioning across the striatum. We show that dopamine increases were readily detectable in DLS and describe transient dopamine kinetics, as well as slowly developing signals during conditioning. Overall, our findings indicate that dLight photometry is well suited to measuring dopamine dynamics in DLS

    Growing a Sustainable Biofuels Industry: Economics, Environmental Considerations, and the Role of the Conservation Reserve Program

    Biofuels are expected to be a major contributor to renewable energy in the coming decades under the Renewable Fuel Standard (RFS). These fuels have many attractive properties including the promotion of energy independence, rural development, and the reduction of national carbon emissions. However, several unresolved environmental and economic concerns remain. Environmentally, much of the biomass is expected to come from agricultural expansion and/or intensification, which may greatly affect the net environmental impact, and economically, the lack of a developed infrastructure and bottlenecks along the supply chain may affect the industry\u27s economic vitality. The approximately 30 million acres (12 million hectares) under the Conservation Reserve Program (CRP) represent one land base for possible expansion. Here, we examine the potential role of the CRP in biofuels industry development, by (1) assessing the range of environmental effects on six end points of concern, and (2) simulating differences in potential industry growth nationally using a systems dynamics model. The model examines seven land-use scenarios (various percentages of CRP cultivation for biofuel) and five economic scenarios (subsidy schemes) to explore the benefits of using the CRP. The environmental assessment revealed wide variation in potential impacts. Lignocellulosic feedstocks had the greatest potential to improve the environmental condition relative to row crops, but the most plausible impacts were considered to be neutral or slightly negative. Model simulations revealed that industry growth was much more sensitive to economic scenarios than land-use scenarios—similar volumes of biofuels could be produced with no CRP as with 100% utilization. The range of responses to economic policy was substantial, including long-term market stagnation at current levels of first-generation biofuels under minimal policy intervention, or RFS-scale quantities of biofuels if policy or market conditions were more favorable. In total, the combination of the environmental assessment and the supply chain model suggests that large-scale conversion of the CRP to row crops would likely incur a significant environmental cost, without a concomitant benefit in terms of biofuel production

    Storm Gloria: sea state evolution based on in situ measurements and modeled data and its impact on extreme values

    Storm Gloria, generated on January 17th, 2020 in the Eastern North Atlantic, crossed the Iberian Peninsula and impacted the Western Mediterranean during the following days. The event produced relevant damages on the coast and the infrastructures at the Catalan-Balearic Sea, due to extraordinary wind and wave fields, concomitant with anomalously intense rain and ocean currents. Puertos del Estado (the Spanish holding of harbors) has developed and operates a complex monitoring and forecasting system (PORTUS System), in collaboration with the Spanish Met Office (AEMET). The present work shows how Gloria was correctly forecasted by this system, alerts were properly issued (with special focus to the ports), and the buoys were able to monitor the sea state conditions during the event, measuring several new records of significant wave height and exceptional high mean wave periods. The paper describes, in detail, the dynamic evolution of the atmospheric conditions, and the sea state during the storm. It is by means of the study of both in situ and modeled PORTUS data, in combination with the AEMET weather forecast system results. The analysis also serves to place this storm in a historical context, showing the exceptional nature of the event, and to identify the specific reasons why its impact was particularly severe. The work also demonstrates the relevance of the PORTUS System to warn, in advance, the main Spanish Ports. It prevents accidents that could result in fatal casualties. To do so, the wave forecast warning performance is analyzed, making special focus on the skill score for the different horizons. Furthermore, it is demonstrated how a storm of this nature results in the need of changes on the extreme wave analysis for the area. It impacts all sorts of design activities at the coastline. The paper studies both how this storm fits into existing extreme analysis and how these should be modified in the light of this particular single event. This work is the first of a series of papers to be published on this issue. They analyze, in detail, other aspects of the event, including evolution of sea level and description of coastal damages

    CLASH: Mass Distribution in and around MACS J1206.2-0847 from a Full Cluster Lensing Analysis

    We derive an accurate mass distribution of the galaxy cluster MACS J1206.2-0847 (z=0.439) from a combined weak-lensing distortion, magnification, and strong-lensing analysis of wide-field Subaru BVRIz' imaging and our recent 16-band Hubble Space Telescope observations taken as part of the Cluster Lensing And Supernova survey with Hubble (CLASH) program. We find good agreement in the regions of overlap between several weak and strong lensing mass reconstructions using a wide variety of modeling methods, ensuring consistency. The Subaru data reveal the presence of a surrounding large scale structure with the major axis running approximately north-west south-east (NW-SE), aligned with the cluster and its brightest galaxy shapes, showing elongation with a \sim 2:1 axis ratio in the plane of the sky. Our full-lensing mass profile exhibits a shallow profile slope dln\Sigma/dlnR\sim -1 at cluster outskirts (R>1Mpc/h), whereas the mass distribution excluding the NW-SE excess regions steepens further out, well described by the Navarro-Frenk-White form. Assuming a spherical halo, we obtain a virial mass M_{vir}=(1.1\pm 0.2\pm 0.1)\times 10^{15} M_{sun}/h and a halo concentration c_{vir} = 6.9\pm 1.0\pm 1.2 (\sim 5.7 when the central 50kpc/h is excluded), which falls in the range 4 <7 of average c(M,z) predictions for relaxed clusters from recent Lambda cold dark matter simulations. Our full lensing results are found to be in agreement with X-ray mass measurements where the data overlap, and when combined with Chandra gas mass measurements, yield a cumulative gas mass fraction of 13.7^{+4.5}_{-3.0}% at 0.7Mpc/h (\approx 1.7r_{2500}), a typical value observed for high mass clusters.Comment: Accepted by ApJ (30 pages, 17 figures), one new figure (Figure 10) added, minor text changes; a version with high resolution figures available at http://www.asiaa.sinica.edu.tw/~keiichi/upfiles/MACS1206/ms_highreso.pd

    Distinctive nuclear zone for RAD51-mediated homologous recombinational DNA repair

    Genome-based functions are inseparable from the dynamic higher-order architecture of the cell nucleus. In this context, the repair of DNA damage is coordinated by precise spatiotemporal controls that target and regulate the repair machinery required to maintain genome integrity. However, the mechanisms that pair damaged DNA with intact template for repair by homologous recombination (HR) without illegitimate recombination remain unclear. This report highlights the intimate relationship between nuclear architecture and HR in mammalian cells. RAD51, the key recombinase of HR, forms spherical foci in S/G2 phases spontaneously. Using super-resolution microscopy, we show that following induction of DNA double-strand breaks RAD51 foci at damaged sites elongate to bridge between intact and damaged sister chromatids; this assembly occurs within bundle-shaped distinctive nuclear zones, requires interactions of RAD51 with various factors, and precedes ATP-dependent events involved the recombination of intact and damaged DNA. We observed a time-dependent transfer of single-stranded DNA overhangs, generated during HR, into such zones. Our observations suggest that RAD51-mediated homologous pairing during HR takes place within the distinctive nuclear zones to execute appropriate recombination

    Air Pollution–Associated Changes in Lung Function among Asthmatic Children in Detroit

    In a longitudinal cohort study of primary-school–age children with asthma in Detroit, Michigan, we examined relationships between lung function and ambient levels of particulate matter ≤ 10 μm and ≤ 2.5 μm in diameter (PM(10) and PM(2.5)) and ozone at varying lag intervals using generalized estimating equations. Models considered effect modification by maintenance corticosteroid (CS) use and by the presence of an upper respiratory infection (URI) as recorded in a daily diary among 86 children who participated in six 2-week seasonal assessments from winter 2001 through spring 2002. Participants were predominantly African American from families with low income, and > 75% were categorized as having persistent asthma. In both single-pollutant and two-pollutant models, many regressions demonstrated associations between higher exposure to ambient pollutants and poorer lung function (increased diurnal variability and decreased lowest daily values for forced expiratory volume in 1 sec) among children using CSs but not among those not using CSs, and among children reporting URI symptoms but not among those who did not report URIs. Our findings suggest that levels of air pollutants in Detroit, which are above the current National Ambient Air Quality Standards, adversely affect lung function of susceptible asthmatic children
