    A Reputation and Trust Management Broker Framework for Web Applications

    This paper presents a distributed reputation and trust management framework that addresses the challenges of eliciting, evaluating and propagating reputation for web applications. We propose a broker framework where every service user is associated with a broker who may represent multiple users. A broker collects for its users the distributed reputation ratings about any web service. In return, a user provides its broker the transaction rating after every transaction with any service in order to build up the reputation database on all services. In addition, brokers form a trust network where they exchange and collect reputation data about services. By delegating trust management to brokers, individual users only need to ask their brokers about the reputation of a service before any transaction with a server. The only overhead for a user is the responsibility to share the reputation feedback with its broker. We present the distributed reputation and trust management framework and show the performance of the system by simulations. 1

    Development and Validation of a Prognostic Model for Esophageal Adenocarcinoma Based on Necroptosis-Related Genes

    Necroptosis is a newly developed cell death pathway that differs from necrosis and apoptosis; however, the potential mechanism of necroptosis-related genes in EAC and whether they are associated with the prognosis of EAC patients remain unclear. We obtained 159 NRGs from the Kyoto Encyclopedia of Genes and Genomes (KEGG) and performed differential expression analysis of the NRGs in 9 normal samples and 78 EAC tumor samples derived from The Cancer Genome Atlas (TCGA). Finally, we screened 38 differentially expressed NRGs (DE-NRGs). The results of the GO and KEGG analyses indicated that the DE-NRGs were mainly enriched in the functions and pathways associated with necroptosis. Protein interaction network (PPI) analysis revealed that TNF, CASP1, and IL-1B were the core genes of the network. A risk score model based on four DE-NRGs was constructed by Least Absolute Shrinkage and Selection Operator (LASSO) regression, and the results showed that the higher the risk score, the worse the survival. The model achieved more efficient diagnosis compared with the clinicopathological variables, with an area under the receiver operating characteristic (ROC) curve of 0.885. The prognostic value of this model was further validated using Gene Expression Omnibus (GEO) datasets. Gene set enrichment analyses (GSEA) demonstrated that several metabolism-related pathways were activated in the high-risk population. Single-sample GSEA (ssGSEA) provided further confirmation that this prognostic model was remarkably associated with the immune status of EAC patients. Finally, the nomogram map exhibited a certain prognostic prediction efficiency, with a C-index of 0.792 and good consistency. Thus, the prognostic model based on four NRGs could better predict the prognosis of EAC and help to elucidate the mechanism of necroptosis-related genes in EAC, which can provide guidance for the target prediction and clinical treatment of EAC patients

    Glutamate-releasing BEST1 channel is a new target for neuroprotection against ischemic stroke with wide time window

    Many efforts have been made to understand excitotoxicity and develop neuroprotectants for the therapy of ischemic stroke. The narrow treatment time window is still to be solved. Given that the ischemic core expanded over days, treatment with an extended time window is anticipated. Bestrophin 1 (BEST1) belongs to a bestrophin family of calcium-activated chloride channels. We revealed an increase in neuronal BEST1 expression and function within the peri-infarct from 8 to 48 h after ischemic stroke in mice. Interfering the protein expression or inhibiting the channel function of BEST1 by genetic manipulation displayed neuroprotective effects and improved motor functional deficits. Using electrophysiological recordings, we demonstrated that extrasynaptic glutamate release through BEST1 channel resulted in delayed excitotoxicity. Finally, we confirmed the therapeutic efficacy of pharmacological inhibition of BEST1 during 6–72 h post-ischemia in rodents. This delayed treatment prevented the expansion of infarct volume and the exacerbation of neurological functions.     Surface Magnesium Substitution at Spinel Lithium Manganate 8a Tetrahedral Sites for Suppressed Manganese Dissolution and Enhanced Cycle Stability

    Surface degradation induced by the dissolution of manganese is a major failure mechanism that has hampered the large-scale adoption of spinel LiMn2O4 cathode material for decades. Although modifications based on restraining manganese disproportionation reactions have received much attention, many of these methods may sacrifice batteries’ specific capacity and rate capability. Here, we present a strategy that surface-populated magnesium ion substitution at 8a tetrahedral sites in LiMn2O4 could significantly suppress spinel surface evolution and Mn dissolution during cycling. With a total trace amount of magnesium surface doping (0.3 wt %) introduced by ALD coating and annealing, the LiMn2O4 cathode shows a capacity retention of 98.6% after 250 cycles.
