    Effect of Phellodendron chinense Schneid Extract on Chronic Bacterial Prostatitis Induced by Chlamydia in Rats

    Purpose: To explore the effects of Phellodendron chinense Schneid (PCS) extract on chlamydiainduced chronic bacterial prostatitis (CBP).Methods: Sixty 8-week-old male Sprague-Dawley rats were used in this study. Prostate index (PI) and prostate specific antigen (PSA) were determined after 4 weeks of oral administration of PCS extract (80, 160 or 320 mg/kg) or tetracycline (80 mg/kg) and compared to PI and PSA in untreated rats with CBP and healthy control rats (n = 10). Chronic inflammatory cell infiltrates, acinar changes, and interstitial fibrosis were evaluated by histopathological examination. In addition, pertinent inflammatory factors, including tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), cyclooxygenase-2 (COX-2), prostaglandin E2 (PEG2), transforming growth factor β1 (TGF-β1), and connective tissue growth factor (CTGF), were measured in prostate tissues using ELISA kits.Results: High doses of PCS (160 and 320 mg/kg) significantly decreased PI and PSA relative to model group (p < 0.01). PCS treatment also significantly reduced chronic inflammatory cell infiltrates and interstitial fibrosis in prostate tissue of CBP rats. In addition, TNF-α, IL-1β, COX-2, PEG2, TGF-β1, and CTGF decreased in PCS-treated rats compared to untreated control (p < 0.01).Conclusion: PCS extract has significant anti-inflammatory effects on chlamydia-induced CBP.Keywords: Phellodendron chinense Schneid, Bacterial prostatitis, Inflammatory factors, Morphometric analysis, Interstitial fibrosis, Prostate specific antigen, Prostate inde

    X-ray absorption of liquid water by advanced ab initio methods

    Oxygen K-edge X-ray absorption spectra of liquid water are computed based on the configurations from advanced ab initio molecular dynamics simulations, as well as an electron excitation theory from the GW method. One one hand, the molecular structures of liquid water are accurately predicted by including both van der Waals interactions and hybrid functional (PBE0). On the other hand, the dynamic screening effects on electron excitation are approximately described by the recently developed enhanced static Coulomb hole and screened exchange approximation by Kang and Hybertsen [Phys. Rev. B 82, 195108 (2010)]. The resulting spectra of liquid water are in better quantitative agreement with the experimental spectra due to the softened hydrogen bonds and the slightly broadened spectra originating from the better screening model.Comment: 10 pages, 5 figures, accepted by Phys.     Postpartum practices of puerperal women and their influencing factors in three regions of Hubei, China

    BACKGROUND: 'Sitting month' is a Chinese tradition for women's postpartum custom. The present study aims to explore the postpartum dietary and health practices of puerperal women and identify their influential factors in three selected regions of Hubei, China. METHODS: A cross-sectional retrospective study was conducted in the selected urban, suburban and rural areas in the province of Hubei from 1 March to 30 May 2003. A total of 2100 women who had given birth to full-term singleton infants in the past two years were selected as the participants. Data regarding postpartum practices and potentially related factors were collected through questionnaire by trained investigators. RESULTS: During the puerperium, 18% of the participants never ate vegetables, 78.8% never ate fruit and 75.7% never drank milk. Behaviour taboos such as no bathing, no hair washing or teeth brushing were still popular among the participants. About half of the women didn't get out of the bed two days after giving birth. The average time they stayed in bed during this period was 18.0 h. One third of them didn't have any outdoor activities in that time periods. The educational background of both women and their spouses, location of their residence, family income, postnatal visit, nutrition and health care educational courses were found to be the influencing factors of women's postpartum practices. CONCLUSION: Traditional postpartum dietary and health behaviours were still popular among women in Hubei. Identifying the factors associated with traditional postpartum practices is critical to develop better targeting health education programs.     Structural basis of DNA binding by the WhiB-like transcription factor WhiB3 in Mycobacterium tuberculosis

    Mycobacterium tuberculosis (Mtb) WhiB3 is an iron–sulfur cluster-containing transcription factor belonging to a subclass of the WhiB-Like (Wbl) family that is widely distributed in the phylum Actinobacteria. WhiB3 plays a crucial role in the survival and pathogenesis of Mtb. It binds to the conserved region 4 of the principal sigma factor (σA4) in the RNA polymerase holoenzyme to regulate gene expression like other known Wbl proteins in Mtb. However, the structural basis of how WhiB3 coordinates with σA4 to bind DNA and regulate transcription is unclear. Here we determined crystal structures of the WhiB3:σA4 complex without and with DNA at 1.5 Å and 2.45 Å, respectively, to elucidate how WhiB3 interacts with DNA to regulate gene expression. These structures reveal that the WhiB3:σA4 complex shares a molecular interface similar to other structurally characterized Wbl proteins and also possesses a subclass-specific Arg-rich DNA-binding motif. We demonstrate that this newly defined Arg-rich motif is required for WhiB3 binding to DNA in vitro and transcriptional regulation in Mycobacterium smegmatis. Together, our study provides empirical evidence of how WhiB3 regulates gene expression in Mtb by partnering with σA4 and engaging with DNA via the subclass-specific structural motif, distinct from the modes of DNA interaction by WhiB1 and WhiB7

    Phase behavior study of human antibody solution using multi-scale modeling

    Phase transformation in antibody solutions is of growing interest in both academia and the pharmaceutical industry. Recent experimental studies have shown that, as in near-spherical proteins, antibodies can undergo a liquid-liquid phase separation under conditions metastable with respect to crystallization. However the phase diagram of the Y-shaped antibodies exhibit unique features that differ substantially from those of spherical proteins. Specifically, antibody solutions have an exceptionally low critical volume fraction (CVF) and a broader and more asymmetric liquid-liquid coexistence curve than those of spherical proteins. Using molecular dynamics simulation on a series of trimetric Y-shaped coarse-grained models, we investigate the phase behavior of antibody solutions and compare the results with the experimental phase diagram of human IgG antibodies. With the fitted size of spheres, our simulation reproduces both the low CVF and the asymmetric shape of the experimental coexistence curve of IgG antibodies. The broadness of the coexistence curve can be attributed to the anisotropic nature of the inter-protein interaction. In addition, the repulsion between the inner parts of the spherical domains of IgG dramatically expands the coexistence region in the scaled phase diagram, while the hinge length has only a minor effect on the CVF and the overall shape of the coexistence curve. However the meta-stability gap between the solubility line and the coexistence curve sharply increases as the hinge length increases. We thus propose a seven-site model with empirical parameters that characterize the exclusion volume and the hinge length of the IgG molecules that provides a base for simulation studies of the phase behavior of IgG antibodies

    Functional imaging of interleukin 1 beta expression in inflammatory process using bioluminescence imaging in transgenic mice

    <p>Abstract</p> <p>Background</p> <p>Interleukin 1 beta (IL-1β) plays an important role in a number of chronic and acute inflammatory diseases. To understand the role of IL-1β in disease processes and develop an <it>in vivo </it>screening system for anti-inflammatory drugs, a transgenic mouse line was generated which incorporated the transgene firefly luciferase gene driven by a 4.5-kb fragment of the human IL-1β gene promoter. Luciferase gene expression was monitored in live mice under anesthesia using bioluminescence imaging in a number of inflammatory disease models.</p> <p>Results</p> <p>In a LPS-induced sepsis model, dramatic increase in luciferase activity was observed in the mice. This transgene induction was time dependent and correlated with an increase of endogenous IL-1β mRNA and pro-IL-1β protein levels in the mice. In a zymosan-induced arthritis model and an oxazolone-induced skin hypersensitivity reaction model, luciferase expression was locally induced in the zymosan injected knee joint and in the ear with oxazolone application, respectively. Dexamethasone suppressed the expression of luciferase gene both in the acute sepsis model and in the acute arthritis model.</p> <p>Conclusion</p> <p>Our data suggest that the transgenic mice model could be used to study transcriptional regulation of the IL-1β gene expression in the inflammatory process and evaluation the effect of anti-inflammatory drug <it>in vivo</it>.</p
