    A Novel Peptide-Based SILAC Method to Identify the Posttranslational Modifications Provides Evidence for Unconventional Ubiquitination in the ER-Associated Degradation Pathway.

    The endoplasmic reticulum-associated degradation (ERAD) pathway is responsible for disposing misfolded proteins from the endoplasmic reticulum by inducing their ubiquitination and degradation. Ubiquitination is conventionally observed on lysine residues and has been demonstrated on cysteine residues and protein N-termini. Ubiquitination is fundamental to the ERAD process; however, a mutant T-cell receptor α (TCRα) lacking lysine residues is targeted for the degradation by the ERAD pathway. We have shown that ubiquitination of lysine-less TCRα occurs on internal, non-lysine residues and that the same E3 ligase conjugates ubiquitin to TCRα in the presence or absence of lysine residues. Mass-spectrometry indicates that WT-TCRα is ubiquitinated on multiple lysine residues. Recent publications have provided indirect evidence that serine and threonine residues may be modified by ubiquitin. Using a novel peptide-based stable isotope labeling in cell culture (SILAC) approach, we show that specific lysine-less TCRα peptides become modified. In this study, we demonstrate that it is possible to detect both ester and thioester based ubiquitination events, although the exact linkage on lysine-less TCRα remains elusive. These findings demonstrate that SILAC can be used as a tool to identify modified peptides, even those with novel modifications that may not be detected using conventional proteomic work flows or informatics algorithms

    Ubiquitin Ligase RNF146 Regulates Tankyrase and Axin to Promote Wnt Signaling

    Canonical Wnt signaling is controlled intracellularly by the level of β-catenin protein, which is dependent on Axin scaffolding of a complex that phosphorylates β-catenin to target it for ubiquitylation and proteasomal degradation. This function of Axin is counteracted through relocalization of Axin protein to the Wnt receptor complex to allow for ligand-activated Wnt signaling. AXIN1 and AXIN2 protein levels are regulated by tankyrase-mediated poly(ADP-ribosyl)ation (PARsylation), which destabilizes Axin and promotes signaling. Mechanistically, how tankyrase limits Axin protein accumulation, and how tankyrase levels and activity are regulated for this function, are currently under investigation. By RNAi screening, we identified the RNF146 RING-type ubiquitin E3 ligase as a positive regulator of Wnt signaling that operates with tankyrase to maintain low steady-state levels of Axin proteins. RNF146 also destabilizes tankyrases TNKS1 and TNKS2 proteins and, in a reciprocal relationship, tankyrase activity reduces RNF146 protein levels. We show that RNF146, tankyrase, and Axin form a protein complex, and that RNF146 mediates ubiquitylation of all three proteins to target them for proteasomal degradation. RNF146 is a cytoplasmic protein that also prevents tankyrase protein aggregation at a centrosomal location. Tankyrase auto-PARsylation and PARsylation of Axin is known to lead to proteasome-mediated degradation of these proteins, and we demonstrate that, through ubiquitylation, RNF146 mediates this process to regulate Wnt signaling

    Detta är en socialpsykologisk vinjettstudie med syfte att ta reda på observatörens inställning till att ingripa i en mobbningssituation, där vi tittar på frågeställningarna: Påverkar maktrelationerna mellan mobbaren och mobbningsoffret observatörens vilja att ingripa? Påverkar maktrelationen mellan observatören och mobbaren observatörens vilja att ingripa? Påverkar könet på mobbaren och mobbningsoffret observatörens vilja att ingripa? Forskningen kring arbetsplatsmobbning är inte lika omfattande som den forskning som finns kring mobbning i skolmiljö. I denna studie använder vi oss av Björks definition av mobbning som är kontinuerliga inkompetensförklaringar. Studien utfördes på 144 personer som arbetar inom vården, eftersom den offentliga sektorn är ett yrkesområde som statistiskt sett drabbas mycket av mobbning. Mobbarens och den mobbades kön och status var de oberoende variablerna och ingripande, acceptans, oro, motivation och allvarlighetsgrad var de beroende variablerna. Resultatet visade på att mobbningsbeteendet anses som mer oacceptabelt om det är en chef som mobbar än en kollega (p=.049). Det visade sig också att status i kombination med kön (p=.042) var mest oacceptabelt om det var en kvinnlig chef som mobbade
