Who do they think they are? Wnt-responsive cells reveal their family trees
The nature of stem and progenitor cells in the mammary epithelium, and the relevance of cleared fat pad transplantation as a functional assay for them, has been thrown into doubt by recent lineage-tracking studies. Now two new studies based on tracking the progeny of Wnt-responsive cells are starting to help make sense of this fascinating problem.
Background: The mammary gland undergoes dramatic morphological changes during puberty, pregnancy and involution. Transplantation studies of tissue fragments into a cleared mammary fat pad pioneered by DeOme and colleagues showed reconstitution of the mammary gland [1], demon strating the presence of mammary stem cells. Work on serial transplantation of small numbers of purified basal mammary epithelial cells showed that they could regenerate the entire mammary epithelial tree with high efficiency and, indeed, that a single basal cell could do so [2-5].
These studies suggested the presence of multipotent mammary stem cells, located in the basal compartment. In contrast, purified luminal cells have limited fat pad reconstitution ability, although c-Kit-positive luminal cells were capable of forming small epithelial outgrowths in the transplant assay [5]. This model was recently challenged by Van Keymeulen and colleagues, who suggested that the basal and luminal lineages were uncoupled in the adult mammary epithelium and that each lineage contained a unipotent stem cell population [6]. The lineage contribution of stem/progenitor cells during pregnancy was unclear, however, as Van Keymeulen and colleagues were able to label very few luminal cells for lineage tracking - many fewer than the number of pregnancy-responsive progenitors, thought to be luminal, in the gland [5,6]. These progenitors generate alveoli, containing milk-secreting alveolar luminal cells and contractile myoepithelial cells, during pregnancy. Previous studies on lineage tracking of pregnancy-responsive mammary epithelial cells suggested that luminal pregnancy-responsive progenitors could contribute to both luminal and myoepithelial layers in the alveoli [7].
Our understanding of the relationship of stem and progenitor populations in the mammary epithelium with the different epithelial lineages is therefore currently very much in flux; two new publications examining the lineage contributions of Wnt-responsive cells are starting to clear these muddy waters
NOTCH and AKT signalling interact to drive mammary tumour heterogeneity
A better understanding of the mechanisms generating tumour heterogeneity will allow better targeting of current therapies, identify potential resistance mechanisms and highlight new approaches for therapy. We have previously shown that in genetically modified mouse models carrying conditional oncogenic alleles, mammary tumour histotype varies depending on the combination of alleles, the cell type to which they are targeted and, in some cases, reproductive history. This suggests that tumour heterogeneity is not a purely stochastic process; rather, differential activation of signalling pathways leads to reproducible differences in tumour histotype. We propose the NOTCH signalling pathway as one such pathway. Here, we have crossed conditional knockout Notch1 or Notch2 alleles into an established mouse mammary tumour model. Notch1/2 deletion had no effect on tumour-specific survival; however, loss of Notch alleles resulted in a dose-dependent increase in metaplastic adenosquamous carcinomas (ASQCs). ASQCs and adenomyoepitheliomas (AMEs) also demonstrated a significant increase in AKT signalling independent of Notch status. Therefore, the NOTCH pathway is a suppressor of the ASQC phenotype, while increased PI3K/AKT signalling is associated with ASQC and AME tumours. We propose a model in which PI3K/AKT and NOTCH signalling act interact to determine mouse mammary tumour histotype

Methodologies for Housing Justice Resource Guide
This Resource Guide is the outcome of a Summer Institute on Methodologies for Housing Justice convened by the Institute on Inequality and Democracy at UCLA Luskin as part of the Housing Justice in Unequal Cities Network, which is supported by the National Science Foundation (BCS 1758774). Held in Los Angeles in August 2019, the Summer Institute brought together participants from cities around the world. As is the case with the overall scope and purpose of the Housing Justice in Unequal Cities Network, it created a shared terrain of scholarship for movement-based and university-based scholars. Dissatisfied with the canonical methods that are in use in housing studies and guided by housing justice movements that are active research communities, the Summer Institute was premised on the assertion that methodology is political. Methodology is rooted in arguments about the world and involves relations of power and knowledge. The method itself – be it countermapping or people’s diaries – does not ensure an ethics of solidarity and a purpose of justice. Such goals require methodologies for liberation. Thus, as is evident in this Resource Guide, our endeavor foregrounds innovative methods that are being used by researchers across academia and activism and explicitly situates such methods in an orientation towards housing justice

El síndrome de Burnout constituye una respuesta al estrés laboral crónico, donde se distinguen tres dimensiones: agotamiento emocional, despersonalización y realización personal. Los profesionales que tienen interacción directa y constante con otras personas, son los de mayor riesgo; los enfermeros, se consideran uno de los grupos más vulnerables para desarrollar el síndrome. Objetivo: Diagnosticar el Síndrome de Burnout e identificar las variables estadísticamente significativas relacionadas con el desarrollo del síndrome en enfermeros(as) del área clínica y administrativa del Hospital Universitario San José, Popayán. Materiales y Métodos: Estudio de tipo Descriptivo, enfoque cuantitativo, correlacional, de corte transversal, con participación voluntaria de 120 enfermeros(as), se aplicó un cuestionario de datos sociodemográficos y laborales y el Inventario de Maslach Burnout Inventory validado al español. Los datos se procesaron en el programa SPSS versión 23. Para el análisis estadístico se emplearon medidas de tendencia central y para estimar la correlación entre las variables y las tres subescalas del síndrome. Resultados: La muestra estuvo constituida por 80,8% mujeres, 76,7% tenían vinculación a término indefinido, el 79,2% laboraban en el área asistencial, con una experiencia laboral mayor a 10 años del 52,5%. El 15,8% presentaron nivel alto en la subescala de Agotamiento Emocional, el 10,8% alta Despersonalización y el 9,2% nivel bajo en Realización Personal; la prevalencia del síndrome de Burnout fue del 1,7%. Conclusión: La presencia del síndrome de Burnout, en enfermeros que laboran en el Hospital Universitario San José, Popayán fue baja, el 65,8% presentaron alto nivel en realización personal.

Rapid activation of epithelial-mesenchymal transition drives PARP inhibitor resistance in Brca2-mutant mammary tumours
Tumours defective in the DNA homologous recombination repair pathway can be effectively treated with poly (ADP-ribose) polymerase (PARP) inhibitors; these have proven effective in clinical trials in patients with BRCA gene function-defective cancers. However, resistance observed in both pre-clinical and clinical studies is likely to impact on this treatment strategy. Over-expression of phosphoglycoprotein (P-gp) has been previously suggested as a mechanism of resistance to the PARP inhibitor olaparib in mouse models of Brca1/2-mutant breast cancer. Here, we report that in a Brca2 model treated with olaparib, P-gp upregulation is observed but is not sufficient to confer resistance. Furthermore, resistant/relapsed tumours do not show substantial changes in PK/PD of olaparib, do not downregulate PARP1 or re-establish double stranded DNA break repair by homologous recombination, all previously suggested as mechanisms of resistance. However, resistance is strongly associated with epithelial-mesenchymal transition (EMT) and treatment-naïve tumours given a single dose of olaparib upregulate EMT markers within one hour. Therefore, in this model, olaparib resistance is likely a product of an as-yet unidentified mechanism associated with rapid transition to the mesenchymal phenotype

Metodologías para la justicia de la vivienda: Guia de recursos
Esta Guía de Recursos es el resultado de un Instituto de Verano sobre Metodologías para la Justicia en la Vivienda convocado por el Instituto sobre Desigualdad y Democracia de UCLA Luskin como parte de la Red de Justicia en la Vivienda en Ciudades Desiguales, que es apoyada por la Fundación Nacional de Ciencias (BCS 1758774). Celebrado en Los Ángeles en agosto de 2019, el Instituto de Verano reunió a participantes de ciudades de todo el mundo. Al igual que el alcance y el propósito general de la Red de Justicia en las Ciudades Desiguales, creó un terreno compartido de para estudiosos del movimiento y académicos de universidades. Con una insatisfacción a los métodos canónicos que se utilizan en los estudios sobre la vivienda y guiado por los movimientos de justicia de la vivienda que son comunidades de investigación activa, el Instituto de verano se basó en la afirmación de que la metodología es política. La metodología se basa en argumentos sobre el mundo e implica relaciones de poder y conocimiento. El método por sí mismo -ya sea el contra ataque al mapeo o los diarios de la genteno asegura una ética de solidaridad y un propósito de justicia. Tales objetivos requieren metodologías para la liberación. Por lo tanto, como es evidente en esta Guía de Recursos, nuestro esfuerzo pone en primer plano los métodos innovadores que están siendo utilizados por los investigadores en todo el mundo académico y el activismo y sitúa explícitamente tales métodos en una orientación hacia la vivienda la justicia
