    Relationship between polymorphisms in vitamin D metabolism-related genes and the risk of rickets in Han Chinese children

    BACKGROUND: Vitamin D deficiency rickets is common in China. Genetic factors may play an important role in the susceptibility to rickets. Our study aimed to identify the relationship between three vitamin D-related genes (group specific component [GC], cytochrome P450, family 2, subfamily R, polypeptide 1 (CYP2R1), and 7-dehydrocholesterol reductase/nicotinamide-adenine dinucleotide synthetase 1 (DHCR7/NADSYN1) and rickets in Han Chinese children from northeastern China. METHODS: A total of 506 Han children from northeastern China were enrolled in the current study. Twelve SNPs in three candidate genes were genotyped using the SNaPshot assay. Linear regression was used to examine the effect of 12 single-nucleotide polymorphisms (SNPs) on the risk of rickets. RESULTS: In our case–control cohort, six alleles of the 12 SNPs conferred a significantly increased risk of rickets in GC (rs4588 C, P = 0.003, OR: 0.583, 95% CI: 0.412-0.836; rs222020 C, P = 0.009, OR: 1.526, 95% CI: 1.117-2.0985; rs2282679 A, P = 0.010, OR: 0.636, 95% CI: 0.449-0.900; and rs2298849 C, P = 0.001, OR: 1.709, 95% CI: 1.250-2.338) and in CYP2R1 (rs10741657 G, P = 0.019, OR: 1.467, 95% CI: 1.070-2.011; and rs2060793 G, P = 0.023, OR: 0.689, 95% CI: 0.502-0.944). The results remained significant after adjustment for sex and body mass index. We further analyzed the effect of genotypes under three different genetic models. After using Bonferroni’s method for multiple corrections, rs4588, rs2282679, and rs2298849 of the GC gene were significantly associated with rickets under the dominant (P =0.003 for rs4588, P =0.024 for rs2282679, and P =0.005 for rs2298849) and additive models (P = 0.006 for rs4588, P = 0.024 for rs2282679, and P = 0.005 for rs2298849). Haplotype analysis showed that the CAT haplotype of the GC gene (P = 0.005) and the GAA haplotype of the CYP2R1 gene (P = 0.026) were associated with susceptibility to rickets. CONCLUSIONS: This case–control study confirmed the strong effect of GC and CYP2R1 loci on rickets in Han children from northeastern China

    A Novel 3D Imaging Method for Airborne Downward-Looking Sparse Array SAR Based on Special Squint Model

    Three-dimensional (3D) imaging technology based on antenna array is one of the most important 3D synthetic aperture radar (SAR) high resolution imaging modes. In this paper, a novel 3D imaging method is proposed for airborne down-looking sparse array SAR based on the imaging geometry and the characteristic of echo signal. The key point of the proposed algorithm is the introduction of a special squint model in cross track processing to obtain accurate focusing. In this special squint model, point targets with different cross track positions have different squint angles at the same range resolution cell, which is different from the conventional squint SAR. However, after theory analysis and formulation deduction, the imaging procedure can be processed with the uniform reference function, and the phase compensation factors and algorithm realization procedure are demonstrated in detail. As the method requires only Fourier transform and multiplications and thus avoids interpolations, it is computationally efficient. Simulations with point scatterers are used to validate the method

    Impaired left ventricular function is an independent predictor of adverse clinical outcomes in patients with aortic stenosis. The aim of this study is to evaluate the short-term changes of echocardiographic parameters, New York Heart Association (NYHA) class and B-type natriuretic peptide (BNP) level and adverse events amongst patients with heart failure (HF) after transcatheter aortic valve replacement (TAVR) procedure. This was a retrospective cohort study conducted at affiliated Yantai Yuhuangding Hospital of Qingdao University between September 2017 and September 2022. TAVR cases were stratified into three groups [heart failure with reduced ejection fraction (HFrEF), heart failure with mildly reduced ejection fraction (HFmrEF), heart failure with preserved ejection fraction (HFpEF)] by left ventricular ejection fraction (LVEF). Baseline characteristics, changes in echocardiographic parameters (1 week and 1 month), BNP (1 month), and NYHA class (6 months) post-TAVR were compared across the three groups. Meanwhile, we observed the adverse events of the patients after TAVR. A total of 96 patients were included, of whom 15 (15.6%) had HFrEF, 15 (15.6%) had HFmrEF, and 66 (68.8%) had HFpEF. Compared to the HFpEF subgroup, patients in the HFrEF subgroup were younger (p < 0.05), and with a higher BNP (p < 0.05). The left ventricular end-diastolic dimension (LVEDD) in HFrEF group decreased significantly after TAVR. HFmrEF and HFrEF patients showed significant improvements in LVEF after TAVR. The pulmonary artery systolic pressure (PASP), aortic valve peak gradient (AVPG) and aortic valve peak gradient (V ) decreased significantly 1 month after TAVR in all three groups compared to the baseline (all p < 0.05). BNP significantly reduced in HFrEF group compared to HFpEF patients after TAVR (p < 0.05). The majority of patients experienced an improvement at least one NYHA class in all three groups 6 months post-TAVR. There is no significant increase in the risk of adverse events in the HFrEF group. Patients who underwent TAVR achieved significant improvements in BNP, NYHA class, LVEDD, LVEF, and PASP across the three HF classes, with a more rapid and pronounced improvement in the HFrEF and HFmrEF groups. Complication rates were low in the different HF groups. There is no significant increase in the risk of periprocedural complications in the HFrEF and HFmrEF groups. [Abstract copyright: © 2023.     Facile, high efficiency immobilization of lipase enzyme on magnetic iron oxide nanoparticles via a biomimetic coating

    BackgroundImmobilization of lipase on appropriate solid supports is one way to improve their stability and activity, and can be reused for large scale applications. A sample, cost- effective and high loading capacity method is still challenging.ResultsA facile method of lipase immobilization was developed in this study, by the use of polydopamine coated magnetic nanoparticles (PD-MNPs). Under optimal conditions, 73.9% of the available lipase was immobilized on PD-MNPs, yielding a lipase loading capacity as high as 429 mg/g. Enzyme assays revealed that lipase immobilized on PD-MNPs displayed enhanced pH and thermal stability compared to free lipase. Furthermore, lipase immobilized on PD-MNPs was easily isolated from the reaction medium by magnetic separation and retained more than 70% of initial activity after 21 repeated cycles of enzyme reaction followed by magnetic separation.ConclusionsImmobilization of enzyme onto magnetic iron oxide nanoparticles via poly-dopamine film is economical, facile and efficient

    LDL-C: An important independent risk factor for new-onset heart block in patients with severe aortic stenosis and heart failure after TAVR

    Transcatheter aortic valve replacement (TAVR) is an effective alternative treatment for patients with aortic stenosis (AS) who have intermediate to high surgical risk or who are inoperable. However, the incidence of conduction abnormalities is high after TAVR, which can reduce the effectiveness of the surgery. Our research objective is to explore the risk factors of new-onset conduction abnormalities after TAVR, providing reference value for clinical doctors to better prevent and treat conduction abnormalities. Patients who underwent TAVR were divided into those who developed heart block and those who did not. Baseline clinical characteristics, cardiac structural parameters, procedural characteristics, electrocardiogram (ECG) changes before and after TAVR ( = postoperative minus preoperative), and surgical complications were compared. Logistic regression was applied to identify significant risk factors for new-onset heart block. We studied 93 patients, of whom 34.4% developed heart blocks. Univariate logistic regression showed that prior history of malignancy, atrial fibrillation, preoperative high-level total cholesterol and low-density lipoprotein cholesterol (LDL-C), HR, QRS interval, QT interval, and QTc interval were risk factors of new-onset heart block after TAVR. Multivariate analysis showed that preoperative high-level LDL-C and QRS interval remained significant independent risk factors after adjusting for potential confounds. Heart block is the most common complication of TAVR, and its significant independent risk factors include high-level LDL-C and QRS interval. [Abstract copyright: Copyright: © 2023 The Author(s).     Clinical meaning of serum trimethylamine oxide, N-terminal-pro-brain natriuretic peptide, hypoxia-inducible factor-1a and left ventricular function and pregnancy outcome in patients with pregnancy-induced hypertension

    Background: To figure out the clinical meaning of serum trimethylamine oxide (TMAO), N-terminal-pro-brain natriuretic peptide (NT-proBNP) and hypoxia-inducible factor-1a (HIF-1a) with left ventricular function and pregnancy outcome in patients with pregnancy-induced hypertension. Methods: From January 2018 to October 2020, 117 patients with gestational hypertension were taken as the research objects and grouped into the gestational hypertension (pregnancy-induced hypertension, 55 cases), mild preeclampsia (mild PE, 43 cases) and severe preeclampsia (severe PE, 19 cases) in the light of the severity of the disease. Analysis of the relation of serum TMAO, NT-proBNP and HIF-1a with the severity of disease and cardiac function indexes in patients with gestational hypertension was conducted. All patients were followed up to the end of pregnancy, and the predictive value of serum TMAO, NT-proBNP and HIF-1a on pregnancy outcome in patients was analyzed. Results: Serum TMAO and NT-proBNP of patients were elevated, while HIF-1a was reduced with the severity of the disease (P < 0.05). Serum TMAO and NT-proBNP in patients with gestational hypertension were positively correlated but HIF-1a was negatively correlated with the severity of the disease (P < 0.05). Left ventricular end-diastolic volume (LVEDV) and left ventricular end-systolic volume (LVESV) were elevated in gestational hypertension patients, while ejection fraction (LVEF) was reduced with the severity of disease (P < 0.05). Serum TMAO, NT-proBNP and HIF1a were associated with LVEDV, LVESV and LVEF values in patients with gestational hypertension (P < 0.05). Serum TMAO and NT-proBNP were elevated but HIF-1a was reduced in patients with a poor pregnancy outcome (P < 0.05). The AUC of the combined detection of serum TMAO, NT-proBNP and HIF-1a on pregnancy outcome was greater (P < 0.05). Conclusions: Serum TMAO, NT-proBNP and HIF-1a in patients with gestational hypertension are associated with disease severity and cardiac function, and have predictive and evaluative values for disease severity and pregnancy outcome
